As a quantitative approach to the life histories of fishes, the present paper attempted to predict a relation among reproduction, growth and mortality numerically with a technique of control theory, the discrete maximum principle. A method for predicting the relation was derived on the postulate that natural selection maximized the net reproductive rate subject to a few constraints. The derived method was applied to Atlantic cod and Atlantic herring populations in the Southern Gulf of St. Lawrence as numerical examples. The examples demonstrated that the theoretical reproductive effort and body weight were well consistent with the observed ones every age but the theoretical survival rates were slightly different from the observed ones. For the reasons mentioned below, however, it should be interpreted that the examples rather support the adopted postulate to a certain degree. First, in general, it is very difficult to obtain good estimates of the rates with traditional methods. Second, intense fishing pressure possibly changes the life history parameters to some extent in fish populations. Moreover, the examples also suggested that, to examine the postulate in further detail, similar analyses had to be made with the data of many fish populations on which intense fishing pressure had not been exerted.  相似文献

Japanese encephalitis virus (JEV), which is transmitted by mosquitoes, infects many animal species and causes serious acute encephalitis in humans and horses. In this study, a serosurvey of JEV in Japanese macaques (Macaca fuscata) reared in Aichi Prefecture was conducted using purified JEV as an antigen for ELISA. The results revealed that 146 of 332 monkeys (44 %) were seropositive for JEV. In addition, 35 of 131 monkeys (27 %) born in the facility were seropositive, and the annual infection rate in the facility was estimated as 13 %. Our results provide evidence of the frequent exposure of many Japanese macaques to JEV, suggesting that there is a risk of JEV transmission to humans by mosquitoes.  相似文献

We compared volatiles from lima bean leaves (Phaseolus lunatus) infested by either beet armyworm (Spodoptera exigua), common armyworm [Mythimna (Pseudaletia) separata], or two-spotted spider mite (Tetranychus urticae). We also analyzed volatiles from the leaves treated with jasmonic acid (JA) and/or methyl salicylate (MeSA). The volatiles induced by aqueous JA treatment were qualitatively and quantitatively similar to those induced by S. exigua or M. separata damage. Furthermore, both S. exigua and aqueous JA treatment induced the expression of the same basic PR genes. In contrast, gaseous MeSA treatment, and aqueous JA treatment followed by gaseous MeSA treatment, induced volatiles that was qualitatively and quantitatively more similar to the T. urticae-induced volatiles than those induced by aqueous JA treatment. In addition, T. urticae damage resulted in the expression of the acidic and basic PR genes that were induced by gaseous MeSA treatment and by aqueous JA treatment, respectively. Based on these data, we suggest that in lima bean leaves, the JA-related signaling pathway is involved in the production of caterpillar-induced volatiles, while both the SA-related signaling pathway and the JA-related signaling pathway are involved in the production of T. urticae-induced volatiles.  相似文献

It has been speculated that a soluble form of G-CSFR might be physiologically present in humans, since G-CSFR mRNA that lacks a transmembrane domain has been identified from a human myelomonocytic cell line. Here, we demonstrate human soluble G-CSFR (sG-CSFR) of two different molecular sizes (80 and 85 kDa) on an immunoblot analysis using Abs generated against the amino-terminal, extracellular domain of the full-length G-CSFR. Both isoforms of sG-CSFR were able to bind recombinant human G-CSF (rhG-CSF). RT-PCR analysis with primers targeted outside of the transmenbrane region revealed that membrane-anchored G-CSFR is expressed at all maturation stages of purified myeloid cells, including CD34+CD13+ cells (blasts), CD11b-CD15+ cells (promyelocytes or myelocytes), CD11b+CD15+ cells (metamyelocytes and mature neutrophils), and CD14+ cells (monocytes). On the other hand, sG-CSFR mRNA was detectable in CD11b-CD15+, CD11b+CD15+, and CD14+ cells, but not in the CD34+CD13+ blast population. The serum concentration of both isoforms of sG-CSFR appeared to be correlated with the numbers of neutrophils/monocytes before and after rhG-CSF treatment in normal individuals. Thus, two isoforms of sG-CSFR are physiologically secreted from relatively mature myeloid cells and might play an important role in myelopoiesis through their binding to serum G-CSF.  相似文献

