Chemical techniques were investigated in order to eradicate Caulerpa taxifolia, a green alga spreading at a remarkable rate in the Mediterranean Sea. The action of copper, potassium and sodium ions on survival rates and photosynthetic parameters was compared, in order to optimise the conditions of further in situ treatments. The lethal doses were determined and the impact of the studied cations on photosynthesis and respiration rates and PSII photochemistry was analysed from measurements of net oxygen exchanges and chlorophyll fluorescence. The Cu2+ concentrations required to obtain 100% mortality were 15 × 102 to 104 times lower than those of K+ and Na+. Respiration was slightly affected whatever the salt concentration,while photosynthesis could be totally inhibited depending on the applied treatment. Changes in the structure of the Ribulose-1,5-biphosphate carboxylase/oxygenase (RubisCO, EC: 4.1.1.39) were also detected when C. taxifolia under went cation treatments (10 mg L-1 Cu2+, 1h; 20 gL-1 K+, 3 h; 20 g L-1 Na+, 1 h). Given the high concentration and long incubation periods required with K+ and Na+ ions, these cations are not suitable to be used in situ. Our results make possible the utilisation of copper cations following technical approaches such asion-exchange textile covers, which allows a controlled release of cupric ions without dissemination in the marine environment. Background

Colorectal cancer (CRC) is one of the most common malignancies but the current therapeutic approaches for advanced CRC are less efficient. Thus, novel therapeutic approaches are badly needed. The purpose of this study is to investigate the involvement of nuclear protein kinase CK2 α subunit (CK2α) in tumor progression, and in the prognosis of human CRC.

Methodology/Principal Findings

Expression levels of nuclear CK2α were analyzed in 245 colorectal tissues from patients with CRC by immunohistochemistry, quantitative real-time PCR and Western blot. We correlated the expression levels with clinicopathologic parameters and prognosis in human CRC patients. Overexpression of nuclear CK2α was significantly correlated with depth of invasion, nodal status, American Joint Committee on Cancer (AJCC) staging, degree of differentiation, and perineural invasion. Patients with high expression levels of nuclear CK2α had a significantly poorer overall survival rate compared with patients with low expression levels of nuclear CK2α. In multi-variate Cox regression analysis, overexpression of nuclear CK2α was proven to be an independent prognostic marker for CRC. In addition, DLD-1 human colon cancer cells were employed as a cellular model to study the role of CK2α on cell growth, and the expression of CK2α in DLD-1 cells was inhibited by using siRNA technology. The data indicated that CK2α-specific siRNA treatment resulted in growth inhibition.

Conclusions/Significance

Taken together, overexpression of nuclear CK2α can be a useful marker for predicting the outcome of patients with CRC.  相似文献

Al-sensitive (als) mutants of Arabidopsis were isolated and characterized with the aim of defining mechanisms of Al toxicity and resistance. Most als mutants selected on the basis of root growth sensitivity to Al were recessive, and together the mutants constituted eight complementation groups. Also, in most als mutants, Al sensitivity appeared to be specific for Al relative to La (another trivalent cation), except als2, which was more sensitive to La than wild type. The tendency of roots on mutant seedlings to accumulate Al was examined by staining with morin and hematoxylin, dyes used to indicate the presence of Al. A significant increase in morin staining was observed in als5, consistent with its increased sensitivity to Al. Unexpectedly, als7 and als4 showed less morin staining, suggesting that the roots on these mutants accumulate less Al than wild type seedlings after exposure to Al-containing solutions. Roots of wild-type seedlings produce callose in response to AlCl3 concentrations that inhibit root growth. Only als5 accumulated more callose than wild type in response to low levels (25 mu M) of AICI3 However, als4 and als7 did not accumulate callose at this AlCl3 concentration even though root growth was significantly inhibited. The lack of callose accumulation in als4 and als7 suggests that there is not an obligatory relationship between callose deposition and Al-induced inhibition of root growth.  相似文献

One of the major symptoms of diabetes mellitus (DM) is delayed wound healing, which affects large populations of patients worldwide. However, the underlying mechanism behind this illness remains elusive. Skin wound healing requires a series of coordinated processes, including fibroblast cell proliferation and migration. Here, we simulate DM by application of high glucose (HG) in human foreskin primary fibroblast cells to analyze the molecular mechanism of DM effects on wound healing. The results indicate that HG, at a concentration of 30 mM, delay cell migration, but not cell proliferation. bFGF is known to promote cell migration that partially rescues HG effects on cell migration. Molecular and cell biology studies demonstrated that HG enhanced ROS production and repressed JNK phosphorylation, but did not affect Rac1 activity. JNK and Rac1 activation were known to be important for bFGF regulated cell migration. To further confirm DM effects on skin repair, a type 1 diabetic rat model was established, and we observed the efficacy of bFGF on both normal and diabetic rat skin repair. Furthermore, proteomic studies identified an increase of Annexin A2 protein nitration in HG-stressed fibroblasts and the nitration was protected by activation of bFGF signaling. Treatment with FGFR1 and JNK inhibitors delayed cell migration and increased Annexin A2 nitration levels, indicating that Annexin A2 nitration is modulated by bFGF signaling via activation of JNK. Together with these results, our data suggests that the HG-mediated delay of cell migration is linked to the inhibition of bFGF signaling, specifically through JNK suppression.  相似文献

