Coeliac disease is characterised by inflammation of small intestinal mucosa accompanied by abnormal villous architecture. It is now accepted that some patients with positive coeliac serology tests may have minor mucosal lesions that may not be apparent on routine histopathological analysis. The aim of the study was to perform detailed examination of enterocyte morphology and cytoskeletal structures using a high content analysis technology. Duodenal biopsies from 14 untreated and 10 treated coeliac patients and from 20 non-coeliac controls were examined. Tissue sections from six patients (study group subjects) before and after the development of gluten-sensitive enteropathy were also investigated. Immunohistochemical studies were performed on paraffin-embedded sections using an anti-α-tubulin antibody. Significant differences in enterocyte morphology and intracellular cytoskeletal structures were demonstrated in patients with proven coeliac disease and in the study group subjects. These changes were present in study group biopsies before evidence of enteropathy, as assessed by routine microscopy. This is the first study to demonstrate detailed characteristics of enterocyte morphology in coeliac patients using a high content analysis approach. The use of this technology allows a quantitative analysis of enterocyte intracellular structures from routine biopsy material and permits detection of subtle changes that precede the characteristic histological lesion.  相似文献

Glycosyl hydrolase family 1 β-glucosidases are important enzymes that serve many diverse functions in plants including defense, whereby hydrolyzing the defensive compounds such as hydroxynitrile glucosides. A hydroxynitrile glucoside cleaving β-glucosidase gene (Llbglu1) was isolated from Leucaena leucocephala, cloned into pET-28a (+) and expressed in E. coli BL21 (DE3) cells. The recombinant enzyme was purified by Ni–NTA affinity chromatography. The optimal temperature and pH for this β-glucosidase were found to be 45 °C and 4.8, respectively. The purified Llbglu1 enzyme hydrolyzed the synthetic glycosides, pNPGlucoside (pNPGlc) and pNPGalactoside (pNPGal). Also, the enzyme hydrolyzed amygdalin, a hydroxynitrile glycoside and a few of the tested flavonoid and isoflavonoid glucosides. The kinetic parameters K m and V max were found to be 38.59 μM and 0.8237 μM/mg/min for pNPGlc, whereas for pNPGal the values were observed as 1845 μM and 0.1037 μM/mg/min. In the present study, a three dimensional (3D) model of the Llbglu1 was built by MODELLER software to find out the substrate binding sites and the quality of the model was examined using the program PROCHEK. Docking studies indicated that conserved active site residues are Glu 199, Glu 413, His 153, Asn 198, Val 270, Asn 340, and Trp 462. Docking of rhodiocyanoside A with the modeled Llbglu1 resulted in a binding with free energy change (ΔG) of ?5.52 kcal/mol on which basis rhodiocyanoside A could be considered as a potential substrate.  相似文献

Agrotis segetum Schiff granulosis virus (AsGV) propagated in Denmark was supplied against naturally occurring cutworm populations (A. ipsilon and to a less extentA. segetum) in experimental field plots of tobacco, okra, potato and sugar beet in northern Pakistan. AsGV doses varied between 4 × 107 and 4 × 1011 capsules per m2 plot, and no. of applications between 1 and 3. One treatment with AsGV did not reduce cutworm damage significantly to tobacco seedlings and potato plants. Two treatments with AsGV reduced cutworm damage significantly. In tobacco, reduction was 64–82%, in okra and potato 85% and 77% respectively. Damage in sugar beet was reduced 78%. Three treatments with AsGV dis not reduce damage significantly better than two treatments. AsGV and the chemical insecticides Tamaran and Dieldrin, andBacillus thuringiensis (Thuricide) were about equally effective, reducing damage by 85%, 79%, 87% and 69%, respectively. No difference was found between the efficiency of highly purified AsGV to which activated charcoal was added and partially purified AsGV without charcoal.   相似文献
