Human metapneumovirus (hMPV) is one of the etiological agents of acute respiratory tract infections. From June 2005 to May 2006, we collected 185 clinical specimens from children in Osaka City, Japan, and detected 41 hMPV RNA. Of the 41 specimens, four (9.8%) also contained other viruses (3 with adenovirus [AdV] and 1 with respiratory syncytial virus [RSV]). The clinical symptoms of patients coinfected with AdV were indistinct from those of patients mono-infected with hMPV. The symptoms of the one patient co-infected with RSV were clinically severe. Further research is needed to clarify the effect of hMPV on other respiratory viruses or vice versa. 相似文献

Previous analysis of PCR products derived from total RNA from the abdominal gland of the male newt, Cynops pyrrhogaster, inhabiting the Nara area of Japan led to the identification of a gene encoding [Val(8)]sodefrin, as well as the female-attracting peptide pheromone, sodefrin. In this study, purification of this sodefrin variant from the abdominal glands of male newts from the Nara area was accomplished using gel-filtration chromatography and reversed-phase HPLC. Amino acid sequence analysis and mass spectrometry confirmed that the final product was [Val(8)]sodefrin. A full-length cDNA encoding the biosynthetic precursor of [Val(8)]sodefrin was cloned and characterized. The deduced amino acid sequence of prepro[Val(8)]sodefrin showed 86.2% identity with that of the sodefrin precursor. The [Val(8)]sodefrin variant potently attracted females from the Nara area, but the variant was much less or not effective in attracting females captured in the Niigata and Chiba areas. The term aonirin ("aoni" from "aoni-yoshi", the conventional epithet of Nara) is proposed to designate this region-specific pheromone. It is speculated that the coevolution of a novel pheromone and its complementary receptor in the Nara newts may lead to reproductive isolation and eventual differentiation into a separate species. 相似文献

The gross anatomy of the mastication system of the giant anteater (Myrmecophaga tridactyla) was examined by means of three-dimensional image analysis. The anteater rotates the mandibles medially and laterally to control its tongue when it is elongated and to house it when it is relaxed. Three-dimensional CT image analysis demonstrated that the shape and size of the oral cavity changes drastically when the mandibles are rotated. The oral cavity expands bilaterally when the dorsal part of the mandibles bend medially. Macroscopic observations and muscle-weight data supported the observation that the superficial temporal and medial pterygoid muscles act as the main medial and lateral rotators of the mandible, respectively. The low height of the mandibular ramus and the incomplete zygomatic arch in this species represent adaptations for the rotational movement of the mandibles, since they both contribute to the medially oriented transmission of force from the temporal muscles and to preventing collision between the mandibles and the cranium during the rotational movement. 相似文献

Microfluid filters were fabricated, which possessed 2,100 cylindrical through-bores (psi 40 microm) in 200 microm-thickness polymethylmethacrylate (PMMA) sheets (psi 3 mm), by deep X-ray lithography using synchrotron radiation. To evaluate the microfluid filters as a device for an immunoassay, we bound the goat anti-mouse immunoglobulin G (IgG) antibody to the surface of the filters, and set the filters between reaction vessels stacked vertically in a microreactor. An enzyme-linked immunosorbent assay (ELISA) of mouse IgG using the goat anti-mouse IgG/horseradish-peroxidase (HRP) conjugate indicated that mouse IgG could be quantitatively detected in the range of 0-100 ng/ml, demonstrating the applicability of vertical microfluidic operation to the immunoassay. 相似文献

Recent studies have revealed that cytoplasmic processing bodies (P-bodies) play important roles in the control of eukaryotic gene expression in response to stress. Since the formation of P-bodies is in dynamic competition with translation, the status of translation is reflected in the assembly and disassembly of P-bodies in eukaryotic cells. During the brewing of Japanese sake and the making of wine, yeast cells are exposed to stress caused by increases in the concentration of ethanol. Here we found that ethanol stress enhances the formation of P-bodies in yeast cells in SD medium. In the wine-making process, P-body formation was also enhanced as alcoholic fermentation proceeded, but the formation of P-bodies was not simply affected by the ethanol concentration in the sake mash. These findings suggest differences in the rate of translation and the cytoplasmic mRNA flux during the sake brewing and wine making processes. 相似文献