Apoptosis is a barrier to maintaining high viable cell densities in animal cell culture. Silkworm hemolymph and its 30K protein have been reported to exhibit anti-apoptotic activity in various mammalian and insect cell systems. The 30K protein is thermally unstable at temperatures higher than 60 degrees C; however, the silkworm hemolymph heat-treated at 70-80 degrees C still exhibited anti-apoptotic activity. This indicates that silkworm hemolymph contains another anti-apoptotic compound other than 30K protein. In this article, the anti-apoptotic molecule other than 30K protein was found from the silkworm hemolymph and identified. This molecule was storage-protein 2 (SP2), which has no homology with any known anti-apoptotic protein. This molecule was heat-stable up to 80 degrees C, while 30K protein lost its activity at temperatures higher than 60 degrees C. When apoptosis was induced by staurosporine in HeLa cells, SP2 protein suppressed nuclear fragmentation and apoptotic body formation. Moreover, the generation of reactive oxygen species after apoptosis induction was inhibited, which means the inhibition occurred in an early step of the apoptotic process. Inhibition of apoptosis by the SP2 protein would lead to the minimization of cell death during commercial mammalian cell culture.  相似文献

A number of mutations (45) expressed as cold-sensitive conditional lethal pheno-types were screened by transduction for their linkage to the streptomycin-resistance locus; 7 showed such linkage. Of these, two were studied in greater detail. The sedimentation profiles of ribosomes from cultures grown at low temperature differed from wild type and from one another. Both mutants lost ribonucleic acid control at low temperature. It is suggested that a high proportion of mutants expressing a cold-sensitive phenotype harbor mutations in genes affecting ribosome synthesis or regulation.  相似文献

A seven-generation selection experiment comprising a selected (S) and a control (C) line was conducted with the objective of increasing the number of fertile eggs (F) of the Brown Tsaiya duck after a single artificial insemination (AI) with pooled Muscovy semen. Both lines consisted of about 20 males and 60 females since parents in each generation and each female duck was tested 3 times, at 26, 29 and 32 weeks of age. The fertile eggs were measured by candling at day 7 of incubation. The selection criterion in the S line was the BLUP animal model value for F. On average, 24.7% of the females and 15% of the males were selected. The direct responses to the selection for F, and correlated responses for the number of eggs set (Ie), the number of total dead embryos (M), the maximum duration of fertility (Dm) and the number of hatched mule ducklings (H) were measured by studying the differences across the generations of selection between the phenotypic value averages in the S and C lines. The predicted genetic responses were calculated by studying the differences between the S and C lines in averaged values of five traits of the BLUP animal model. The selection responses and the predicted responses showed similar trends. There was no genetic change for Ie. After seven generations of selection, the average selection responses per generation were 0.40, 0.33, 0.42, 0.41 genetic standard deviation units for F, M, Dm, and H respectively. Embryo viability was not impaired by this selection. For days 2–8 after AI, the fertility rates (F/Ie) were 89.2% and 63.8%, the hatchability rates (H/F) were 72.5% and 70.6%, and (H/Ie) were 64.7% and 45.1% in the S and C lines respectively. It was concluded that upward selection on the number of fertile eggs after a single AI with pooled Muscovy semen may be effective in ducks to increase the duration of the fertile period and the fertility and hatchability rates with AI once a week instead of twice a week.  相似文献

The heartwood of Syringa oblata Lindl. (SO) is one of Mongolian folk medicines to treat insomnia and pain, while its pharmacological evaluation and underlying mechanism remain unclear. In this study, the sedative effect of ethanol extract of SO (ESO) was evaluated with the locomotor activity test and the threshold dose of pentobarbital sodium-induced sleep test in mice, and the hot plate test, acetic acid-induced writhing test, and formalin test in mice were used to evaluate its analgesic effect. The underlying mechanism of ESO analgesia was explored by RT-PCR and western blot analysis, which is associated with the regulation of the NF-κB signaling pathway. Besides, the main constituents of ESO were characterized by LC/MS data analysis and comparison with isolated pure compounds. The current findings brought evidence for clinical application and further pharmacological and phytochemical studies on SO.  相似文献
