NH4 + transport system of a psychrophilic marine bacterium Vibrio sp. strain ABE-1 (Vibrio ABE-1) was examined by measuring the uptake of [14C]methylammonium ion (14CH3NH3 +) into the intact cells. 14CH3NH3 + uptake was detected in cells grown in medium containing glutamate as the sole nitrogen source, but not in those grown in medium containing NH4Cl instead of glutamate. Vibrio ABE-1 did not utilize CH3NH3 + as a carbon or nitrogen source. NH4Cl and nonradiolabeled CH3NH3 + completely inhibited 14CH3NH3 + uptake. These results indicate that 14CH3NH3 + uptake in this bacterium is mediated via an NH4 + transport system and not by a specific carrier for CH3NH3 +. The respiratory substrate succinate was required to drive 14CH3NH3 + uptake and the uptake was completely inhibited by KCN, indicating that the uptake was energy dependent. The electrochemical potentials of H+ and/or Na+ across membranes were suggested to be the driving forces for the transport system because the ionophores carbonylcyanide m-chlorophenylhydrazone and monensin strongly inhibited uptake activities at pH 6.5 and 8.5, respectively. Furthermore, KCl activated 14CH3NH3 + uptake. The 14CH3NH3 + uptake activity of Vibrio ABE-1 was markedly high at temperatures between 0° and 15°C, and the apparent K m value for CH3NH3 + of the uptake did not change significantly over the temperature range from 0° to 25°C. BACKGROUND: The long-term benefit of Helicobacter pylori eradication treatment that includes metronidazole on peptic ulcer disease in Japan is unclear. We investigated the rate of H. pylori re-infection and ulcer relapse after H. pylori eradication. MATERIALS AND METHODS: A total of 266 patients with endoscopically confirmed peptic ulcer disease and H. pylori infection were treated with triple therapy of omeprazole 40 mg (20 mg b.i.d.), clarithromycin 800 mg (400 mg b.i.d.), and tinidazole 1000 mg (500 mg b.i.d.) for 7 days. Endoscopy with gastric biopsy was performed before and 1 month, 6 months, 1.5 years, and 3.5 years after therapy. H. pylori status was determined by H. pylori culture, rapid urease test, and histopathology. 13C-urea breath test was done at 6 months after eradication therapy. Treatment was deemed successful when all tests were negative at 6 months after therapy by endoscopic biopsy. RESULTS: Successful H. pylori eradication was achieved in 262/266 (98.5%) patients with peptic ulcer. Total relapse of peptic ulcer occurred in 8/262 (3%) patients after eradication, with 3/262 (1.1%) occurring within 1.5 years after treatment and 5/262 (1.9%) within 3.5 years. All relapsed patients were found to be H. pylori-positive at the time of relapse. Of the 262 patients who experienced eradication, 20 (7.6%) were subsequently re-infected, six (2.3%) within 1.5 years and 14 (5.3%) within 3.5 years. CONCLUSION: Triple therapy with omeprazole, clarithromycin, and tinidazole (OCT) is useful for H. pylori eradication in Japan, but there is an appreciable re-infection rate in this population.  相似文献

Variations in cardiovascular responses to orthostatic stress were investigated in terms of physiological polymorphism. Variations of physiological measurements are subdivided into individual differences and measurement errors. However, individual differences are often considered to be an error in statistical analysis due to its limitations in experimental design. In order to discuss about the relative contribution of individual difference in cardiovascular responses to postural changes, percent contribution (PC) was estimated using the Taguchi method. Six healthy male adults (age range: 21-27) were subjected to orthostatic stress by inducing a postural inclination of 60 degrees head-up-tilting to the horizontal, and the responses were measured thrice in each subject on different days. The respective changes of heart rate (HR) and stroke volume (SV) in the period from the resting supine to the head-up-tilt position were significantly increased (p < 0.01) and decreased (p < 0.01) without affecting the mean blood pressure (MBP). The PC of individual difference in HR showed a significantly higher ratio of individual difference during the head-up-tilt (71.4-76.2%) compared with supine rest (0.0-50.4%). While the main variations of HR during supine rest were not the individual differences between the subjects, the day-to-day differences within the subject were significant. The PC of individual differences in MBP and SV constantly displayed a significant difference between the subjects. These results suggest that the strategy for maintaining stable cardiovascular regulation may be different even in normal subjects. In the perspective of physiological parameters, PC monitoring may serve as an empirical approach to evaluate physiological polymorphism.  相似文献

