A new amide, isoseryl S-methylcysteamine sulfoxide, was isolated from the tubers of Sarutori-Ibara (Smilax china, Liliaceae). Conjugated metabolites of abscisic acid (ABA) have been characterized by their chemical ionization (CI) mass spectra using isobutane and ammonia as reagent gases. The CI mass spectra usually consist of quasimolecular ions ([QM]+), and constituent aglycone- and.sugar-derived ions. Hence, the spectra are simply interpreted and afford useful information for structutal characterization. CI examinations of ABA and its unconjugated metabolites and collisionally activated dissociation (CAD) measurements of selected ions were helpful in characterizing the constituent aglycones. The distinction between the two glycosidal forms, glycoside and glycosyl ester, has been also discussed in the corresponding conjugates. Confirmation of the fragmentation pathways via recognition of the diagnostic ions can be made by the extensive use of ammonia- d3 as an additional reagent gas. 相似文献

The effects of sera from diabetic patients and healthy donors on the synthesis of prostaglandin I2 (PGI2) and PGI3in vitro were studied in confluent bovine aortic endothelial cells cultured with EPA. The products 6-keto-PGF1 and Δ17-6-keto-PGF1 were measured by GC/SIM as markers of PGI2 and PG73 formation in growth medium after 60 min of incubation. PGI2 and PG73 synthesis with 10% diabetic sera were less than with sera from healthy donors (p < 0.05). However, the total prostacyclin production (PGI2 and PGI3) in the cell cultures incubated with 10 μM EPA and 10% diabetic sera approximated that of the cultures incubated with the sera of healthy donors without EPA. These results suggest that the diabetic sera inhibits PGI2 and PGI3 synthesis in the cultured endothelial cells, and that EPA intake may reduce the complications of diabetes mellitus, such as microangiopathy and vaso-occlusive diseases, and enhances the production of PGI3 which seems to exert a strong anti-aggregatory effect. 相似文献

During dismantling and relocation of the Takamatsuzuka Tumulus stone chamber, many Cladophialophora and chaetothyrialean black fungi, such as Exophiala and Phialophora, were isolated from samples taken from the joints between the stone walls. However, inside the stone chamber of the Kitora Tumulus, after intermittent UV irradiation in 2009, these black fungi were also isolated from samples taken from the stone walls. Molecular phylogenetic analyses based on only nrLSU and the concatenated (nrLSU D1/D2 + ITS) sequences revealed that the 35 Takamatsuzuka and Kitora Tumuli isolates of Cladophialophora and the chaetothyrialean black fungi were divergent. Two new species of Cladophialophora are described herein: C. tumulicola from the viscous gels and various substrates on the stone walls of the Takamatsuzuka and Kitora Tumuli and C. tumbae from black substances on the plastic cover over the “thief hole,” soil and plaster pieces between the stone walls, and the exterior of the Takamatsuzuka Tumulus chamber. Also, molecular phylogenetic placements for the remaining eight Takamatsuzuka and Kitora Tumuli isolates of chaetothyrialean black fungi have been determined or suggested. 相似文献

Although organisms can alter dynamics of elements in ecosystems via physiological results, the effects of tree species on ecosystem nutrient dynamics are highly uncertain. A four-fold variation in the calcium concentrations of streams, soils and leaf litters were caused by the planting of Cryptomeria japonica in south-central Japan. In this study, we examined how the calcium dynamics were affected by the planting of C. japonica through strontium isotope analysis. We predicted the planting of C. japonica would result in the calcium concentration increasing because of the significant dissolution of calcium from bedrock. In a forest ecosystem, calcium is usually derived from precipitation and bedrock weathering, and their relative contributions can be estimated using a strontium isotope mixing model. Therefore, we collected stream water, litter, soil, precipitation and bedrock samples from 17 sites in catchments dominated by C. japonica plantation or evergreen broad-leaved forest; after collection, we analyzed the sample chemical compositions and strontium isotope ratios. The calcium concentrations in the stream water and the water-soluble calcium in the soil were significantly higher at sites dominated by C. japonica than at broad-leaved forest sites. Strontium isotope analysis indicated that there was more calcium from the bedrock present in stream water at sites dominated by C. japonica than in stream water at broad-leaved forest sites. Our results showed that watershed-scale dynamics of calcium and other cations can be altered by the type of vegetation in a catchment due to the effects of vegetation on the supply of calcium from bedrock. 相似文献

Background aims
We have previously reported the generation of a current Good Manufacture Practice (cGMP)-compliant induced pluripotent stem cell (iPSC) line for clinical applications. Here we show that multiple cellular products currently being considered for therapy can be generated from a single master cell bank of this or any other clinically compliant iPSC lineMethods
Using a stock at passage 20 prepared from the cGMP-compliant working cell bank (WCB), we tested differentiation into therapeutically relevant cell types of the three germ layers using standardized but generic protocols. Cells that we generated include (i) neural stem cells, dopaminergic neurons and astrocytes; (ii) retinal cells (retinal pigment epithelium and photoreceptors); and (iii) hepatocyte, endothelial and mesenchymal cells. To confirm that these generic protocols can also be used for other iPSC lines, we tested the reproducibility of our methodology with a second clinically compliant lineResults
Our results confirmed that well-characterized iPSC lines have broad potency, and, despite allelic variability, the same protocols could be used with minimal modifications with multiple qualified lines. In addition, we introduced a constitutively expressed GFP cassette in Chr13 safe harbor site using a standardized previously described method and observed no significant difference in growth and differentiation between the engineered line and the control line indicating that engineered products can be made using a standardized methodologyConclusions
