Three-dimensional (3D) cell culture techniques using a bioreactor have been used to co-culture various breast cancer cell lines. Comparisons between 3D co-cultures containing different proportions of breast cancer cell lines have been made with respect to cluster size, cell surface marker distribution, and Ki67 expression. Furthermore, an observed difference in invasion through collagen between co-cultures has been briefly reported. However, these assays have not yet been developed into a quantifiable methodology to assess the effects of drugs and/or microenvironments on cellular invasion. From a cancer perspective, two important aspects of cellular invasion that are often left out of in vitro assays are considerations about the 3D structural heterogeneity of the primary tumor and the ability of cells to migrate in all directions. Accordingly, we have taken advantage of the methodology previously described for 3D cell culture techniques and have developed a 3D invasion assay using cell clusters that can be used to assess the effects of different drugs and treatment conditions on cancer cell invasion. We also describe a novel whole-mount technique that permits fluorescence-based immunolocalization of proteins through the entire tumorsphere, without the need for sectioning. Our assay provides a simple, inexpensive, and physiologically relevant context to study cellular invasion in vitro, in a way that recapitulates an in vivo milieu. 相似文献

Glycosyltransferases are specific enzymes that catalyse the transfer of monosaccharide moieties to biological substrates, including proteins, lipids and carbohydrates. These enzymes are present from prokaryotes to humans, and their glycoconjugate products are often vital for survival of the organism. Many glycosyltransferases found in fungal pathogens such as Cryptococcus neoformans do not exist in mammalian systems, making them attractive potential targets for selectively toxic agents. In this article, we present the features of this diverse class of enzymes, and review the fungal glycosyltransferases that are involved in synthesis of the cell wall, the cryptococcal capsule, glycoproteins and glycolipids. We specifically focus on enzymes that have been identified or studied in C. neoformans, and we consider future directions for research on glycosyltransferases in the context of this opportunistic pathogen. 相似文献

Intraspecific density regulation influences the synchronization of local population dynamics through dispersal. Spatial synchrony in turn may jeopardize metapopulation persistence. Joining results from previous studies suggests that spatial synchrony is highest at moderate over-compensation and is low at compensating and at very strong over-compensating density regulation. We scrutinize this supposition of a unimodal relationship using a process-based metapopulation model with explicit local population dynamics. We extend the usually studied range of density regulation to under-compensation and analyse resulting metapopulation persistence. We find peaks of spatial synchrony not only at over-compensatory but also under-compensatory density regulation and show that effects of local density compensation on synchrony follow a bimodal rather than unimodal relationship. Persistence of metapopulations however, shows a unimodal relationship with a broad plateau of high persistence from compensatory to over-compensatory density regulation. This range of high persistence comprises both levels of low and high spatial synchrony. Thus, not synchrony alone jeopardizes metapopulation persistence, but only in interplay with high local extinction risk. The functional forms of the relations of density compensation with spatial synchrony and persistence are robust to increases in dispersal mortality, landscape dynamics, or density dependence of dispersal. However, with each of these increases the maxima of spatial synchrony and persistence shift to higher over-compensation and levels of synchrony are reduced. Overall, for over-compensation high landscape connectivity has negative effects while for under-compensation connectivity affects persistence positively. This emphasizes the importance of species life-history traits for management decisions with regard to landscape connectivity: while dispersal corridors are essential for species with under-compensatory density regulation, they may have detrimental effects for endangered species with over-compensation. 相似文献

