Fructose 1,6-bisphosphatase (Fru-P2ase,EC 3.1.3.11) purified from livers of fed rabbits has been reported to contain tryptophan, which is not present in the enzyme purified from livers of fasted animals. We now show that the tryptophan arises from small amounts of active or inactive rabbit liver aldolase in the Fru-P2ase preparations. Fru-P2ase free of tryptophan may conveniently be prepared by raising the temperature of the heat step in the purification procedure to 67 °C. 相似文献

Infrared spectroscopy is used to characterize the transitions in the photocycle of bR involving the M intermediate. It has been shown previously that in this part of the photocycle a large protein conformational change takes place that is important for proton pumping. In this work we separate the spectra of the L, M, and N intermediates in order to better describe the timing of the molecular changes. We use the photoreaction of the M intermediate to separate its spectrum from those of L and N. At temperatures between 220 and 270 K a mixture of M and L or N is produced by illumination with green light. Subsequent blue illumination selectively drives M back into the ground state and the difference between the spectra before and after blue excitation yields the spectrum of M. Below about 250 K and L/M mixture is separated; at higher temperatures an M/N mixture is seen. We find that the spectrum of M is identical in the two temperature regions. The large protein conformational change is seen to occur during the M to N transition. Our results confirm that Asp-96 is transiently deprotonated in the L state. The only aspartic protonation changes between M and bR are the protonation of Asp-85 and Asp-212 that occur simultaneously during the L to M transition. Blue-light excitation of M results in deprotonation of both. The results suggest a quadrupolelike interaction of the Schiff base, Asp-85, Asp-212, and an additional positive charge in bR. 相似文献

The dynamic behavior of a laboratory-scale activated sludge biological waste treatment process with recycle and wasting of sludge was investigated by subjecting the system to step changes in the influent waste concentration, the recycle flow rate, or the sludge wasting rate. The dynamic behavior of the system was examined by measuring adenosine triphosphate (ATP) in addition to dissolved chemical oxygen demand (COD) and cell dry weight in the aeration tank. Cell dry weight of the recyle flow and effluent COD were also measured. Analysis of the results and estimation of time constants assuming first order responses showed that the time constants characterizing the dynamic responses of the sludge were directly related to the sludge mean residence time. The time constants estimated from dissolved COD measurements were of the same order of magnitude as the fluid residence time in the aeration tank. The ATP transient response was frequently different from that of the cell dry weight in the aeration tank. 相似文献

A clinically diagnosed case of progressive external ophthalmoplegia was biopsied and ultrastructurally evaluated. The disease was found to be characterized by numerous giant mitochondria. The intramitochondrial changes consisted of either swirls of cristae or of paracrystalline structures which were in apposition with the membranes of the cristae. Homologous DNA fragments were prepared from cloned cDNAs for the pro-alpha 1(I) and pro-alpha 2(I) chains of human type I procollagen. The DNA fragments were then used to develop a dot blot hybridization assay for mRNAs for pro-alpha 1(I) and pro-alpha 2(I) chains in skin fibroblasts. In normal fibroblasts, the ratio of the steady state levels of the two mRNAs was 1.94 +/- 0.34 S.D. The ratio for the rates of synthesis of the two pro-alpha chains in the same cells was 1.84 +/- 0.13 S.D. Since the two ratios were essentially the same, the results indicated that the mRNAs for the two chains are translated at about the same rates. Therefore, there is no need to invoke translational control or more complex mechanisms to explain synthesis of pro-alpha 1(I) and pro-alpha 2(I) chains in a stoichiometry of 2:1. The dot blot hybridization assay was also used to examine the levels of the mRNAs in fibroblasts from several variants of osteogenesis imperfecta. In two of the variants, the ratios of the steady state levels of mRNAs for pro-alpha 1(I) and pro-alpha 2(I) chains were 3.05 and 2.52, respectively. In the same fibroblasts, the ratios for the rates of synthesis of the two chains were 2.99 +/- 0.43 and 2.45 +/- 0.16, respectively. Therefore, even though the ratios of the levels of the two mRNAs in the fibroblasts were abnormal, the two mRNAs were still translated at the same rates, and there was no evidence of differential regulation at the translational level. 相似文献

Two component systems (TCSs) are a primary mechanism of signal sensing and response in bacteria. Systematic characterization of an entire TCS could provide a mechanistic understanding of these important signal transduction systems. Here, genetic selections were employed to dissect the molecular basis of signal transduction by the HitRS system that detects cell envelope stress in the pathogen Bacillus anthracis. Numerous point mutations were isolated within HitRS, 17 of which were in a 50-residue HAMP domain. Mutational analysis revealed the importance of hydrophobic interactions within the HAMP domain and highlighted its essentiality in TCS signaling. In addition, these data defined residues critical for activities intrinsic to HitRS, uncovered specific interactions among individual domains and between the two signaling proteins, and revealed that phosphotransfer is the rate-limiting step for signal transduction. Furthermore, this study establishes the use of unbiased genetic selections to study TCS signaling and provides a comprehensive mechanistic understanding of an entire TCS. 相似文献
