Microfluidics is used increasingly for engineering and biomedical applications due to recent advances in microfabrication technologies. Visualization of bubbles, tracer particles, and cells in a microfluidic device is important for designing a device and analyzing results. However, with conventional methods, it is difficult to observe the channel geometry and such particles simultaneously. To overcome this limitation, we developed a Darkfield Internal Reflection Illumination (DIRI) system that improved the drawbacks of a conventional darkfield illuminator. This study was performed to investigate its utility in the field of microfluidics. The results showed that the developed system could clearly visualize both microbubbles and the channel wall by utilizing brightfield and DIRI illumination simultaneously. The methodology is useful not only for static phenomena, such as clogging, but also for dynamic phenomena, such as the detection of bubbles flowing in a channel. The system was also applied to simultaneous fluorescence and DIRI imaging. Fluorescent tracer beads and channel walls were observed clearly, which may be an advantage for future microparticle image velocimetry (μPIV) analysis, especially near a wall. Two types of cell stained with different colors, and the channel wall, can be recognized using the combined confocal and DIRI system. Whole-slide imaging was also conducted successfully using this system. The tiling function significantly expands the observing area of microfluidics. The developed system will be useful for a wide variety of engineering and biomedical applications for the growing field of microfluidics.  相似文献

Purpose

To investigate the length variation of the posterior auricular artery and propose a novel classification of the posterior auricular artery based on angiographical appearance.

Patients and Methods

A series of 234 consecutive patients who had undergone conventional cerebral angiography was analyzed. The posterior auricular artery was examined on the lateral projection of the external carotid or common carotid arteriography. The posterior auricular artery was classified into four groups by length, using the external auditory canal and the top of the helix as radiographical landmarks. Our proposed classification is as follows: Type A, posterior auricular artery terminates between its origin and the center of the external auditory canal; Type B, posterior auricular artery terminates between the center of the external auditory canal and the top of the helix; Type C, posterior auricular artery terminates between the top of the helix and the vertex; and Type D, posterior auricular artery reaches up to the vertex.

Results

A total of 424 (right, 214; left, 210) posterior auricular arteries were analyzed in 111 men and 123 women aged 11 to 91 years (mean, 61.0 years) examined for aneurysms in 78 cases, occlusive vascular diseases in 56, intracranial hemorrhages in 41, tumors in 35, and others in 24. Types A, B, C, and D were found in 15.1%, 34.9%, 48.8%, and 1.2% of the patients, respectively.

Conclusion

A novel classification of the posterior auricular artery identifies four types based on its length on cerebral angiography.  相似文献

Chromosome segment substitution lines (CSSLs) are a powerful alternative for locating quantitative trait loci (QTL), analyzing gene interactions, and providing starting materials for map-based cloning projects. We report the development and characterization of a CSSL library of a U.S. weedy rice accession ‘PSRR-1’ with genome-wide coverage in an adapted rice cultivar ‘Bengal’ background. The majority of the CSSLs carried a single defined weedy rice segment with an average introgression segment of 2.8 % of the donor genome. QTL mapping results for several agronomic and domestication traits from the CSSL population were compared with those obtained from two recombinant inbred line (RIL) populations involving the same weedy rice accession. There was congruence of major effect QTLs between both types of populations, but new and additional QTLs were detected in the CSSL population. Although, three major effect QTLs for plant height were detected on chromosomes 1, 4, and 8 in the CSSL population, the latter two escaped detection in both RIL populations. Since this was observed for many traits, epistasis may play a major role for the phenotypic variation observed in weedy rice. High levels of shattering and seed dormancy in weedy rice might result from an accumulation of many small effect QTLs. Several CSSLs with desirable agronomic traits (e.g. longer panicles, longer grains, and higher seed weight) identified in this study could be useful for rice breeding. Since weedy rice is a reservoir of genes for many weedy and agronomic attributes, the CSSL library will serve as a valuable resource to discover latent genetic diversity for improving crop productivity and understanding the plant domestication process through cloning and characterization of the underlying genes.  相似文献