Phytoremediation and its efficiency--influenced by several factors-is prime concern in reduction of environment contamination with heavy metals. The aim of the study was to estimate in controlled conditions--in relation to the natural Ca/Mg ratio 4:1--the influence of different Ca/Mg ratios (1:1/4, 20:1, 1:10) on selected heavy metals (Cd, Cu, Pb, and Zn) accumulation efficiency shown by three morphological parts of Salix viminalis 'Cinamomea' (cane bark, shoots and roots). Accumulation of heavy metals in experimental conditions was significantly Ca/Mg ratio dependent, and occurred in the following order: 1:10 > 4:1 > 20:1 > 1:1/4. The highest accumulation under 0.5 mM solutions (as compared to 0.1 mM) was noted for copper and zinc, and the lowest for cadmium. Biometric parameters of Salix viminalis cuttings revealed the highest biomass productivity under 20:1 and 4:1 Ca/Mg ratios. In the case of shoot length increase, both metal concentration and kind of metal present in the solution were important. The results indicate that higher calcium ion concentration in solution (Ca/Mg 20:1) stimulates Salix viminalis growth by 3 fold when compared to other Ca/Mg ratio. The trajectory of hurricane-induced succession was evaluated in a network of forest plots measured immediately before and 3 mo, 5, 10, and 15 yr after the direct impact of a Category 4 hurricane. Comparisons of forest structure, composition, and aboveground nutrients pools were made through time, and between species, life-history groups and geomorphic settings. The hurricane reduced aboveground biomass by 50 percent, causing an immediate decrease in stem density and diversity indices among all geomorphic settings. After 15 yr, basal area and aboveground biomass returned to pre-hurricane levels, while species richness, diversity indices, and stem densities exceeded pre-hurricane levels. Differences in species composition among geomorphic settings had not returned after 15 yr but differences in stem densities and structure were beginning to emerge. Significant differences were observed in the nutrient concentration of the three species that comprised the most aboveground biomass, and between species categorized as secondary high-light species and primary, low-light species. Species whose abundance was negatively correlated with the mature forest dominant also had distinct nutrient concentrations. When total aboveground nutrient pools were compared over time, differences in leaf nutrients among species were hidden by similarities in wood nutrient concentrations and the biomass dominance of a few species. The observed successional trajectory indicates that changes in species composition contributed to fast recovery of aboveground biomass and nutrient pools, while the influence of geomorphic setting on species composition occurs at time scales >15 yr of succession. 相似文献

The current diagnostic work-up and monitoring of pulmonary infections may be perceived as invasive, is time consuming and expensive. In this explorative study, we investigated whether or not a non-invasive exhaled breath analysis using an electronic nose would discriminate between cystic fibrosis (CF) and primary ciliary dyskinesia (PCD) with or without various well characterized chronic pulmonary infections. We recruited 64 patients with CF and 21 with PCD based on known chronic infection status. 21 healthy volunteers served as controls. An electronic nose was employed to analyze exhaled breath samples. Principal component reduction and discriminant analysis were used to construct internally cross-validated receiver operator characteristic (ROC) curves. Breath profiles of CF and PCD patients differed significantly from healthy controls p = 0.001 and p = 0.005, respectively. Profiles of CF patients having a chronic P. aeruginosa infection differed significantly from to non-chronically infected CF patients p = 0.044. We confirmed the previously established discriminative power of exhaled breath analysis in separation between healthy subjects and patients with CF or PCD. Furthermore, this method significantly discriminates CF patients suffering from a chronic pulmonary P. aeruginosa (PA) infection from CF patients without a chronic pulmonary infection. Further studies are needed for verification and to investigate the role of electronic nose technology in the very early diagnostic workup of pulmonary infections before the establishment of a chronic infection. 相似文献

Participants were gloved and asked to present the contents of six pouches of three different sizes to a simulated sterile field. The exterior of pouches and gloves of participants were coated with a simulated contaminant prior to each opening trial. After presentation to the simulated sterile field, the presence of the contaminant on package contents was recorded as indicative of contact with non-sterile surfaces and analyzed in a binary fashion using a generalized linear mixed model.Results
Recruited subjects were 26–64 years of age (81 females, 16 males), with 2.5–44 years of professional experience. Results indicated a significant main effect of pouch size on contact rate of package contents (P = 0.0108), whereby larger pouches induced greater rates of contact than smaller pouches (estimates±SEM: 14.7±2.9% vs. 6.0±1.7%, respectively).Discussion and Conclusion
This study utilized novel methodologies which simulate contamination in aseptic presentation. Results of this work indicate that increased contamination rates are associated with larger pouches when compared to smaller pouches. The results add to a growing body of research which investigate packaging''s role in serving as a pathway for product contamination during aseptic presentation. A new labeling technique attaching a fluorescent pteridine derivative (3, 5) via a linker onto the 3'-OH group of 5'-O-dimethoxytritylthymidine (7) was developed to lead to the conjugates 8 and 11.