To determine the role of endothelium in hypoxic pulmonary vasoconstriction (HPV), we measured vasomotor responses to hypoxia in isolated seventh-generation porcine pulmonary arteries < 300 microm in diameter with (E+) and without endothelium. In E+ pulmonary arteries, hypoxia decreased the vascular intraluminal diameter measured at a constant transmural pressure. These constrictions were complete in 30-40 min; maximum at PO(2) of 2 mm Hg; half-maximal at PO(2) of 40 mm Hg; blocked by exposure to Ca(2+)-free conditions, nifedipine, or ryanodine; and absent in E+ bronchial arteries of similar size. Hypoxic constrictions were unaltered by indomethacin, enhanced by indomethacin plus N(G)-nitro-L-arginine methyl ester, abolished by BQ-123 or endothelial denudation, and restored in endothelium-denuded pulmonary arteries pretreated with 10(-10) M endothelin-1 (ET-1). Given previous demonstrations that hypoxia caused contractions in isolated pulmonary arterial myocytes and that ET-1 receptor antagonists inhibited HPV in intact animals, our results suggest that full in vivo expression of HPV requires basal release of ET-1 from the endothelium to facilitate mechanisms of hypoxic reactivity in pulmonary arterial smooth muscle.  相似文献

Neural recognition molecule NB-2/contactin 5 is expressed transiently during the first postnatal week in glutamatergic neurons of the central auditory system. Here, we investigated the effect of NB-2 deficiency on the auditory brainstem in mouse. While almost all principal neurons are wrapped with the calyces of Held in the medial nucleus of the trapezoid body (MNTB) in wild type, 8% of principal neurons in NB-2 knockout (KO) mice lack the calyces of Held at postnatal day (P) 6. At P10 and P15, apoptotic principal neurons were detected in NB-2 KO mice, but not in wild type. Apoptotic cells were also increased in the ventral cochlear nucleus (VCN) of NB-2 KO mice, which contains bushy neurons projecting to the MNTB and the lateral superior olive (LSO). At the age of 1 month, the number of principal neurons in the MNTB and of glutamatergic synapses in the LSO was reduced in NB-2 KO mice. Finally, interpeak latencies for auditory brainstem response waves II-III and III-IV were significantly increased in NB-2 KO mice. Together, these findings suggest that NB-2 deficiency causes a deficit in synapse formation and then induces apoptosis in MNTB and VCN neurons, affecting auditory brainstem function.  相似文献

To determine whether reactive oxygen species (ROS) play an essential role in hypoxic pulmonary vasoconstriction (HPV) and the cellular locus of ROS production and action during HPV, we measured internal diameter (ID) at constant transmural pressure, lucigenin-derived chemiluminescence (LDCL), and electron paramagnetic resonance (EPR) spin adduct spectra in small distal porcine pulmonary arteries, and dichlorofluorescein (DCF) fluorescence in myocytes isolated from these arteries. Hypoxia (4% O2) decreased ID, increased DCF fluorescence, tended to increase LDCL, and in some preparations produced EPR spectra consistent with hydroxyl and alkyl radicals. Superoxide dismutase (SOD, 150 U/ml) or SOD + catalase (CAT, 200 U/ml) did not alter ID during normoxia but reduced or abolished the constriction induced by hypoxia. SOD also blocked HPV in endothelium-denuded arteries after restoration of the response by exposure to 10-10 M endothelin-1. Confocal fluorescence microscopy demonstrated that labeled SOD and CAT entered pulmonary arterial myocytes. SOD, SOD + CAT, and CAT blocked the increase in DCF fluorescence induced by hypoxia, but SOD + CAT and CAT also caused a stable increase in fluorescence during normoxia, suggesting that CAT diminished efflux of DCF from cells or oxidized the dye directly. We conclude that HPV required increased concentrations of ROS produced by and acting on pulmonary arterial smooth muscle rather than endothelium.  相似文献

Meimuna opalifera males emit extremely complex calling songs, with the main section comprising two parts. To investigate whether these two parts of the calling song have different meanings for the receivers, we conducted playback experiments. We counted and compared the number of response calls when different types of sound stimuli were presented under laboratory conditions. Meimuna opalifera males emitted significantly greater response calls during the playback of stimuli containing the former part of the songs than those containing the latter part only. This suggests that M. opalifera males tend to respond by detecting characteristics in the former part of the songs. Because the peak frequencies were similar between the former and the latter parts, the temporal patterns of the songs might be important in short-range communication between males of M. opalifera.  相似文献

