Chronic pain and cognitive decline are characteristic symptoms of rheumatoid arthritis. One of the immediate early gene c-fos is overexpressed during peripheral and central noxious conditions and can be used as a marker for neuronal activity/excitability. In the adjuvant-induced arthritis Sprague–Dawley rat model, we examined the dynamics of c-Fos protein and mRNA expression in the amygdala, cortex, hippocampus, and thalamus and evaluated the effects of N-(2-hydroxy phenyl) acetamide (NA-2), a derivative of salicylic acid. The paw volume was assessed as an indicator of peripheral edema and the hyperalgesia associated with arthritis was monitored by gait analysis. The region of interests of the brain from arthritic and non-arthritic animals were used to isolate the RNA and were then reverse transcribed into cDNA. The PCR products were electrophoresed on 1 % agarose gel and the gels were visualized in gel-doc system. The frozen brain sections were stained for c-Fos using immunohistochemistry. Negative control experiments were performed without the primary and secondary antibodies to rule out the nonspecific tissue binding of antibodies. We report a significant increase in the c-Fos expression in the arthritic control animals. In comparison to the control group, the treatment of NA-2 treatment was found to block the development of the arthritis-induced c-Fos protein and mRNA expression and peripheral edema. It also significantly reduces the gait deficits which were otherwise observed in the arthritic control group. Both the immunohistochemistry and PCR analysis revealed NA-2 to be more potent in comparison to member of non-steroidal anti-inflammatory drug.  相似文献

Sea bream broodstock were immunised 1 or 2 months before spawning with a novel photobacteriosis vaccine. Sixty-seven-day-old larvae (mean weight 22.3 mg) originating from immunised and non-immunised parents were experimentally infected with the Photobacterium damsela subsp. piscicida (Phdp). Larvae from immunised fish showed delayed onset and lower mortality (66.67%) compared with larvae from control fish (80%). Eighty-nine-day-old larvae (mean weight 162.2 mg) from both groups were bath vaccinated with Phdp and Escherichia coli lipopolysaccharides (LPS) and larval samples were collected for measurement of humoral parameters. Larvae vaccinated with Phdp and LPS showed significantly higher anti-protease activity, lysozyme activity and total immunoglobulin compared to the controls. One-hundred-and-twenty-day-old larvae (mean weight 297.85 mg) from both parental groups were challenged with (LD70) virulent Phdp bacterial cells. Vaccinated larvae from both groups showed significantly less mortality compared to the respective controls. The RPS values of larvae from immunised parents vaccinated with Phdp and LPS was 95.83% and 72.22%, respectively. The RPS values of larvae from non-immunised parents vaccinated with Phdp and LPS was 62.5% and 70.83%, respectively. Results are discussed with respect to the beneficial effect of broodstock immunisation prior to spawning and the immunisation of larvae on their survival against photobacteriosis.  相似文献

Stretchable piezoelectric nanogenerators (SPENGs) for human kinematics energy harvesting have limited use due to the low stretchability or mechanical robustness and the difficulty of structural design for omnidirectional stretchability. This study reports an efficient, omnidirectionally stretchable, and robust SPENG based on a stretchable graphite electrode on a 3D micropatterned stretchable substrate and a stacked mat of piezoelectric nanofibers. The stacked mat of free‐standing nanofibers is alternatively composed of nanocomposite nanofibers of barium titanate nanoparticles embedded in polyurethane and poly(vinylidene fluoride‐trifluoroethylene) nanofibers. The nanofiber SPENG (nf‐SPENG) exhibits a high stretchability of 40% and high mechanical durability up to 9000 stretching cycles at 30% strain, which are attributed to the stress‐relieving nature of the 3D micropattern on the substrate and the free‐standing stacked hybrid nanofibers. The nf‐SPENG produces a peak open circuit voltage (Voc) and short circuit current (Isc) of 9.3 V and 189 nA, respectively. The nf‐SPENG is demonstrated to harvest the energy from human kinematics while walking when placed over the knee cap of a subject, generating a maximum Voc of 10.1 V. The omnidirectional stretchability, efficiency, facile fabrication process, mechanical durability, environmentally friendly lead‐free components, and response to multimodal straining make this device suitable for self‐powered wearable sensing systems.  相似文献

The present study estimated the length–weight relationship (LWR) and length‐length relationships (LLR) of four fish species, Ompok pabo (Hamilton, 1822), Acentrogobius cyanomos (Bleeker, 1849), Acentrogobius caninus (Valenciennes, 1837) and Gagata cenia (Hamilton, 1822) collected from the Payra river over a period of 1 year between July 2015–June 2016 using set bag nets and cast nets with a mesh size of 0.5 cm and <0.5 cm, respectively. Altogether 593 individuals of O. pabo (59), A. cyanomos (221), A. caninus (240) and G. cenia (73) were sampled and studied. LWRs of O. pabo, A. cyanomos, and A. caninus are presented for the first time in Fishbase.  相似文献

