The critical importance of cytoskeletal function for correct neuronal migration during development of the cerebral cortex has been underscored by the identities of germline mutations underlying a number of human neurodevelopmental disorders. The proteins affected include TUBA1A, a major α-tubulin isoform, and microtubule-associated components such as doublecortin, and LIS1. Mutations in these genes are associated with the anatomical abnormality lissencephaly, which is believed to reflect failure of neuronal migration. An important recent observation has been the dependence of cortical neuronal migration upon acetylation of α-tubulin at lysine 40 by the histone acetyltransferase Elongator complex. Here, we describe a recognizable autosomal recessive syndrome, characterized by generalized polymicrogyria in association with optic nerve hypoplasia (PMGOH). By autozygosity mapping, we show that the molecular basis for this condition is mutation of the TUBA8 gene, encoding a variant α-tubulin of unknown function that is not susceptible to the lysine 40 acetylation that regulates microtubule function during cortical neuron migration. Together with the unique expression pattern of TUBA8 within the developing cerebral cortex, these observations suggest a role for this atypical microtubule component in regulating mammalian brain development.  相似文献

Barley (Hordeum vulgare L.) seeds were soaked in aqueous 10−4 M dihydroquercetin (DHQ) to examine its influence on seed germination and further growth of seedlings under optimal soil watering and flooding conditions. The adaptive potential of the plants was estimated by the content of thiobarbituric acid reactive substances (TBARs) and the activity of ascorbate peroxidase (AsP). High-grade seeds were germinated evenly under (−DHQ)- and (+DHQ)-treatments. Low-grade seeds soaked in DHQ, showed no mold and twofold germination rate in comparison with the same seeds soaked in water. The seedlings grown from the similarly germinated seeds did not differ from each other in the shoot growth, independent of the DHQ-pretreatment. The root growth was higher in DHQ-pretreated plants. Soil flooding suppressed the shoot and root growth rates in non-pretreated and DHQ-pretreated plants, however TBARs content was lower in the roots and leaves of (+DHQ)-seedlings as compared to the (−DHQ)-ones. The activity of AsP increased more significantly in the (+DHQ)-plants. The ratio between TBARs content and the AsP activity was lower in the leaves of (+DHQ)-plants both under optimal soil conditions and flooding. Thus, the treatment of low-grade barley seeds with DHQ protects the seeds against mold and increases adaptive potential of the seedlings.  相似文献

The vertebrate olfactory system has fascinated neurobiologists over the last six decades because of its ability to replace its neurons and synaptic connections continuously throughout adult life, under both physiological and pathological conditions. Among the factors that are proposed to be involved in this regenerative potential, brain-derived neurotrophic factor (BDNF) is a candidate for having an important role in the neuronal turnover in the olfactory epithelium (OE) because of its well-documented neurogenic and trophic effects throughout the nervous system. The aim of the present study was to generate a suitable model to study the participation of BDNF in the recovery of the OE after injury in vivo. We developed an experimental design in which the OE of Rhinella arenarum tadpoles could be easily and selectively damaged by immersing the animals in ZnSO4 solutions of various concentrations for differing time periods. Image analysis of histological sections showed that different combinations of each of these conditions produced statistically different degrees of injury to the olfactory tissue. We also observed that the morphology of the OE was restored within a few days of recovery after ZnSO4 treatment. Immunohistochemical analysis of BDNF was performed with an antiserum whose specificity was confirmed by Western blotting, and which showed drastic changes in the abundance and distribution pattern of this neurotrophin in the damaged olfactory system. Our results thus suggest that BDNF is involved in the regeneration of the OE of amphibian larvae, and that our approach is suitable for further investigations of this topic. The trajectory of hurricane-induced succession was evaluated in a network of forest plots measured immediately before and 3 mo, 5, 10, and 15 yr after the direct impact of a Category 4 hurricane. Comparisons of forest structure, composition, and aboveground nutrients pools were made through time, and between species, life-history groups and geomorphic settings. The hurricane reduced aboveground biomass by 50 percent, causing an immediate decrease in stem density and diversity indices among all geomorphic settings. After 15 yr, basal area and aboveground biomass returned to pre-hurricane levels, while species richness, diversity indices, and stem densities exceeded pre-hurricane levels. Differences in species composition among geomorphic settings had not returned after 15 yr but differences in stem densities and structure were beginning to emerge. Significant differences were observed in the nutrient concentration of the three species that comprised the most aboveground biomass, and between species categorized as secondary high-light species and primary, low-light species. Species whose abundance was negatively correlated with the mature forest dominant also had distinct nutrient concentrations. When total aboveground nutrient pools were compared over time, differences in leaf nutrients among species were hidden by similarities in wood nutrient concentrations and the biomass dominance of a few species. The observed successional trajectory indicates that changes in species composition contributed to fast recovery of aboveground biomass and nutrient pools, while the influence of geomorphic setting on species composition occurs at time scales >15 yr of succession.  相似文献