β-D-galactofuranose (Galf) is a component of polysaccharides and glycoconjugates and its transferase has been well analyzed. However, no β-D-galactofuranosidase (Galf-ase) gene has been identified in any organism. To search for a Galf-ase gene we screened soil samples and discovered a strain, identified as a Streptomyces species by the 16S ribosomal RNA gene analysis, that exhibits Galf-ase activity for 4-nitrophenyl β-D-galactofuranoside (pNP-β-D-Galf) in culture supernatants. By draft genome sequencing of the strain, named JHA19, we found four candidate genes encoding Galf-ases. Using recombinant proteins expressed in Escherichia coli, we found that three out of four candidates displayed the activity of not only Galf-ase but also α-L-arabinofuranosidase (Araf-ase), whereas the other one showed only the Galf-ase activity. This novel Galf-specific hydrolase is encoded by ORF1110 and has an optimum pH of 5.5 and a Km of 4.4 mM for the substrate pNP-β-D-Galf. In addition, this enzyme was able to release galactose residue from galactomannan prepared from the filamentous fungus Aspergillus fumigatus, suggesting that natural polysaccharides could be also substrates. By the BLAST search using the amino acid sequence of ORF1110 Galf-ase, we found that there are homolog genes in both prokaryotes and eukaryotes, indicating that Galf-specific Galf-ases widely exist in microorganisms.  相似文献

The Wnt-induced planar cell polarity (PCP) signaling pathway is essential for polarized cell migration and morphogenesis. Dishevelled (Dvl) and its binding protein Daam1 mediate RhoA activation in this pathway. WGEF, a member of the Rho-guanine nucleotide exchange factor (Rho-GEF) family, was shown to play a role in Wnt-induced RhoA activation in Xenopus embryos. However, it has remained unknown which member(s) of a Rho-GEF family are involved in Wnt/Dvl-induced RhoA activation in mammalian cells. Here we identified p114-RhoGEF and Lfc (also called GEF-H1) as the Rho-GEFs responsible for Wnt-3a–induced RhoA activation in N1E-115 mouse neuroblastoma cells. We screened for Rho-GEF–silencing short-hairpin RNAs (shRNAs) that are capable of suppressing Dvl-induced neurite retraction in N1E-115 cells and found that p114-RhoGEF and Lfc shRNAs, but not WGEF shRNA, suppressed Dvl- and Wnt-3a–induced neurite retraction. p114-RhoGEF and Lfc shRNAs also inhibited Dvl- and Wnt-3a–induced RhoA activation, and p114-RhoGEF and Lfc proteins were capable of binding to Dvl and Daam1. Additionally, the Dvl-binding domains of p114-RhoGEF and Lfc inhibited Dvl-induced neurite retraction. Our results suggest that p114-RhoGEF and Lfc are critically involved in Wnt-3a– and Dvl-induced RhoA activation and neurite retraction in N1E-115 cells.  相似文献

Alzheimer's disease (AD) is characterized by progressive cognitive impairment and the formation of senile plaques. Silymarin, an extract of milk thistle, has long been used as a medicinal herb for liver diseases. Here we report marked suppression of amyloid β-protein (Aβ) fibril formation and neurotoxicity in PC12 cells after silymarin treatment in vitro. In vivo studies had indicated a significant reduction in brain Aβ deposition and improvement in behavioral abnormalities in amyloid precursor protein (APP) transgenic mice that had been preventively treated with a powdered diet containing 0.1% silymarin for 6 months. The silymarin-treated APP mice also showed less anxiety than the vehicle-treated APP mice. These behavioral changes were associated with a decline in Aβ oligomer production induced by silymarin intake. These results suggest that silymarin is a promising agent for the prevention of AD.  相似文献

