The search for biomarkers to diagnose psychiatric disorders such as schizophrenia has been underway for decades. Many molecular profiling studies in this field have focused on identifying individual marker signals that show significant differences in expression between patients and the normal population. However, signals for multiple analyte combinations that exhibit patterned behaviors have been less exploited. Here, we present a novel approach for identifying biomarkers of schizophrenia using expression of serum analytes from first onset, drug-naïve patients and normal controls. The strength of patterned signals was amplified by analyzing data in reproducing kernel spaces. This resulted in the identification of small sets of analytes referred to as targeted clusters that have discriminative power specifically for schizophrenia in both human and rat models. These clusters were associated with specific molecular signaling pathways and less strongly related to other neuropsychiatric disorders such as major depressive disorder and bipolar disorder. These results shed new light concerning how complex neuropsychiatric diseases behave at the pathway level and demonstrate the power of this approach in identification of disease-specific biomarkers and potential novel therapeutic strategies.Schizophrenia is a debilitating neuropsychiatric disorder that affects more than 1% of the world population and costs hundreds of billions of United States dollars in healthcare provision and lost earnings (1). The diagnosis of this disease has not changed substantially over several decades and currently relies on subjective psychopathological ratings such as the Diagnostic and Statistical Manual of Mental Disorders (DSM)1-IV. Thus, diagnosis can be complicated by the presence of overlapping symptoms frequently occurring in other psychiatric illnesses such as bipolar disorder (BD) and major depressive disorder (MDD) and by the presence of confounding factors such as drug abuse and co-morbidities. This often results in diagnosis being delayed for several months to years. A delay in establishing an accurate diagnosis can have serious deleterious implications because a late or imprecise diagnosis can contribute to unsatisfactory outcomes to currently used drug therapies and to higher rates of relapse (2). Most importantly, more than half of schizophrenia subjects develop a progressive course of the disease associated with deficit symptoms (3).In contrast, early therapeutic intervention holds promise in preventing or diminishing such effects (4–6). An empirical assay for early and accurate diagnosis of schizophrenia would deliver improved patient outcomes and reduce the costs of the disease for healthcare services and society (7–9). Such an assay could also provide a means of stratifying patients and monitoring drug responses and may also lead to the development of translational medicine tools that are critical for discovery of novel therapeutic strategies. Molecular profiling methods that afford the simultaneous measurement of multiple analytes in clinical and preclinical samples have considerable promise in this endeavor. These methods have been aimed predominantly at identifying individual molecules that show differences in expression between the disease and control conditions. However, such studies have often been fraught with small fold-changes in analyte levels, a common obstacle when investigating complex neuropsychiatric disorders (10–12). Thus, standard statistical techniques such as t tests will not be able to explore patterned behaviors involving proteins that have subtle expression changes but still contribute to the development of schizophrenia.The main objective of this study was to determine whether unique patterns of biomarkers can be identified for subjects with first onset antipsychotic-naïve schizophrenia. Analyte expression lists were generated using the Multi-Analyte Profiling (MAP®) fluorescent bead-based technology for profiling serum samples from 77 male schizophrenia patients and 66 matched male controls. For comparison with other psychiatric disorders, we also analyzed the serum samples of 13 male BD and 17 male MDD patients. In parallel, serum samples from four relevant animal models were also profiled for comparison with the human disease state. Analysis of the respective expression lists was carried out using non-linear statistical analysis, which identifies small sets of analytes called targeted analyte clusters (TACs) that have the power to discriminate the patients from normal controls. We present here the performance of these clusters for diagnosis of schizophrenia. In addition, we show how this method can also contribute to increasing our understanding of the etiology of the disorder by determining its ability to classify various preclinical models of psychiatric disorders. We have used multiplexed high-throughput sequencing to characterize changes in small RNA populations that occur during viral infection in animal cells. Small RNA-based mechanisms such as RNA interference (RNAi) have been shown in plant and invertebrate systems to play a key role in host responses to viral infection. Although homologs of the key RNAi effector pathways are present in mammalian cells, and can launch an RNAi-mediated degradation of experimentally targeted mRNAs, any role for such responses in mammalian host-virus interactions remains to be characterized. Six different viruses were examined in 41 experimentally susceptible and resistant host systems. We identified virus-derived small RNAs (vsRNAs) from all six viruses, with total abundance varying from “vanishingly rare” (less than 0.1% of cellular small RNA) to highly abundant (comparable to abundant micro-RNAs “miRNAs”). In addition to the appearance of vsRNAs during infection, we saw a number of specific changes in host miRNA profiles. For several infection models investigated in more detail, the RNAi and Interferon pathways modulated the abundance of vsRNAs. We also found evidence for populations of vsRNAs that exist as duplexed siRNAs with zero to three nucleotide 3′ overhangs. Using populations of cells carrying a Hepatitis C replicon, we observed strand-selective loading of siRNAs onto Argonaute complexes. Colonization of roots by Pseudomonas chlororaphis O6 ( Pc O6) involves root surface coverage through surface motility and biofilm formation. Root colonization and the production of antifungal phenazines are important in the ability of the bacterium to protect plants against pathogens. In this in vitro study we report that both biofilm formation and phenazine production are differentially influenced by nutrition and the presence of polyethylene oxide/polypropylene oxide triblock copolymer surfactants (Pluronics). Such surfactants are used for many purposes including agricultural formulations. Four Pluronics differing in molecular weight and in hydrophobic/hydrophilic proportions had distinct effects on biofilm formation and secondary metabolite production, although each increased surface motility, termed swarming, to a similar extent. These findings show that Pluronics had specific metabolic impacts on the bacterium, where both up- and downregulation was achieved depending on the medium and the Pluronic composition. In environmental and agricultural settings, Pluronics may have unanticipated effects on soil microorganisms, while in bioprocessing these effects may be leveraged to regulate metabolite yield. 相似文献