Bone morphogenetic signaling (BMP) is a key pathway during neurogenesis and depends on many downstream intermediators to carry out its signaling. One such signaling pathway utilizes neurotrophin receptor-interacting MAGE protein (NRAGE), a member of the melanoma-associated antigen (MAGE) family, to upregulate p38 mitogen activated protein kinase (p38MAPK) in response to cellular stress and activate caspases which are critical in leading cells to death. NRAGE consists of two conserved MAGE homology domains separated by a unique hexapeptide repeat domain. Although we have previously implicated NRAGE in inducing apoptosis in neural progenitors and P19 cells, a model system for neural progenitors, its domains have yet to be explored in determining which one may be responsible for setting up the signaling for apoptosis. Here, we overexpressed a series of deletion mutations in P19 cells to show that only those with at least half of the repeat domain, activated p38MAPK and underwent apoptosis offering intriguing incite into NRAGE’s contribution in BMP apoptotic signaling.  相似文献

Transitional cell carcinoma (TCC) of urinary bladder belongs to glutathione S-transferase P1 (GSTP1) overexpressing tumors. Upregulated GSTP1 in TCC is related to apoptosis inhibition. This antiapoptotic effects of GSTP1 might be mediated through protein:protein interaction with c-Jun NH(2) -terminal kinase (JNK). Herein, we analyzed whether a direct link between GSTP1 and JNK exists in TCC. The presence of GSTP1/JNK complexes was analyzed by immunoprecipitation and Western blotting in 20 TCC specimens, obtained after surgery. Co-localization of GSTP1 and JNK was also investigated in the 5637 TCC cell line by immunofluorescence confocal microscopy. By means of immunoprecipitation we show for the first time the presence of GSTP1/JNK complexes in all TCC samples studied. A co-localization of GSTP1 and JNK was also demonstrated in the 5637 TCC cell line by means of confocal microscopy. Protein-protein interactions, together with co-localization between GSTP1 and JNK provide evidence that GSTP1 most probably inhibits apoptosis in TCC cells by non-covalent binding to JNK.  相似文献

Cocoa bean fermentation is a spontaneous process involving a succession of microbial activities, starting with yeasts, followed by lactic acid bacteria and acetic acid bacteria. So far, all microbiological studies about cocoa bean fermentation were based on culture-dependent (isolation, cultivation, and identification), or, more recently, culture-independent (PCR-DGGE, or polymerase chain reaction denaturing gradient gel electrophoresis) methods. Using a metagenomic approach, total DNA was extracted from heap and box fermentations at different time points and from different locations (Ghana and Brazil, respectively) to generate a 16 S rDNA clone library that was sequenced. The sequencing data revealed a low bacterial diversity in the fermentation samples and were in accordance with the results obtained through culture-dependent and a second, culture-independent analysis (PCR-DGGE), suggesting that almost all bacteria involved in the fermentation process are cultivable. One exception was the identification by 16 S rDNA library sequencing of Gluconacetobacter species of acetic acid bacteria that were not detected by the two other approaches. The presence of Enterobacteriaceae related to Erwinia/Pantoea/Tatumella, as revealed by 16 S rDNA library sequencing, suggests an impact of these bacteria on fermentation.  相似文献
