The effects of three catecholamines, dopamine, epinephrine, and dobutamine, on the systemic circulation, especially on systemic vascular capacitance, were studied using cardiopulmonary bypass in dogs anesthetized with pentobarbital. Venous outflow was divided into three compartments: splanchnic, renal, and other; changes in systemic blood volume (SBV) were calculated from the changes in total venous outflow. To examine the contribution of sympathetic discharge to these vascular responses, sympathetic efferent nerve activity (SENA) from the ventral ansa subclavian nerve was recorded simultaneously. Experiments were done under three conditions: control, after baroreceptor deafferentation, and after hexamethonium injection with low and high doses of each catecholamine. During control and after baroreceptor deafferentation, dopamine- and epinephrine-induced changes in SBV were less than those after hexamethonium, and not significant except with low dose epinephrine. After hexamethonium, dopamine (200 micrograms/kg), epinephrine (10 micrograms/kg), and dobutamine (100 micrograms/kg) reduced SBV by 10.6 +/- 3.4, 13.1 +/- 1.7, and 1.9 +/- 0.3 mL/kg, respectively. Splanchnic outflow increased significantly with dopamine and epinephrine after hexamethonium. High dose dopamine and epinephrine significantly suppressed SENA to 38 +/- 9 and 15 +/- 6% of baseline, respectively. Low dose dopamine decreased arterial pressure and SENA. This suppression in SENA was attenuated but still observed after baroreceptor deafferentation. Dobutamine reduced SBV, but had no effect on SENA. These results suggest that dopamine and epinephrine primarily decrease SBV by venoconstriction in the splanchnic region, however, these effects are greatly modified by basal sympathetic discharge and changes in SENA and vascular tone.  相似文献

The enhancement of antitumor activities of the tumoricidal soluble factor (SF) from a streptococcal preparation (OK-432)-activated macrophages by the pretreatment with a protein-bound polysaccharide (PSK) was investigated in tumor-bearing mice.Two-step stimulations with OK-432 atin vivo priming andin vitro eliciting were required for the production of the tumoricidal SF by macrophages, and the tumoricidal activity of the SF apparently correlated with the uptake of OK-432 by macrophages at priming phase.Tumoricidal activity of the SF from OK-432-activated macrophages in proteose-peptone (P-P)-pretreated mice significantly decreased with the development of the tumor, whereas in PSK-pretreated mice did not. Pretreatment of tumor-bearing mice with PSK saved a decrease in the macrophages carrying Iak or asialo GM1 antigens and an increase in wheat germ agglutinin (WGA) receptors. Furthermore, the uptake of OK-432 by macrophages at priming phase was enhanced. The tumoricidal activity of the SF from OK-432-activated macrophages was augmented.Thus, PSK may restore the depressed functions of macrophages, and the combination therapy with PSK and OK-432 may be effective to enhance the production of tumoricidal SF in tumor-bearing mice.  相似文献

Effects of administration of the LHRH agonist D-Leu6-LHRH ethylamide (LHRH-A), gonadotropin (PMS), and their interaction on testicular prolactin (PRL) receptor levels were investigated in rats. LHRH-A (2 micrograms/100 g body wt.) or saline was injected SC daily, and PMS (5 IU) injected every other day. In intact rats, the testicular PRL receptor levels were about 400 fmoles/testis after either 1 or 7 daily injections of saline. Administration of LHRH-A decreased PRL receptors to 12% of that of saline-injected control rats at day 1, and to 20% at day 2, and PRL receptor levels were partially restored to 55% at day 7. In hypophysectomized rats given daily injections of saline for 7 days PRL receptor levels were only 20% of those in saline-injected intact rats. Injections of LHRH-A in hypophysectomized animals did not further decrease PRL receptor numbers at this time. Administration of PMS to hypophysectomized rats for 7 days partially reversed the reduction of PRL receptors that occurred after hypophysectomy, to 46% of those in intact controls. Injections of LHRH-A into hypophysectomized. PMS-treated animals did not significantly alter PRL receptors on day 1 (117% of that of saline-injected, hypophysectomized, PMS-treated rats at day 1) or day 2 (96% of same-day controls), but decreased PRL receptors on day 7 to 102 fmoles/testis (55% of same-day controls). This latter concentration is nearly the same as that in saline-injected, 7-day hypophysectomized rats not treated with PMS. These findings suggest that: (1) the effects of LHRH-A on testicular PRL receptors differ depending on the presence or absence of gonadotropin, (2) gonadotropin, primarily FSH, maintains some population of testicular PRL receptors, and these gonadotropin-dependent PRL receptors are suppressed by direct action of LHRH-A upon the testes, and (3) there is a population of PRL receptors which is not affected by LHRH-A or gonadotropin.  相似文献

Summary Cultured human pancreatic carcinoma cells (MIA PaCa-2) have been shown previously to be very sensitive toE. coli l-asparaginase (EC II). The present studies have demonstrated that another enzyme,Acinetobacter glutaminase-asparaginase (AGA) is much more effective in inhibiting cell growth. At the concentration of 0.0025 U/ml of AGA activity the enzyme totally inhibited cell growth, whereas the EC II with the same concentration did not show any effect. The inhibition of cell growth correlated well with inhibition of protein and glycoprotein synthesis. The addition ofl-glutamine at the concentration of 1 mM completely reversed the inhibition of protein synthesis. Similarly, the addition ofl-glutamine at the concentration of 3 mM daily on 3 successive days after adding AGA resulted in significant reversal of growth inhibition. The results of this study indicate that the action of AGA on MIA PaCa-2 is, to a great extent, exerted through itsl-glutaminase activity. This work was supported in part by USPHS Grant CA 19182. Summary The superior cervical ganglia (SCG), celiac superior mesenteric ganglia (CMG), and splanchnic nerve of unoperated guinea pigs, as well as both proximal and distal stumps of a previously transected branch of the postganglionic plexus of the CMG, were immunostained for somatostatin (SS). In addition, the PAP technique was adapted for fine-structural visualization of SS. A greater proportion of cells were labeled for SS in the CMG than in the SCG. PAP molecules were present in one type of intraganglionic axons. Only two labeled axons were found in the splanchnic nerve. Neither proximal nor the distal stump of the transected CMG postganglionic nerve contained labeled axons. The present results support the hypothesis that the intraganglionic axons labeled for SS arise from SS-containing intraganglionic neurons.  相似文献

Small circular DNA complexes in eucaryotic cells   总被引:7,自引:0,他引:7  
A small number of eucaryotic cells (100 to 1000 cells) were pressed by mica sheet; then the extruded contents were adsorbed on mica and processed for electron microscopy. In the absence of divalent cation, small polydisperse circular DNA molecules bound to proteins or membrane material were preferentially adsorbed. The small circular DNA complexes have been found in every eucaryotic cell, primary lymphoid tissue cells of bursa and thymus, primary cell lines of retina and liver, and established cultured cell lines of embryonal teratocarcinoma, F9 and PCC3, HeLa and 3T6. Size distribution of these DNA complexes varies, depending on the cell source. The circles less than 1 μm in contour length predominate in cultured cell lines and the larger ones in primary cell lines and cells in situ. Polydisperse covalently closed circular DNAs were recovered from thymus lymphocytes by the conventional dye-CsCl buoyant density method. Their size distribution was similar to that of the small circular DNA complexes detected by the mica-press-adsorption method. They are present in several tens to hundreds of copies per cell representing, at a maximum, 0.02% of the total cellular DNA. The possibility that small circular DNA complexes may result from gene rearrangement as well as from replicon “misfiring” (A. Varshavsky, 1981, Proc. Nat. Acad. Sci.
