To explore the protein kinase family enzymes expressed in cells, we attempted to generate antibodies that could detect a wide variety of protein kinases. For the production of such antibodies, synthetic peptides corresponding to amino acid sequences of a highly conserved subdomain (subdomain VIB) of the protein kinase family were used for immunization. Among the various peptide antigens, a peptide with 16 amino acids, CVVHRDLKPENLLLAS, effectively produced polyclonal antibodies with broad cross-reactivities to protein kinases. Two monoclonal antibodies, designated M8C and M1C, detected a variety of protein kinases such as calmodulin-dependent protein kinase II, calmodulin-dependent protein kinase IV, cAMP-dependent protein kinase, and mitogen-activated protein kinases, on Western blotting. The antibodies also immunoprecipitated various protein kinases in cell extracts. Furthermore, these antibodies could be used for detection of positive clones in the expression cloning of various protein kinases. Among 39 positive clones obtained from mouse brain cDNA library, 36 clones were identified as cDNA clones for various known and novel protein serine/threonine kinases, suggesting that the antibodies reacted highly specifically with various protein kinases. These results indicate that the present monoclonal antibodies directed to multiple protein kinases will be a powerful tool for the detection of a variety of known and novel protein kinases in cells.  相似文献

Caspase-activated DNase (CAD) is a key protein in the process of apoptosis that degrades DNA through the action of caspases. Its N-terminal region, the CAD domain (CAD-CD), is highly conserved among CAD family proteins and is responsible for the interaction with its inhibitor. We report here that CAD-CD spontaneously aggregates to form amyloid fibrils, without a lag time, under the conditions of low pH (below 4) and the presence of anions. Interestingly, the secondary structure of CAD-CD in the fibril state comprised not only beta-sheet but also alpha-helix, as found in CD, FTIR, and x-ray fiber diffraction experiments. Aromatic side chains have a defined orientation and are in the hydrophobic environment occurring with the CAD-CD fibrillogenesis. These findings provide new insights into the architecture of amyloid fibrils.  相似文献

Ca(2+)-binding sites I and II in the N-terminal lobe of molluscan troponin C (TnC) have lost the ability to bind Ca(2+) due to substitutions of the amino acid residues responsible for Ca(2+) liganding. To evaluate the functional importance of the Ca(2+)-deficient N-terminal lobe in the Ca(2+)-regulatory function of molluscan troponin, we constructed chimeric TnCs comprising the N-terminal lobes from rabbit fast muscle and squid mantle muscle TnCs and the C-terminal lobe from akazara scallop TnC, TnC(RA), and TnC(SA), respectively. We characterized their biochemical properties as compared with those of akazara scallop wild-type TnC (TnC(AA)). According to equilibrium dialysis using (45)Ca(2+), TnC(RA), and TnC(SA) bound stoichiometrically 3 mol Ca(2+)/mol and 1 mol Ca(2+)/mol, respectively, as expected from their primary structures. All the chimeric TnCs exhibited difference-UV-absorption spectra at around 280-290 nm upon Ca(2+) binding and formed stable complexes with akazara scallop troponin I, even in the presence of 6M urea, if Ca(2+) was present. However, when the troponin complexes were constructed from chimeric TnCs and akazara scallop troponin T and troponin I, they showed different Ca(2+)-regulation abilities from each other depending on the TnC species. Thus, the troponin containing TnC(SA) conferred as high a Ca(2+) sensitivity to Mg-ATPase activity of rabbit actomyosin-akazara scallop tropomyosin as did the troponin containing TnC(AA), whereas the troponin containing TnC(RA) conferred virtually no Ca(2+) sensitivity. Our findings indicate that the N-terminal lobe of molluscan TnC plays important roles in molluscan troponin regulation, despite its inability to bind Ca(2+).  相似文献

As demonstrated with alr2835 (hepA) and alr2834 (hepC) mutants, heterocysts of Anabaena sp. strain PCC 7120, a filamentous cyanobacterium, must have an envelope polysaccharide layer (the Hep+ phenotype) to fix dinitrogen in an oxygen-containing milieu (the Fox+ phenotype). Transpositions presumptively responsible for a Fox- phenotype were localized in open reading frames (ORFs) near hepA and hepC. A mutation in each of nine of these ORFs was complemented by a clone bearing only that single, intact ORF. Heterocysts of the nine mutants were found to lack an envelope polysaccharide layer. Complementation of mutations in alr2832 and alr2840 may have resulted from recombination. However, alr2825, alr2827, alr2831, alr2833, alr2837, alr2839, and alr2841, like hepA and hepC, are required for a Hep+ Fox+ phenotype.  相似文献

