The ventral prostrate was fractionated into one mitochondrial and three microsomal fractions. The different fractions were characterized morphologically and chemically. An interesting finding was that upon homogenization the endoplasmic reticulum membranes often turned ‘inside-out’ giving rise to microsomes with ribosomes attached to the inside of the vesicles. The secretion of the protatic secretion was studied by means of isotopic pulse labeling using radioactive leucine. Peak radioactivity in the secretory fluid was obtained at 2 h after injection with a relativity rapid fall. The radioactivity in the secretory fluid displayed a continuous increase up to 8 h followed by a plateau. When prostatic secretion was purified from secretory fluid and microsomes using a Con A-Sepharose column it showed a typical precursor-product relationship with an early peak at 60 min in microsomal prosatatic secretion protein followed by a peak in secretory fluid at 4 h. Vinblastine blocked the release of labeled secretion protein into the secretory fluid, a phenomenon characteristic for secretory proteins which are exocytosed by means of fusion between secretory granules and the plasma membrane. Following intravenous injection of [3H]estramustine, accumulation was seen in the secretory fluid. Some estramustine probably binds to newly synthesized protatic secretion protein and follows the same route of intracellular transport and extracellular discharge as does prostatic secretion protein.  相似文献

Obstetric selection acts on the female pelvic canal to accommodate the human neonate and contributes to pelvic sexual dimorphism. There is a complex relationship between selection for obstetric sufficiency and for overall body size in humans. The relationship between selective pressures may differ among populations of different body sizes and proportions, as pelvic canal dimensions vary among populations. Size and shape of the pelvic canal in relation to body size and shape were examined using nine skeletal samples (total female n = 57; male n = 84) from diverse geographical regions. Pelvic, vertebral, and lower limb bone measurements were collected. Principal component analyses demonstrate pelvic canal size and shape differences among the samples. Male multivariate variance in pelvic shape is greater than female variance for North and South Africans. High‐latitude samples have larger and broader bodies, and pelvic canals of larger size and, among females, relatively broader medio‐lateral dimensions relative to low‐latitude samples, which tend to display relatively expanded inlet antero‐posterior (A‐P) and posterior canal dimensions. Differences in canal shape exist among samples that are not associated with latitude or body size, suggesting independence of some canal shape characteristics from body size and shape. The South Africans are distinctive with very narrow bodies and small pelvic inlets relative to an elongated lower canal in A‐P and posterior lengths. Variation in pelvic canal geometry among populations is consistent with a high degree of evolvability in the human pelvis. Summary Membrane introduction mass spectrometry has been employed for on-line determination of the major products and volatile metabolites ofBacillus polymyxa fermentation. Samples were introduced into the mass spectrometer via a direct insertion membrane probe in which the aqueous solution flowed past a membrane located in the ion source of the mass spectrometer. Concentrations of the products 2,3-butanediol, acetoin, ethanol and acetic acid in fermentation broth were measured by tandem mass spectrometry after permeation through the membrane and ionization by chemical ionization. External standards were employed for quantification and a large linear response range was available for each of the major products observed. Dissolved CO2 and O2, as well as CO2 in the off gases, were also monitored on-line by mass spectrometry. The use of tandem mass spectrometry has allowed the identification of products that were not previously known to be present in measurable amounts.  相似文献

Summary We have studied the consequences of mutational alteration in the structure of EF-Tu on the missense errors and proofreading activity of bacterial ribosomes in vitro. Our data show that the EF-Tu Bo mutant form of EF-Tu (van der Meide et al. 1983a) is inactive in polypeptide synthesis on the ribosome, even though it binds aminoacyl-tRNA. A second mutant form, EF-Tu Ar (van der Meide et al. 1983a), is active in polypeptide synthesis but supports a much higher messense incorporation with either leucine isoacceptor 2 or leucine isoacceptor 4 in the in vitro system. Further analysis of the kinetic basis of this enhanced missense frequency revealed that the mutation responsible for the alteration in EF-Tu Ar increases the errors at both the proofreading step and the initial selection. In this respect the effect of this particular mutation is similar to the mode of action of the antibiotic kanamycin (Jelenc and Kurland 1984).  相似文献

The growth of the Finnish wolf (Canis lupus) population during the last years has highlighted people’s contradictory attitudes toward wolves. The supranational conservation objectives brought on by Finland’s membership in the European Union (EU) and the regional application of the official policy on wolves have led to conflicts. This article is based on the preparation process of the wolf management plan in Finland. As a part of the process, a nationwide hearing was arranged in 2004. Fear of wolves is widespread and even more common in areas with low wolf population. People living in areas where wolves occur feel that they can no longer influence decision making which affects them and that the authorities, conservationists, and the EU do not listen to their opinions. Numerous requests have been made for the legislation and its interpretation, and concessions to these requests would promote consensus and increase tolerance for wolves. Proposed changes include reforming the damage compensation system and formulating a clearer interpretation of the conservation status of wolves. However, it seems impossible to create a policy that would be universally supported. As the wolf question has become more central in environmental policy, the conservation and management of the species have become more complicated, and the cooperation between various interest groups has declined. The wolf conflict could be mitigated by compromises, but because the conflict is value-based, solutions are illusive.  相似文献

Hyperlipidemic rabbit serum and its lipid extract were found to impair the precipitation of glycosaminoglycans by cetylpyridinium chloride. The inhibition was 60–80% in the case of sulfated glycosaminoglycans and 75–85% in the case of hyaluronic acid. The interfering compounds could be removed by extracting the samples twice with a six-fold volume of ethanol-ether (2:1) for 1 h.  相似文献

According to parental investment theory, nest defence activity should be related to the reproductive value of the offspring. Alternative hypotheses suggest that defence activity may, for example, depend upon the conspicuousness of the young. Studies concerning this topic have been carried out almost exclusively on birds and experimental data on the diversity of organisms is lacking. Bank voles Clethrionomys glareolus were used to study the effects of the number and age of offspring upon the pup defence activity of mothers. Male bank voles are infanticidal and thus an adult male was used as a predator. Defence trials were conducted in the laboratory and filmed for subsequent analysis. Litter sizes were divided into three treatment groups: reduced (−2 pups), control (±0 pups) and enlarged (+2 pups). In order to study the effect of offspring age upon maternal defence activity the trials were conducted twice: when the pups were 3 and 8 days old. Defence activity increased with the number of offspring and enlarged litters were most actively defended. This result supports parental investment theory and conclusions drawn by earlier studies of birds. However, in contrast to the conclusions of earlier studies, older offspring were defended less than the younger ones. Whilst new-born pups are totally defenceless against predators their vulnerability decreases as they age. Therefore, we suggest that maternal aggression in female bank voles is related to the value as well as to the vulnerability of the offspring. The validity of this explanation and the determinants of parental investment decisions in small mammals in general deserve further study.