A new dinoflagellate Durinskia capensis Pienaar, Sakai et Horiguchi sp. nov. (Peridiniales, Dinophyceae), from tidal pools along the west coast of the Cape Peninsula, Republic of South Africa, is described. The dinoflagellate produces characteristic dense orange-red colored blooms in tidal pools. The organism is characterized by having a eukaryotic endosymbiotic alga. Ultrastructure study revealed the organism has a cellular construction similar to that of other diatom-harboring dinoflagellates. The cell is thecate and the plate formula is: Po, x, 4', 2a, 6', 5c, 4s, 5', 2', which is the same as that of Durinskia baltica, the type species of the genus Durinskia. D. capensis can, however, be distinguished from D. baltica by overall cell shape, the relative size of the 1a and 2a plates, the degree of cingular displacement, and the shape of the eyespot. Our molecular analysis based on SSU rDNA revealed that D. capensis is closely allied to D. baltica, thus supporting the assignment of this new species to this genus. This Durinskia clade takes a sister position to another diatom-harboring dinoflagellate clade, which includes Kryptoperidinium foliaceum and Galeidinium rugatum. Molecular analysis based on the rbcL gene sequence and ultrastructure study revealed that the endosymbiont of D. capensis is a diatom. The SSU rDNA gene trees indicated that four species with a diatom endosymbiont formed a clade, suggesting a single endosymbiotic origin. 相似文献

BACKGROUND: Brain natriuretic peptide (BNP) has been widely used for the diagnosis and prognostic evaluation of chronic heart failure (CHF). In the present study, we performed association study of single nucleotide polymorphisms (SNPs) surrounding the natriuretic peptide precursor B (NPPB) gene with plasma BNP levels in 2970 adult Japanese. METHODS AND RESULTS: Association analysis between SNPs of the NPPB gene and plasma BNP revealed significant associations of the 8 SNPs surrounding the entire NPPB gene with plasma BNP levels. For instance, as to SNP rs198389 (T-381C), plasma BNP levels among the three genotypic categories, i.e., 2189 homozygous T-allele carriers (BNP 26.4+/-0.6pg/ml), 697 heterozygous carriers (35.0+/-1.1pg/ml), and 52 homozygous C-allele carriers (46.0+/-4.1pg/ml) indicated a co-dominant effect of the minor C-allele on elevating plasma BNP levels (P<0.0001). Linkage disequilibrium (LD) analysis among the 8 SNPs revealed that the region consisted of two, 5' major and 3' minor, LD blocks. Haplotype-based association analysis demonstrated that plasma BNP levels were associated closely with the haplotypes-1 and -2 of the major LD block. CONCLUSION: These results suggest that genetic variation at the primary locus NPPB gene, represented by definition of risk haplotypes, may be an important determinant of plasma BNP levels. 相似文献

Antisense targeting of tumor with fluorescent conjugated DNA oligomers has the potential of improving tumor/normal tissue ratios over that achievable by nuclear antisense imaging. When administered as a linear duplex of two fluorophore-conjugated oligomers arranged in a manner that inhibits fluorescence as the duplex and designed to dissociate only in the presence of the target mRNA, the fluorescence signal should in principle be inhibited everywhere except in the target cell. Optical imaging by fluorescence quenching using linear fluorophore-conjugated oligomers has not been extensively investigated and may not have been previously considered for antisense targeting. We evaluated in cell culture and in KB-G2 tumor bearing nude mice a 25-mer phosphorothioate (PS) anti- mdr1 antisense DNA conjugated with the Cy5.5 emitter on its 3' equivalent end and hybridized as a linear duplex with a shorter 18-mer phosphodiester (PO) complementary DNA (cDNA) with the Black Hole inhibitor BHQ3 on its 5' end. In serum environments, 90% of the DNA25-Cy5.5 fluorescence was inhibited immediately following addition of the cDNA18-BHQ3 and showed only slight loss of inhibition over 24 h at 37 degrees C. As evidence of antisense specific binding, when incubated with the DNA25-Cy5.5/cDNA18-BHQ3 duplex, the fluorescence was lower in KB-31 (Pgp +/-) cells compared to KB-G2 (Pgp++) cells, but when incubated with the control cDNA18-Cy5.5/DNA25-BHQ3 duplex in which the fluorophores were reversed, the fluorescence of both cell types was low. As further evidence of specific binding, the fluorescent intensity of total RNA from KB-G2 cells incubated with the study duplex showed evidence of dissociation and hybridization with the target mRNA. Furthermore, the fluorescence microscopy images of KB-G2 cells incubated with DNA25-Cy5.5 as the singlet or study duplex show that migration in both cases is to the nucleus. The animal studies were performed in mice bearing KB-G2 tumor in one thigh and receiving iv the study or control duplexes. The tumor/normal thigh fluorescence ratio was clearly positive as early as 30 min postinjection in the study mice and reached a maximum at 5 h. By contrast, much lower fluorescence was observed in mice receiving the control duplex at the same dosage. Fluorescence microscope imaging showed that the Cy5.5 fluorescence was much higher in tumor sections from the animal that had received the study rather than control duplex. Thus combining a fluorophore-conjugated antisense DNA with an inhibitor-conjugated shorter complementary cDNA inhibited fluorescence both in cell culture and in tumored animals except in the presence of the target mRNA. This proof of concept investigation of optical antisense targeting therefore suggests that further studies including optimization of this approach are appropriate. 相似文献