Cardiomyocytes derived from mouse embryonic stem (mES) cells have been demonstrated to exhibit a time-dependent expression of ion channels and signal transduction pathways in electrophysiological studies. However, ion transporters, such as Na+/K+ ATPase (Na+ pump) or Na+/Ca2+ exchanger, which play crucial roles for cardiac function, have not been well studied in this system. In this study, we investigated the functional expression of Na+/K+ ATPase and Na+/Ca2+ exchanger in mES cells during in vitro differentiation into cardiomyocytes, as well as the functional coupling between the two transporters. By measuring [Na+]i and Na+ pump current (Ip), it was shown that an ouabain-high sensitive Na+/K+ ATPase was expressed functionally in undifferentiated mES cells and these activities increased during a time course of differentiation. Using RT-PCR, the expression of mRNA for alpha1-subunit and alpha3-subunit of the Na+/K+ ATPase could be detected in both undifferentiated mES cells and derived cardiomyocytes. In contrast alpha2-subunit mRNA could be detected only in derived cardiomyocytes but not in undifferentiated mES cells. mRNA for the Na+/Ca2+ exchanger 1 isoform (NCX1) could be detected in undifferentiated mES cells and its expression levels seemed to gradually increase throughout the differentiation accompanied by increasing its Ca2+ extrusion function. At the middle stages of differentiation (after 10-day induction), more than 75% derived cardiomyocytes exhibited [Ca2+]i oscillations by blocking of Na+/K+ ATPase, suggesting the functional coupling with Na+/Ca2+ exchanger. From these results and RT-PCR analysis, we conclude that alpha2-subunit Na+/K+ ATPase mainly contributes to establish the functional coupling with NCX1 at the middle stages of differentiation of cardiomyocytes.  相似文献

NADPH-dependent erythrose reductases (ERs) in erythritol-producing fungi, Trichosporonoides megachiliensis SNG-42, catalyze the reduction of D-erythrose. We previously characterized the biochemical properties of three isozymes of ERs (ER-I, ER-II, and ER-III). Using internal amino acid sequences of ER-III and ER-I with peptide mapping, we cloned three cDNAs (er1, 1121-bp (AB191474); er2, 1077-bp (AB191475); er3, 1119-bp (AB191476)). The er3 cDNA encoded a polypeptide 36,044 Da, and its deduced amino acid sequence was same as that of the native ER-III. The three recombinant enzymes expressed in Escherichia coli were purified to homogeneity. The recombinant enzymes of ER1, ER2, and ER3 showed similar electrophoretic properties to that of the native ER-I, ER-II, and ER-III on SDS- and Native- but not on IEF-PAGE. All three recombinant enzymes showed substrate specificity towards C-4 and C-3 aldehydes similar to that of the native ER-III. These results strongly suggest that cloned er1, er2, and er3 cDNAs encode erythrose reductases.  相似文献

Five amino acid residues responsible for extreme stability have been identified in cytochrome c(552) (HT c(552)) from a thermophilic bacterium, Hydrogenobacter thermophilus. The five residues, which are spatially distributed in three regions of HT c(552), were replaced with the corresponding residues in the homologous but less stable cytochrome c(551) (PA c(551)) from Pseudomonas aeruginosa. The quintuple HT c(552) variant (A7F/M13V/Y34F/Y43E/I78V) showed the same stability against guanidine hydrochloride denaturation as that of PA c(551), suggesting that the five residues in HT c(552) necessarily and sufficiently contribute to the overall stability. In the three HT c(552) variants carrying mutations in each of the three regions, the Y34F/Y43E mutations resulted in the greatest destabilization, by -13.3 kJ mol(-1), followed by A7F/M13V (-3.3 kJ mol(-1)) and then I78V (-1.5 kJ mol(-1)). The order of destabilization in HT c(552) was the same as that of stabilization in PA c(551) with reverse mutations such as F34Y/E43Y, F7A/V13M, and V78I (13.4, 10.3, and 0.3 kJ mol(-1), respectively). The results of guanidine hydrochloride denaturation were consistent with those of thermal denaturation for the same variants. The present study established a method for reciprocal mutation analysis. The effects of side-chain contacts were experimentally evaluated by swapping the residues between the two homologous proteins that differ in stability. A comparative study of the two proteins was a useful tool for assessing the amino acid contribution to the overall stability.  相似文献
