Peripheral administration of the endogenous Y(2) and Y(4) receptor selective agonists, PYY(3-36) and PP, have been shown to inhibit food intake and body weight gain in rodents, and to reduce appetite and caloric intake in humans. We have previously developed a long-acting, potent and highly selective Y(2) receptor selective agonist, N-alpha-Ac-[Nle(24,28), Trp(30), Nva(31), Psi(35-36)]PYY(22-36)-NH(2) (BT-48). BT-48 (ip) dose-dependently inhibited ad lib food intake and also decreased the respiratory quotient in mice during both the light and dark periods. The latter observation is indicative of enhanced fat metabolism. Moreover, BT-48 also inhibited food intake in fasted mice. Combined ip administration of BT-48 (50nmol/mouse) with a highly potent and selective Y(4) anorectic peptide, BVD-74D (50nmol/mouse), resulted in a powerful and long lasting inhibitory effect on food intake. As expected, this inhibitory effect on food intake was nearly double that exhibited by either peptide (50nmol/mouse) alone. In summary, BT-48, unlike PYY(3-36), exhibits little or no affinity to other "Y" receptors, and may therefore have a better clinical potential than PYY(3-36) for control of food intake. Moreover, it appears that treatment with a combination of Y(2) and Y(4) receptor selective agonists may constitute a more powerful approach to control food intake than treatment with either of these agonists alone.  相似文献

A diatomaceous earth formulation enhanced with bitterbarkomycin (DEBBM) combined with Beauveria bassiana (Balsamo) Vuillemin was evaluated against lesser grain borer Rhyzopertha dominica F. (Coleoptera: Bostrychidae) under laboratory conditions. DEBBM was applied at the rates of 15 and 30 ppm alone as well as in combination with 6.69 × 106, 6.69 × 108 and 6.69 × 1010 conidia/kg of wheat. Mortality of treated adults was recorded after 5, 10 and 15 days of exposure. Bioassays were carried out at 20, 25 and 30°C with 55 and 75% relative humidity. The emergence of progeny was also assessed 60 days post exposure. The combined use of DEBBM and B. bassiana considerably increased adult mortality especially at increasing temperatures and longer exposure intervals compared with DEBBM and B. bassiana alone. Progeny production was less in wheat treated with high dose rates of DEBBM +B. bassiana. The per cent mycosis in the cadavers was maximum where B. bassiana was applied at low dose rates. The results of the present study indicated that a combination of DEBBM and B. bassiana may provide effective control of R. dominica.  相似文献

The efficacy of one new modified and two old meridic diets on Helicoverpa armigera Hübner (Lepidoptera: Noctuidae) for rearing six successive generations was studied. Duration of larval development for insects fed on the modified diet was considerably shortened as most of them went through only five stadia before pupation, while the per cent pupation and per cent eclosion were relatively higher than on other diets. The lowest pupal mortality (6.33 ± 0.13%) was recorded in the F1 generation reared on the modified diet, whereas the highest pupal mortality (19.49 ± 0.15%) was observed in insects reared on a natural diet in the F6 generation. Blending of chickpea Cicer arietinum L. and red kidney bean Phaseolus vulgaris L. flours with tomato paste proved highly favorable for adult reproduction. These results suggest that the vitality of the tomato fruitworm did not decline obviously after rearing on a modified diet for several generations.  相似文献

The polysaccharide structure of potato cell walls: Chemical fractionation   总被引:6,自引:0,他引:6  
Cell walls of potato tubers were fractionated by successive extraction with various reagents. A slightly degraded pectic fraction with 77% galacturonic acid was extracted in hot, oxalate-citrate buffer at pH 4. A further, major pectic fraction with 38% galacturonic acid was extracted in cold 0.1 M Na2CO3 with little apparent degradation. These two pectic fractions together made up 52% of the cell wall. Most of the oxalate-citrate fraction could alternatively be extracted with cold acetate-N,N,N-tetracetic acid (CDTA) buffer, a non-degradative extractant which nevertheless removed essentially all the calcium ions. This fraction was therefore probably held only by calcium binding, and the remainder of the pectins by covalent bonds. Electrophoresis showed that both pectic fractions contained a range of molecular types differing in composition, with a high arabinose: galactose ratio as well as much galacturonic acid in the most extractable fractions. From methylation data, the main side-chains were 1,4-linked galactans and 1,5-linked arabinans, with smaller quantities of covalently attached xyloglucan. Extraction with NaOH-borate removed a small hemicellulose fraction and some cellulose. Properties of large unilamellar vesicles (LUV), composed of phosphatidylcholine and prepared by reverse-phase evaporation and subsequent extrusion through Unipore polycarbonate membranes, have been investigated and compared with those of small unilamellar vesicles (SUV) and of multilamellar vesicles (MLV). The unilamellar nature of the LUV is shown by 1H-NMR using Pr3+ as a shift reagent. The gel to liquid-crystalline phase transition of LUV composed of dipalmitoylphosphatidylcholine (DPPC) monitored by differential scanning calorimetry, fluorescence polarization of diphenylhexatriene and 90 degrees light scattering, occurs at a slight lower temperature (40.8 degrees C) than that of MLV (42 degrees C) and is broadened by about 50%. The phase transition of SUV is shifted to considerably lower temperatures (mid-point, 38 degrees C) and extends over a wide temperature range. In LUV a well-defined pretransition is not observed. The permeability of LUV (DPPC) monitored by leakage of carboxyfluorescein, increases sharply at the phase transition temperature, and the extent of release is greater than that from MLV. Leakage from SUV occurs in a wide temperature range. Freeze-fracture electron microscopy of LUV (DPPC) reveals vesicles of 0.1-0.2 micron diameter with mostly smooth fracture faces. At temperatures below the phase transition, the larger vesicles in the population have angled faces, as do extruded MLV. A banded pattern, seen in MLV at temperatures between the pretransition and the main transition, is not observed in the smaller LUV, although the larger vesicles reveal a dimpled appearance.  相似文献