We characterized peptidyl hydroxyproline (Hyp) O-galactosyltransferase (HGT), which is the initial enzyme in the arabinogalactan biosynthetic pathway. An in vitro assay of HGT activity was established using chemically synthesized fluorescent peptides as acceptor substrates and extracts from Arabidopsis (Arabidopsis thaliana) T87 cells as a source of crude enzyme. The galactose residue transferred to the peptide could be detected by high-performance liquid chromatography and matrix-assisted laser desorption-ionization time-of-flight mass spectrometry analyses. HGT required a divalent cation of manganese for maximal activity and consumed UDP-d-galactose as a sugar donor. HGT exhibited an optimal pH range of pH 7.0 to 8.0 and an optimal temperature of 35°C. The favorable substrates for the activity seemed to be peptides containing two alternating imino acid residues including at least one acceptor Hyp residue, although a peptide with single Hyp residue without any other imino acids also functioned as a substrate. The results of sucrose density gradient centrifugation revealed that the cellular localization of HGT activity is identical to those of endoplasmic reticulum markers such as Sec61 and Bip, indicating that HGT is predominantly localized to the endoplasmic reticulum. To our knowledge, this is the first characterization of HGT, and the data provide evidence that arabinogalactan biosynthesis occurs in the protein transport pathway.O-glycosylation is the addition of a sugar to hydroxy amino acids such as Thr, Ser, Hyp, Hyl, or Tyr (Lehle et al., 2006). This type of protein modification occurs in many organisms to modify a large variety of proteins. Several types of sugars can be linked to proteins via O-glycosylation, including Man, N-acetylgalactosamine, Glc, Xyl, N-acetylglucosamine, Fuc, Gal, and arabinofuranose (Araf). In addition, elongation of the added sugar residues yields a large variety of oligo- and polysaccharide extensions on the substrate proteins. These modifications are known to play important roles in various phenomena, including pathways required to maintain biological systems and basic cellular functions.Structural analysis of oligo- and polysaccharides in plant cell walls has revealed the presence of three types of O-linked structures, Gal-O-Hyp, Araf-O-Hyp, and Gal-O-Ser (Kieliszewski and Shpak, 2001; Seifert and Roberts, 2007). A part of these three structures has been found on proteins in the super family that includes arabinogalactan protein (AGP) and extensin, which are localized to the cell surface. AGPs contain O-linked arabinogalactan oligo- or polysaccharides attached to Hyp residues (Gal-O-Hyp). It is known that arabinogalactan polysaccharides mainly consist of β-1,3 linkages of Gal polymers (Seifert and Roberts, 2007). Extensin contains short arabino-oligosaccharide chains attached to Hyp residues (Araf-O-Hyp) and single Gal residues linked to Ser residues (Gal-O-Ser). It has been suggested that these O-linked structures play an important role in many stages of growth and development in plants, including signaling, embryogenesis, and programmed cell death (Knox, 2006; Seifert and Roberts, 2007). However, our understanding of the biosynthesis of these O-linked structures is limited at present.Shpak et al. described a novel strategy to elucidate O-glycosylation of AGPs via introduction of synthetic genes encoding a protein substrate of glycosyltransferases into plant cells (Shpak et al., 1999; Estevez et al., 2006). This strategy provided good evidence for the substrate specificities of Hyp O-galactosyltransferase (HGT). Hyp galactosylation occurs on clustered noncontiguous Hyp residues such as Xaa-Hyp-Xaa-Hyp repeats of AGPs (where Xaa is any amino acid except Hyp; Tan et al., 2003). However, the arabinogalactosylation site is not limited to clustered noncontiguous Hyp residues, as isolated Hyp residues with appropriate surrounding sequences can be modified with arabinogalactan (Matsuoka et al., 1995; Shimizu et al., 2005). Therefore, the mechanism of glycosylation to Hyp residues seems complex in plants, while we have little information about the glycosyltransferase(s) involved in arabinogalactan biosynthesis. To examine the enzymatic properties and to identify genes involved in arabinogalactan biosynthesis, we first attempted to establish an in vitro assay for HGT activity, which catalyzes the initial step in arabinogalactan biosynthesis in plants.Here, we report a novel assay for HGT activity based on the use of endoplasmic reticulum (ER)-enriched cell lysates extracted from Arabidopsis (Arabidopsis thaliana) T87 cells as a source of the enzyme and chemically synthesized fluorescent peptides as enzyme substrates. The method enabled us to characterize the enzymatic properties of HGT and to determine the localization of HGT in Arabidopsis cells. Properties of the enzyme and the usefulness of our assay for various studies are discussed.  相似文献

The transesterification reactions of triolein with ethanol using various ion-exchange resin catalysts were conducted to produce ethyl oleate as a biodiesel. The anion-exchange resins exhibited much higher catalytic activities than the cation-exchange resin. The anion-exchange resin with a lower cross-linking density and a smaller particle size gave a high reaction rate as well as a high conversion. By combining the three-step regeneration method, the resin could be repeatedly used for the batch transesterification without any loss in the catalytic activity. A continuous transesterification reaction was carried out using an expanded bed reactor packed with the most active resin. The reactor system permitted the continuous production of ethyl oleate with a high conversion.  相似文献