Ferric binding protein in Neisseria gonorrhoeae (nFbpA) transports iron from outer membrane receptors for host proteins across the periplasm to a permease in an alternative pathway to the use of siderophores in some pathogenic bacteria. Phosphate and nitrilotriacetate, both at pH 8, and vanadate at pH 9 are shown to be synergistic in promoting ferric binding to nFbpA, in contrast to carbonate and sulfate. Interestingly, only phosphate produces the fully closed conformation of nFbpA as defined by native electrophoresis. The role of phosphate was probed by constructing three mutants: Q58E, Q58R, and G140H. The anion and iron binding properties of the Q58E mutant are similar to the wild-type protein, implying that one phosphate oxygen is a hydrogen bond donor and may in part define the specificity of nFbpA for phosphate over sulfate. Phosphate is a weakly synergistic anion in the Q58R and G140H mutants, and these mutants do not form completely closed structures. Ferric binding was investigated by both isothermal titration and differential scanning calorimetry. The apparent affinity of nFbpA for iron in a solution of 30 mM citrate is 1 order of magnitude larger in the presence (K(app)= 1.7 x 10(5) M(-1)) of phosphate than in its absence (K(app) = 1.6 x 10(4) M(-1)) at pH 7. Similar results were obtained at pH 8. This increase in affinity with phosphate as well as the formation of closed structure allows nFbpA to compete for free ferric ions in solution and suggests that ferric binding to nFbpA is regulated by the synergistic phosphate anion at sites of iron uptake.  相似文献

High-density herbivore species often play an important role in forest regeneration. Native sika deer (Cervus nippon yakushimae) inhabit a high density (51.5–63.8head/km2, estimated by a pellet count method) area in the western part of a lowland natural forest on Yakushima Island, Japan. To test experimentally the impact of sika deer on the mortality and the survivability of current-year seedlings, which are at a more vulnerable stage than the later stages, we constructed fenced exclosures, planted seeds of nine sapfruit tree species and compared the mortality and the survivability of current-year seedlings between fenced and unfenced quadrats. Large seeded species had significantly greater survivability in fenced quadrats than in unfenced quadrats. However, the survivability disagreed with feeding preferences. Sika deer activity increased seedling mortality of large-seeded species more than that of small-seeded species, and did not decrease much seedling survivability of not-preferred species. We found that the physical disturbance by the high density of sika deer resulted in mortality for both preferred and not-preferred species, and that deer herbivory was important for preferred species.  相似文献

L-threo-beta-Benzyloxyaspartate (L-TBOA) and (2S,3S)-3-[3-[4-(trifluoromethyl)benzoylamino]benzyloxy]aspartate (L-TFB-TBOA) are potent nontransportable blockers for glutamate transporters. We synthesized a carbamate-type coumarin derivative of L-TBOA 3a as a caged blocker and compared 3a with the corresponding ester-type analogs 1. The carbamate 3a was less sensitive to photolysis than the ester 1 but was more stable in the aqueous solution. The [6,7-bis(carboxymethoxy)-coumarin-4-yl]methylcarbonyl (BCMCMC) group exhibited good results both in photoreactivity and stability. Therefore, we examined photolysis of N-BCMCMC-TBOA 3b and N-BCMCMC-TFB-TBOA 4, which immediately released blockers to show glutamate uptake inhibition.  相似文献

Based on our reexamination of the 9 specimens including the neotype, Bregmaceros lanceolatus is recharacterized and diagnosed by the following combination of features: caudal fin rounded; scales present on gill cover; dorsal surface of snout unpigmented or with a few chromatophores; isthmus pigmented with punctate chromatophores; two parapophyses on abdominal vertebrae; dorsal rays (D) 65–74; anal rays (A) 67–74; vertebrae (V) 58–61; longitudinal scales (LS) ca. 82–88; principal caudal rays (PC) 16–18; head length (HL)/standard length (SL) 14.0–15.5%; caudal peduncle depth/SL 3.2–4.2%. Based on 27 specimens, B. pseudolanceolatus sp. nov. is described. This species is closely similar to B. lanceolatus, but is diagnosed by the following combination of features: caudal fin rounded; scales present on gill cover; dense concentration of chromatophores on dorsal surface of snout; isthmus colorless; one board-like parapophysis on the last three abdominal vertebrae; D 58–64; A 58–67; V 52–55; LS ca. 68–77; PC 14–16; HL 15.5–18.4% SL; caudal peduncle depth 4.1–5.2% SL. Bregmaceros pseudolanceolatus is known from around the Taiwan Strait, southern East China Sea, South China Sea, Gulf of Thailand, Timor Sea, Arafura Sea, and eastern Bay of Bengal.  相似文献

We studied the effects of the timing of urea treatment on the succession of ammonia fungi. In two evergreen Castanopsis cuspidata forests and in one deciduous Quercus serrata forest, we applied 343g urea to 25 and 15 plots of 0.5m2, respectively, at three different times of the year. Ten of the early-phase (EP) species, considered to be saprotrophic, and 6 of the late-phase (LP) ones, considered ectomycorrhizal, fruited. In both phases, the commencement, peak, and cessation of fruiting took place simultaneously among all the plots treated at the same time. The fruiting occurred in summer and autumn. Quantity and size of the fruit bodies was larger in the LP than in the EP species. Fruiting of EP species was affected by the treatment time and that of LP species by interaction of the treatment time and vegetation type. EP was short and occurred as one period, whereas LP was long and occurred as two or more fruiting seasons. We found that species composition, dominant species, and degree of its dominance in fruiting of the ammonia fungi are predictable for different treatment times of the year and different vegetation types.  相似文献