Previous reports of adrenal progesterone (P4) contributions during late gestation in cattle, and ACTH-induced P4 responses in the non-pregnant heifer, prompted a retrospective investigation to evaluate the plasma P4 response and the relative ratio of plasma cortisol (CT) to P4 following ACTH administration during mid-gestation in pregnant Brahman heifers. Twenty-three pregnant (139.0 +/- 5.0 days of gestation) Brahman heifers received one of the following treatments: 0 (saline; n = 5), 0.125 (n = 4), 0.25 (n = 5), 0.5 (n = 4), or 1.0 (n = 5)IU of ACTH per kg BW. Blood samples were collected at -15 and -0.5 (time 0), 15, 30, 45, 60, 75, 105, 135, 165, 195, and 255-min post-ACTH challenge. Plasma P4 and CT were quantified by RIA. Pre-ACTH P4 did not differ (P > 0.10) among ACTH treatment groups (pooled, 12.1 +/- 0.6 ng/mL). Among peak P4 values at 15-min post-ACTH infusion, control P4 (9.6 +/- 1.2 ng/mL) tended to be lower (P < 0.07) than 0.5 IU ACTH-treated heifers (13.3 +/- 1.1 ng/mL); and were lower (P < 0.02) than 0.25 and 1.0 IU ACTH-treated heifers (14.7 +/- 1.1 and 22.2 +/- 3.7 ng/mL, respectively). During the primary P4 response period (0 to 75-min post-ACTH), the area under the curve (AUC) was greater (P < 0.05) for 1.0 IU ACTH-treated heifers than all other groups. The CT:P4 ratios were lower (time x treatment, P < 0.01) for control heifers than all ACTH-treated heifers. Among ACTH-treated heifers, CT:P4 ratio response and CT:P4 ratio AUC were similar (P > 0.10) following ACTH challenge. In conclusion, acute increases in ACTH elevated plasma P4, likely of adrenal origin, in mid-gestation pregnant heifers, while the CT:P4 ratio (relative output) remained constant irrespective of ACTH dose (0.125-1.0 IU). Whether ACTH-induced increases in P4 in pregnant animals are of physiological significance (e.g., an accessory role in the maintenance of pregnancy during periods of acute stress) remains to be determined.  相似文献

The objective of the investigation was to evaluate the effect of immobilizing substances and NaCl salinity on the availability of heavy metals: Zn, Cd, Cu, Ni, and Pb to wheat (Triticum aestivum L.). In greenhouse pot experiment, a sewage sludge amended soil was treated with the following immobilizing substances: three clay minerals (Na-bentonite, Ca-bentonite and zeolite), iron oxides (goethite and hematite), and phosphate fertilizers (superphosphate and Novaphos). The pots were planted with wheat and were irrigated either with deionized or saline water containing 1600 mg L?1 NaCl. Wheat was harvested two times for shoot metal concentrations and biomass measurements. Metal species in soil solution were estimated using the software MINEQL+.

The addition of metal immobilizing substances to the soil significantly decreased metal availability to wheat. The largest reduction in metal bioavailability was found for bentonites. The irrigation with saline water (1600 mg L?1 NaCl) resulted in a significant increase in metal chloride species (MCl+ and MCl2 0). The highest metal complexation with Cl occurred for Cd, which was about 53% of its total soil solution concentration. The total concentration of Cd (CdT) in soil solution increased by 1.6–2.8-fold due to saline water. The NaCl salinity caused a significant increase in uptake and shoot concentration of Cd for two harvests and small but significant increase in shoot Pb concentration for the second harvest. It was concluded that the use of bentonites is the most promising for the reduction of heavy metal availability to plants. Saline water containing 1600 mg L? 1 NaCl increased the availability of Cd and Pb to wheat and decreased the efficiency of bentonites to immobilize soluble Cd.  相似文献

Trichinella spiralis infection elicits a vigorous IgE response and pronounced intestinal and splenic mastocytosis in mice. Since IgE both activates mast cells (MC) and promotes their survival in culture, we examined its role in MC responses and parasite elimination in T. spiralis-infected mice. During primary infection, wild-type but not IgE-deficient (IgE(-/-)) BALB/c mice mounted a strong IgE response peaking 14 days into infection. The splenic mastocytosis observed in BALB/c mice following infection with T. spiralis was significantly diminished in IgE(-/-) mice while eosinophil responses were not diminished in either the blood or jejunum. Similar levels of peripheral blood eosinophilia and jejunal mastocytosis occurred in wild-type and IgE-deficient animals. Despite the normal MC response in the small intestine, serum levels of mouse MC protease-1 also were lower in parasite-infected IgE(-/-) animals and these animals were slower to eliminate the adult worms from the small intestine. The number of T. spiralis larvae present in the skeletal muscle of IgE(-/-) mice 28 days after primary infection was about twice that in BALB/c controls, and the fraction of larvae that was necrotic was reduced in the IgE-deficient animals. An intense deposition of IgE in and around the muscle larvae was observed in wild-type but not in IgE null mice. We conclude that IgE promotes parasite expulsion from the gut following T. spiralis infection and participates in the response to larval stages of the parasite. Furthermore, our observations support a role for IgE in the regulation of MC homeostasis in vivo.  相似文献
