The modification and generation of new biomolecules intended to give higher molecular-mass species for biotechnological purposes, can be achieved by enzymatic cross-linking. The versatile peroxidase (VP) from Pleurotus eryngii is a high redox-potential enzyme with oxidative activity on a wide variety of substrates. In this study, VP was successfully used to catalyze the polymerization of low molecular mass compounds, such as lignans and peptides, as well as larger macromolecules, such as protein and complex polysaccharides. Different analytical, spectroscopic, and rheological techniques were used to determine structural changes and/or variations of the physicochemical properties of the reaction products. The lignans secoisolariciresinol and hydroxymatairesinol were condensed by VP forming up to 8 unit polymers in the presence of organic co-solvents and Mn2+. Moreover, 11 unit of the peptides YIGSR and VYV were homogeneously cross-linked. The heterogeneous cross-linking of one unit of the peptide YIGSR and several lignan units was also achieved. VP could also induce gelation of feruloylated arabinoxylan and the polymerization of β-casein. These results demonstrate the efficacy of VP to catalyze homo- and hetero-condensation reactions, and reveal its potential exploitation for polymerizing different types of compounds.  相似文献

This study combines wavelet decomposition and independent component analysis (ICA) to extract mismatch negativity (MMN) from electroencephalography (EEG) recordings. As MMN is a small event-related potential (ERP), a systematic ICA based approach is designed, exploiting MMN’s temporal, frequency and spatial information. Moreover, this study answers which type of EEG recordings is more appropriate for ICA to extract MMN, what kind of the preprocessing is beneficial for ICA decomposition, which algorithm of ICA can be chosen to decompose EEG recordings under the selected type, how to determine the desired independent component extracted by ICA, how to improve the accuracy of the back projection of the selected independent component in the electrode field, and what can be finally obtained with the application of ICA. Results showed that the proposed method extracted MMN with better properties than those estimated by difference wave only using temporal information or ICA only using spatial information. The better properties mean that the deviant with larger magnitude of deviance to repeated stimuli in the oddball paradigm can elicit MMN with larger peak amplitude and shorter latency. As other ERPs also have the similar information exploited here, the proposed method can be used to study other ERPs.  相似文献

The aim of the study was to evaluate the effects of single and repeated whole-body cryotherapy (WBC, air −110 °C) on the neuromuscular performance in healthy subjects (n=14). The flight times in a drop-jump exercise decreased after a single WBC exposure, but these changes almost vanished after repeated WBC for 3 months. This adaptation was accompanied by a decreased co-contraction of lower leg muscles during the drop-jump. In conclusion, in dynamic exercise, neuromuscular functioning may be able to adapt to repeated WBC, which might enhance the effects of therapeutic exercises in patients after the WBC.  相似文献

Concentrations in serum were determined for 18 fatty acids (FAs) and 21 lipoprotein main and subclasses by chromatographic analyses and the average size was calculated for very low density (VLDL), low density (LDL) and high density (HDL) particles. 283 ethnic Norwegian children and adults from the rural Fjord region of Western Norway were compared with the objectives to reveal patterns and gender differences during the development from prepuberty to adulthood and during aging in adults. Both genders showed a large increase in eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) from child to adult. Males, but not females, show a significant increase in most C16–C18 FAs from prepuberty to adulthood. These changes in males correlate to a pattern of increased concentrations of triglycerides, VLDL and LDL particles, especially the atherogenic subclasses of small and very small LDL particles. Furthermore, concentrations of medium, large and very large HDL particles decrease, while concentration of very small HDL particles increase leading to reduced average size of HDL particles. Females only showed significant increase in concentrations of small and very small LDL particles, very small HDL particles and apolipoprotein B. While EPA and DHA continued to increase during aging in women, no validated model for connecting age to FA profile was obtained for men. Women showed significant increase in concentrations of all subclasses of LDL particles during aging, while men exhibited a more complex pattern with increase also in apolipoprotein A1 and HDL particles.  相似文献

Consumption of contaminated drinking water is a significant cause of Campylobacter infections. Drinking water contamination is known to result from septic seepage and wastewater intrusion into non-disinfected sources of groundwater and occasionally from cross-connection into drinking water distribution systems. Wastewater effluents, farm animals and wild birds are the primary sources contributing human-infectious Campylobacters in environmental waters, impacting on recreational activities and drinking water sources. Culturing of Campylobacter entails time-consuming steps that often provide qualitative or semi-quantitative results. Viable but non-culturable forms due to environmental stress are not detected, and thus may result in false-negative assessments of Campylobacter risks from drinking and environmental waters. Molecular methods, especially quantitative PCR applications, are therefore important to use in the detection of environmental Campylobacter spp. Processing large volumes of water may be required to reach the desired sensitivity for either culture or molecular detection methods. In the future, applications of novel molecular techniques such as isothermal amplification and high-throughput sequencing applications are awaited to develop and become more affordable and practical in environmental Campylobacter research. The new technologies may change the knowledge on the prevalence and pathogenicity of the different Campylobacter species in the water environment.  相似文献

Many butterfly genera are characterised by the presence of marginal eyespots on their wings. One hypothesis to account for an occurrence of eyespots is that these wing pattern elements are partly the outcome of visual selection by predators. Bicyclus anynana (Satyrinae) has underside spotting on its wings but there is also a seasonal form in which the eyespots are reduced in size or totally absent. This natural variation gives us a useful tool to test the hypothesis that marginal eyespot patterns can decoy the attacking predator by, at least sometimes, diverting attack from vital body parts to the edges of the wings. We used lizards, Anolis carolinensis , and pied flycatchers, Ficedula hypoleuca , as predators for living spotted and spotless B. anynana . The presence of eyespots did not increase the escape probability of resting butterflies once captured (even a form with enlarged eyespots did not add to effective deflection of attacks). There was also no evidence that eyespots influenced the location of strikes by the predators. This study thus provides no support that marginal eyespot patterns can act as an effective deflection mechanism to avoid lizard or avian predation.  相似文献

Insects' cold tolerance during their development is a surprisingly understudied subject in ecology, despite the fact that subzero temperatures during the growing season are common at high altitudes and latitudes. Subzero temperatures can have detrimental effects on organisms, restricting a species' range. This study addresses the question whether night frosts during the growing season have an instant or delayed negative impact on larval mortality of the Colorado potato beetle, Leptinotarsa decemlineata (Say) (Coleoptera: Chrysomelidae). We also tested whether populations from the centre (Poland) and margins (Russia) of the distribution range of L. decemlineata differ in their responses to subzero exposure and a low rearing temperature. Larvae of three ages were subjected to a subzero temperature (−4 °C for 3 h simulating night frost) twice, after which they were reared on a fluctuating temperature regime of 10–15 °C. These rearing conditions imitated cool summer temperatures beyond the beetles' current range, such as in Finland. Individuals of both populations were highly cold tolerant, as only 3.1% of larvae died immediately following the subzero treatment. Nonetheless, the low rearing temperature was harmful to beetles of both populations. It caused high larval (ca. 90%) and overwintering (ca. 80%) mortality. As beetle performance was affected solely by rearing temperature, low temperatures during the growing season rather than night frosts apparently retard the beetle's northern expansion.  相似文献