A Gram-staining-positive, catalase-positive, oxidase-negative, non-motile, non-flagellate and rod-shaped bacterium, was designated as DCY81T, and isolated from soil of a ginseng field in Pocheon province, Republic of Korea. The 16S rRNA gene sequence analysis revealed that strain DCY81T belonged to the genus Arthrobacter. Major fatty acid was anteiso-C15:0, while major polar lipids were diphosphatidyglycerol, phatidyglycerol, phosphatidylinositol, monogalactosyldiacylglycerol (GL1), and dimannosyldiacylglycerol (GL2). The dominant quinone was MK-9(H2). The peptidoglycan type was A3α with an l-Lys–l-Ala–l-Thr–l-Ala interpeptide bridge. The DNA–DNA hybridization relatedness between strain DCY81T and Arthrobacter siccitolerans LMG 27359T (98.2 %), Arthrobacter sulfonivorans JCM 13520T (97.81 %), Arthrobacter scleromae DSM 17756T (97.59 %), Arthrobacter oxydans KCTC 3383T (97.3 %) was 39.1 ± 0.2, 62.2 ± 1.6, 36.8 ± 1.1 and 48.3 ± 1.6 %, respectively which show that the genotypic separation of strain DCY81T from the closest reference strain of the genus Arthrobacter. The DNA G+C content was 65.2 mol%. The genotypic analysis, physiological, and chemotaxonomic results indicate that strain DCY81T represents a novel species of the genus Arthrobacter. Therefore, Arthrobacter ginsengisoli sp. nov., is proposed as the type strain (=KCTC 29225T = JCM 19357T).  相似文献

The present study was planned to evaluate Co(II) toxicity in silkworm population. The soil was irrigated using synthetic wastewater to determine the effects of pH and initial cobalt concentration in its bioaccumulation in silkworm (Bombyx mori L.) food chain. The amount of cobalt in wastewater, soil, mulberry and silkworm was determined by atomic absorption spectrophotometer (AAS) analysis. The obtained results clearly indicate that silkworm can be used as template to indicate local cobalt pollution as its body length, body weight and mortality rate was found to be strongly related to cobalt concentration. Higher the cobalt amount in mulberry leaves more the toxicity to silkworm population. At 400 mg/L Co concentration and pH 4 there was maximum deposition of Co in the soil from the synthetic effluent. However, in plants and silk worm the accumulation of Co was maximum at pH 4.5 at an initial Co concentration of 400 mg/L in the synthetic effluent. The maximum cobalt found in wastewater, soil, mulberry and silkworm was 400 ± 0.01, 273.5 ± 0.04, 42.85 ± 0.01, 36.62 ± 0.22 mg/kg, respectively.  相似文献

Casein Kinase II (CK2) is a ubiquitous serine/threonine kinase that is highly conserved in eukaryotic cells. CK2 has been shown to impact cell growth and proliferation, as numerous growth-related proteins are substrates of CK2. More importantly, experimental evidence linking increased expression and activity of CK2 to human cancers underscores the relevance of CK2 biology to cellular transformation and carcinogenesis. Due to the critical regulatory role CK2 plays in cell fate determination in cancer cells, there is a tremendous interest in the development of CK2-specific therapies. Supporting this, recent reports have demonstrated that genetic manipulation of CK2 expression as well as pharmacological inhibition of its enzymatic activity sensitizes cancers to apoptotic stimuli. Here we provide a succinct account of the biology of CK2, its cellular substrates, its pro-survival and pro-proliferation activity, and highlight evidence for its involvement in human cancer.  相似文献

The objective of the present study was to develop a once-daily sustained-release (SR) matrix tablet of famotidine. Nine different formulations (F1–F9) were prepared by direct compression method using Avicel PH101 as filler/binder in the range of 41–27% in F1–F3, 18–22% in F4–F7, and 16–18% in F8–F9 and hydroxypropyl methylcellulose (4,000 cps) as hydrophilic matrix was used in F1–F3 from 19% to 30%, around 40% in F4–F7, and 42–45% in F8–F9. Talc and Aerosil were added in the ratio of 0.7–1.2%. The tablets were subjected to various physical parameters including weight variation test, hardness, thickness, diameter, friability, and in vitro release studies. Assay was also performed according to the USP 30 NF 25 procedure. The results of the physical parameters and assay were found to be within the acceptable range. In vitro dissolution results indicated that formulation F4–F7, having around 40% of rate control polymer, produced a SR pattern throughout 24 h. F1–F3 showed drug release at a faster rate, while F8–F9 released much slower, i.e., <80% in 24 h. Model-dependent and model-independent methods were used for data analysis and the best results were observed for F4 in zero order (r 2 = 0.984) and F6 in Korsmeyer and Higuchi (r 2 = 0.992 and 0.988). The parameter n indicated anomalous diffusion, while β in Weibull showed a parabolic curve with higher initial slope. The f 2 similarity test was performed taking F4 as a reference formulation. Only the F5–F7 formulations were similar to the reference formulation F4. The mean dissolution time was around 10 h for the successful formulation.  相似文献
