In this study we examined changes in colonic mucosal permeability induced by dextran sulfate sodium (DSS) during the acute phase of mouse colitis. To induce colitis, the mice were given drinking water containing 5% (w/v) DSS (MW = 40,000) ad libitum. Colonic mucosal permeability was evaluated by the permeation of Evans blue (EB) from the lumen into the wall of the colon on 1, 2, 3 and 7 days postadministration of DSS. Mucosal changes were also histologically examined daily for 7 days postadministration. The permeation of EB increased significantly by days 3 and 7 postadministration. Histological analysis showed that crypt loss was the initial change, with no inflammatory process and the surface mucosal epithelial cells remained morphologically intact. These histological changes developed on 2 to 3 days postadministration. Erosion was first recognized at 5 days postadministration. These findings indicated that the increase in colonic mucosal permeability may have occurred in 3 days postadministration, and the increase in mucosal permeability occurred before the appearance of the inflammatory process. This suggests that an increase in colonic mucosal permeability, leading to the destruction of mucosal barrier function, may play an important role in the induction of DSS-induced murine colitis. 相似文献

Terrestrial hermit crabs in the family Coenobitidae (genera Coenobita and Birgus) must migrate onto land after completing a pelagic larval stage in the ocean. Better knowledge of emigration behavior would assist in the conservation and management of coenobitid populations by helping identify and protect the habitats they need to complete their life cycles. We cultured laboratory‐born individuals of five coenobitid species (Coenobita cavipes, C. purpureus, C. rugosus, C. violascens, and Birgus latro) from megalopae to early juveniles (first, second, and/or third crabs) in vessels containing seawater and a hard substrate, and analyzed their behavior and molting in conjunction with our published data for C. brevimanus. Our results confirm that the coenobitids migrated from sea to land at the megalopal stage. Megalopae and early juveniles tended to select shells based on their body size. Inland‐dwelling coenobitids, such as C. brevimanus, C. cavipes, and B. latro, had a longer duration from landing to first molt and had a prolonged first crab intermolt period compared with those of the beach‐dwelling coenobitids C. purpureus, C. rugosus, and C. violascens, probably because of the adaptive traits for migrating to inland habitats. Little burrowing behavior was observed by megalopae of B. latro, but they had a strong tendency to be cryptic under shelters. Additionally, megalopae and early juveniles of Coenobita spp. created and utilized burrows somewhat differently. Our results suggest that coenobitids require specific microhabitats for completing their early life stages in the wild. In previous clinical studies, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection in cancer patients has a high risk of aggravation and mortality than in healthy infected individuals. Inoculation with coronavirus disease 2019 (COVID-19) vaccine reduces the risk of SARS-CoV-2 infection and COVID-19 severity. However, vaccination-induced anti-SARS-CoV-2 antibody production is said to be lower in cancer patients than in healthy individuals. In addition, the rationale for why the condition of patients with cancer worsens with COVID-19 is not well understood. Therefore, we examined the infection status of SARS-CoV-2 in the primary tumor and micrometastasis tissues of the patient with cancer and COVID-19. In this study, the expression of angiotensin-converting enzyme 2 (ACE2) was observed, and SARS-CoV-2 particles was detected in ovarian tissue cells in contact with the micrometastatic niche of the patient with high-grade serous ovarian cancer. We believe that the severity of COVID-19 in patients with cancer can be attributed to these pathological features. Therefore, the pathological findings of patients with advanced and recurrent ovarian cancer infected with SARS-CoV-2 may help decrease COVID-19 severity in patients with other cancer types. 相似文献

Phosphate (Pi) plays important roles in plant development and architecture. With the goal of identifying genomic regions that influence tolerance to Pi deficiency (TPDE) in hybrid rice (Oryza sativa L.), quantitative trait loci (QTL) were mapped using recombinant inbred lines (RILs) that were derived from a cross between tolerant ‘XieqingzaoB’ (XB) and susceptible ‘Zhonghui9308’. Six TPDE-related traits, including the root length, root dry weight, tillers number, shoot dry weight, total plant dry weight and root-to-shoot ratio, were evaluated for QTL analysis during both the tillering and heading stages. A correlation analysis showed that most of the traits were correlated with each other. Twenty-one additive QTL were detected and jointly explained between 10–49% of the trait variance, tending to cluster on chromosomes 4, 6, 10 and 11. Three QTL, qTPDE4 XB , qTPDE10 XB and qTPDE11.3 XB , were validated by the phenotypic evaluation using near isogenic lines (NILs, BC4F3) during the seedling stage. qTPDE4 XB showed the most stable tolerance against Pi deficiency. In future large-scale molecular dynamics (MD) simulations that will use parallel computing, the isotropic periodic sum (IPS) method is expected to effectively reduce the cost of interaction calculations while maintaining adequate accuracy. To assess the accuracy of this method in estimating low-charge-density polymer systems, we performed atomistic MD simulations of the bulk state of liquid crystal systems based on 4-pentyl-4′-cyanobiphenyl (5CB). In conditions of 270 K ≤ T ≤ 320 K and a normal pressure, the temperature dependence of the density, potential energy and order parameter was estimated using the IPS and Ewald sum method. The results of the IPS method and Ewald sum were consistent within the range of error. In conditions close to the phase transition point, however, the averaged values of potential energy and order parameter had a small difference. We concluded that the fundamental physical properties for the bulk state of 5CB systems are determined reasonably by using the IPS method, at least in conditions that are not close to the phase transition point. 相似文献

Accumulating evidence indicates that epigenetic aberrations have a role in the pathogenesis of rheumatoid arthritis (RA). However, reports on histone modifications are as yet quite limited in RA. Interleukin (IL)-6 is an inflammatory cytokine which is known to be involved in the pathogenesis of RA. Here we report the role of histone modifications in elevated IL-6 production in RA synovial fibroblasts (SFs). The level of histone H3 acetylation (H3ac) in the IL-6 promoter was significantly higher in RASFs than osteoarthritis (OA) SFs. This suggests that chromatin structure is in an open or loose state in the IL-6 promoter in RASFs. Furthermore, curcumin, a histone acetyltransferase (HAT) inhibitor, significantly reduced the level of H3ac in the IL-6 promoter, as well as IL-6 mRNA expression and IL-6 protein secretion by RASFs. Taken together, it is suggested that hyperacetylation of histone H3 in the IL-6 promoter induces the increase in IL-6 production by RASFs and thereby participates in the pathogenesis of RA. 相似文献