HLA B57 and the closely related HLA B5801 are over-represented among HIV-1 infected long-term nonprogressors (LTNPs). It has been suggested that this association between HLA B57/5801 and asymptomatic survival is a consequence of strong CTL responses against epitopes in the viral Gag protein. Moreover, CTL escape mutations in Gag would coincide with viral attenuation, resulting in low viral load despite evasion from immune control. In this study we compared HLA B57/5801 HIV-1 infected progressors and LTNPs for sequence variation in four dominant epitopes in Gag and their ability to generate CTL responses against these epitopes and the autologous escape variants. Prevalence and appearance of escape mutations in Gag epitopes and potential compensatory mutations were similar in HLA B57/5801 LTNPs and progressors. Both groups were also indistinguishable in the magnitude of CD8+ IFN-gamma responses directed against the wild-type or autologous escape mutant Gag epitopes in IFN-gamma ELISPOT analysis. Interestingly, HIV-1 variants from HLA B57/5801 LTNPs had much lower replication capacity than the viruses from HLA B57/5801 progressors, which did not correlate with specific mutations in Gag. In conclusion, the different clinical course of HLA B57/5801 LTNPs and progressors was not associated with differences in CTL escape mutations or CTL activity against epitopes in Gag but rather with differences in HIV-1 replication capacity. 相似文献

Brain abscesses form in response to a parenchymal infection by pyogenic bacteria, with Staphylococcus aureus representing a common etiologic agent of human disease. Numerous receptors that participate in immune responses to bacteria, including the majority of TLRs, the IL-1R, and the IL-18R, use a common adaptor molecule, MyD88, for transducing activation signals leading to proinflammatory mediator expression and immune effector functions. To delineate the importance of MyD88-dependent signals in brain abscesses, we compared disease pathogenesis using MyD88 knockout (KO) and wild-type (WT) mice. Mortality rates were significantly higher in MyD88 KO mice, which correlated with a significant reduction in the expression of several proinflammatory mediators, including but not limited to IL-1beta, TNF-alpha, and MIP-2/CXCL2. These changes were associated with a significant reduction in neutrophil and macrophage recruitment into brain abscesses of MyD88 KO animals. In addition, microglia, macrophages, and neutrophils isolated from the brain abscesses of MyD88 KO mice produced significantly less TNF-alpha, IL-6, MIP-1alpha/CCL3, and IFN-gamma-induced protein 10/CXCL10 compared with WT cells. The lack of MyD88-dependent signals had a dramatic effect on the extent of tissue injury, with significantly larger brain abscesses typified by exaggerated edema and necrosis in MyD88 KO animals. Interestingly, despite these striking changes in MyD88 KO mice, bacterial burdens did not significantly differ between the two strains at the early time points examined. Collectively, these findings indicate that MyD88 plays an essential role in establishing a protective CNS host response during the early stages of brain abscess development, whereas MyD88-independent pathway(s) are responsible for pathogen containment. 相似文献

Spatial features of pollen tube growth and the composition of the extracellular matrix (ECM) of transmitting tissue in carpels of Kadsura longipedunculata, a member of the basal angiosperm taxon Schisandraceae, were characterized to identify features of transmitting tissue that might have been important for pollen-carpel interactions during the early history of angiosperms. In addition to growing extracellularly along epidermal cells that make up stigmatic crests of individual carpels, pollen tubes grow on abaxial carpel epidermal cells between unfused carpels along an extragynoecial compitum to subsequently enter an adjacent carpel, a feature important for enhancing seed set in apocarpous species. Histo- and immunochemical data indicated that transmitting tissue ECM is not freely flowing as previously hypothesized. Rather, the ECM is similar to that of a dry-type stigma whereby a cuticular boundary with associated esterase activity confines a matrix containing methyl-esterified homogalacturonans. The Schisandraceae joins an increasing number of basal angiosperm taxa that have a transmitting tissue ECM similar to a dry-type stigma, thereby challenging traditional views that the ancestral pollen tube pathway was similar to a wet-type stigma covered with a freely flowing exudate. To identify the pharmacophore of a phosphoramidate peptidomimetic inhibitor of prostate-specific membrane antigen (PSMA), a small analog library was designed and screened for inhibitory potency against PSMA. The design of the lead inhibitor was based upon N-acyl derivatives of endogenous substrate folyl-gamma-Glu and incorporates a phosphoramidate group to interact with the PSMA catalytic zinc atoms. The scope of the analog library was designed to test the importance of various functional groups to the inhibitory potency of the lead phosphoramidate. The IC(50) for the lead phosphoramidate inhibitor was 35 nM while the IC(50) values for the analog library presented a range from 0.86 nM to 4.1 microM. Computational docking, utilizing a recently solved X-ray crystal structure of the recombinant protein, along with enzyme inhibition data, was used to propose a pharmacophore model for the PSMA active site. 相似文献