Vitiligo is a common depigmentation disorder characterized by the selective loss of melanocytes. In our daily clinic experience, we noticed that the skin tightness of hypopigmented lesions would be more evident in comparison to that of uninvolved perilesional skin in vitiligo patients. Therefore, we hypothesized that collagen homeostasis might be maintained in vitiligo lesions, irrespective of the substantial excessive oxidative stress that occurs in association with the disease. We found that the expression levels of collagen-related genes and anti-oxidative enzymes were upregulated in vitiligo-derived fibroblasts. Abundant collagenous fibers were observed in the papillary dermis of vitiligo lesions in comparison to uninvolved perilesional skin by electron microscopy. The production of matrix metalloproteinases that degraded collagen fibers was suppressed. The deposition of acrolein adduct protein, which is a product of oxidative stress, was significantly reduced in vitiligo dermis and fibroblasts. As part of the mechanism, we found upregulation of the NRF2 signaling pathway activity, which is an important defense system against oxidative stress. Taken together, we demonstrated that the anti-oxidative action and collagen production were upregulated and that the collagen degeneration was attenuated in vitiligo dermis. These new findings may provide important clues for the maintenance of antioxidant ability in vitiligo lesions.  相似文献

Calmodulin in Schizosaccharomyces pombe is encoded by the cam1+ gene, which is indispensable for both vegetative growth and sporulation. Here, we report how Cam1 functions in spore formation. We found that Cam1 preferentially localized to the spindle pole body (SPB) during meiosis and sporulation. Formation of the forespore membrane, a precursor of the plasma membrane in spores, was blocked in a missense cam1 mutant, which was viable but unable to sporulate. Three SPB proteins necessary for the onset of forespore membrane formation, Spo2, Spo13, and Spo15, were unable to localize to the SPB in the cam1 mutant although five core SPB components that were tested were present. Recruitment of Spo2 and Spo13 is known to require the presence of Spo15 in the SPB. Notably, Spo15 was unstable in the cam1 mutant, and as a result, SPB localization of Spo2 and Spo13 was lost. Overexpression of Spo15 partially alleviated the sporulation defect in the cam1 mutant. These results indicate that calmodulin plays an essential role in forespore membrane formation by stably maintaining Spo15, and thus Spo2 and Spo13, at the SPB in meiotic cells.Calmodulin is a calcium-binding protein that is ubiquitously distributed and highly conserved among eukaryotes. It contains four EF-hand Ca2+-binding sites, which are required for function. Calmodulin controls a variety of cellular processes mostly related to calcium signaling. When bound to calcium, calmodulin undergoes a characteristic conformational change to an active configuration. Activated calmodulin then binds effector proteins and transmits the signal to downstream regulators.Yeast is a genetically tractable model organism suitable for studying the biological function of calmodulin, using conditional-lethal calmodulin mutants (4). In the budding yeast Saccharomyces cerevisiae, calmodulin is encoded by the CMD1 gene (5). Cmd1p is implicated in a wide variety of cellular processes, including initiation of budding and mitotic spindle formation (24). The fission yeast Schizosaccharomyces pombe has a typical calmodulin encoded by the cam1+ gene, which plays an indispensable role in cell proliferation, dependent on its Ca2+-binding activity (18, 19, 30). A green fluorescent protein (GFP)-Cam1 fusion protein localizes to sites of polarized cell growth and to the spindle pole body (SPB) in vegetative cells (19). Thus, an essential role of Cam1 might be its regulatory function in chromosome segregation (19). The role of calmodulin in the sexual cycle has been documented to a lesser extent in previous studies. A missense mutant, cam1-117, in which the Arg117 codon is changed to a Phe codon, exhibits reduced sporulation efficacy (29), suggesting that calmodulin plays a role in sporulation in fission yeast.Spore formation in fission yeast initiates with assembly of the forespore membrane (FSM), composed of double-unit membranes within the cytoplasm of a diploid zygote cell (10, 27, 28, 34). The FSM expands to encapsulate each haploid nucleus generated by meiosis and then forms a nucleated prespore. The inner bilayer of the FSM subsequently becomes the plasma membrane of the newborn spores. During meiosis II, the SPB undergoes morphological alteration from a compact single plaque to a multilayered expanded structure (10). Such modification of the SPB is a prerequisite for FSM assembly, which occurs close to the outermost layer of the modified SPB (9, 10, 20, 21).Three SPB component proteins, Spo2, Spo13, and Spo15, have been identified as essential for SPB modification and formation of the FSM (11, 23). Spo15, a large coiled-coil protein, is associated with the SPB throughout the life cycle and is indispensable for recruitment of Spo2 and Spo13 to the cytoplasmic surface of the meiotic SPB. The latter two proteins are produced only during meiosis (23). These observations imply that the SPB serves as a platform for assembly of the FSM. Cam1 has been reported to localize to the SPB during vegetative growth (19), raising the intriguing possibility that fission yeast calmodulin is involved in sporulation through proper construction of a modified meiotic SPB. To test this possibility, we report herein a detailed analysis of Cam1 localization during meiosis and the consequence of a missense mutation of cam1 on SPB modification and FSM formation.  相似文献
