A heat-stable factor has been extracted from the microsomal membranes of rat brain that inhibits the activities of rat brain ceramide galactosyl- and glucosyltransferases. It is nondialyzable, susceptible to proteolytic enzymes but resistant to DNase and RNase, and has no effect on lysosomal hydrolytic enzymes. The factor could also be prepared from microsomes of bovine, mouse, and rabbit brains and in much lower concentrations, from systemic organs of rats. This inhibitor might play a role in the regulation of ceramide glycosyltransferases in vivo.  相似文献

To evaluate the role of pyrophosphate-dependent phosphofructokinase (PFP. EC 2.7.1.90) in the sugar metabolism of pollen. its occurrence and properties were studied in pollen grains of several plants including camellia ( Camellia japonica L.). In all pollen samples, PFP was strongly activated by fructose-2,6-bisphosphate (F2,6BP), and the activity of F2,6BP-activated PFP was higher than that of phosphofructokinase (PFK. EC 2.7.1.11). PFP partially purified from camellia pollen required Mg2+ for activity with an optimum at 1 m M . and was almost unaflected by a variety of metabolites at 1 m M . Its molecular mass was around 220 kDa, and apparent Km values for F6P, PPi. F1, 6BP and Pi were 294, 4, 20 and 580 u M , respectively. The levels of F2.6BP. PPi and F6P in camellia pollen were sufficent to support the forward reaction by PFP, and PFP, was 20- to 40-fold more active than PFK during pollen growth. These results suggest that pollen PFP plays a role in glycolysis but not gluconeogenesis. and the possible relevance of this to pollen tube growth is discussed.  相似文献

The heat denaturation and renaturation curves of rat liver and ascites hepatoma (AH 108A) chromatins were measured. In these renaturation curves, there are small sigmoidal regions. These sigmoidal regions remained in redenaturation curves and were largely stable to DNAase I digestion. When the chromatins were treated stepwise with NaClO4 and lysine-rich histones were removed, the sigmoidal regions in the renaturation curves disappeared. These results suggested that the sigmoidal regions reflected the interaction of DNA and lysine-rich histones.  相似文献

The Limulus test has been considered specific for the presence of bacterial endotoxins. To synthesize a simple model of endotoxin, palmitoyldextran phosphate was prepared by modification of dextran by palmitoylation and phosphorylation. The present studies indicated that a variety of polysaccharide derivatives, such as palmitoyldextran phosphate, palmitoyldextran, and dextran phosphate, give a positive Limulus test and show pyrogenic activity, except for low molecular dextran derivatives. On the other hand, polysaccharides, such as dextran, starch (soluble), chitosan, xylan, and lentinan, were negative in these assays. The gelation reaction of Limulus lysate by modified dextran derivatives may depend on the molecular weight or modification of polysaccharides by palmitoylation and/or phosphorylation to a great extent.  相似文献

The alkylating agent N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) enhanced the levels of plasminogen activator (PA) activity in fibroblast cells derived from the skin of patients with tuberous sclerosis. The enhanced enzyme levels were not correlated with those of cloning efficiency nor those of DNA synthesis after MNNG treatment. Enzyme enhancement was also observed in fibroblasts of ataxia telangiectasia and in human neoplastic glia cells, but not in fibroblasts of normal children. The PA induction test may be sufficiently sensitive for the detection of the cellular defects of tuberous sclerosis.  相似文献
