Generally, the immobilization of two-dimensional nanoparticles in immersion procedures is time-consuming (over 24 h). In this paper, we report a very effective and simple method to fabricate two-dimensional gold nanoparticle patterns over large areas with high regularity for surface-enhanced Raman scattering (SERS). We achieved a highly sensitive SERS colloid surface by optimizing temperature and immersion time. The surfaces were characterized by X-ray photoelectron spectroscopy, UV–Vis, atomic force microscopy, and scanning electron microscopy. The SERS activity of surfaces was compared by using two techniques: “dip” and “dip and dry” in an aqueous solution of 10−6 M crystal violet. The influence of the morphology of the surface was investigated with both the dip and dip and dry techniques.  相似文献

Attitudinal studies are increasingly being adopted as tools for evaluating public understanding, acceptance and the impact of conservation interventions. The findings of these studies have been useful in guiding the policy interventions. Many factors affect conservation attitudes positively or negatively. The factors inspiring positive attitudes are likely to enhance the conservation objectives while those inducing negative attitudes may detrimentally undermine these objectives. The magnitude of the resultant effects of each particular factor is determined by the historical, political, ecological, socio-cultural and economic conditions and this may call for different management interventions. In this study we examined how conservation attitudes in western Serengeti are shaped by the following factors: level of conflicts with protected areas; wildlife imposed constraints (inadequate pasture, water, diseases, loss of livestock during migration, theft and depredation); participation in the community based project; and socio-demographic factors (age, education level, wealth, immigration, gender and household size). The results indicated that the level of conflicts, participation in the community based project, inadequate pasture, lack of water, diseases, wealth and education were important in shaping peoples’ attitudes. However, in a stepwise linear regression analysis, 59% of the variation in peoples’ attitudes was explained by three variables i.e., conflict level with protected areas, lack of water and participation in the community based project. In addition to these variables, level of education also contributed in explaining 51% of the variation in people’s attitude regarding the status of the game reserves. Five variables (lack of water, level of education, inadequate pasture, participation in the community based project and diseases) explained 12% of the variation in people’s attitude towards Serengeti National Park. The paper discusses the implications for conservation of these results and recommends some measures to realise effective conservation of wildlife resources.  相似文献

This study examined morphological, anatomical and palynological characters for six species, two subspecies and two varieties of perennial Onobrychis growing in northeast Iran. In the morphological study, shape, size and color of vegetative organs and legumes were assessed. For the anatomical study, cross sections of mature fresh or dried stems, leaflets and petioles were prepared and stained. In the palynological study, pollen were extracted, acetolysed and observed by SEM. The morphological study indicated that some characteristics varied among species, such as shape of stipules and calyx teeth, color and shape of corolla segments and legumes and an identification key is provided based on these characters. Also, in internal structure some differences were noticed in the shape of petiole cross sections, the thickness of cortex and pith shape in petioles and the arrangement of vessels in stems. Some anatomical characters proved useful to identify species, but not sections. Palynological results showed circular and obtuse triangular pollen in polar view and elliptical pollen in equatorial view with reticulate ornamentation. In addition, a cluster analysis was done based on morphological, anatomical and palynological features. The resulting dendrogram confirmed the taxonomic treatment suggested by Sirjaev and Rechinger.  相似文献

Leishmaniasis is a worldwide uncontrolled parasitic disease due to the lack of effective drug and vaccine. To speed up effective drug development, we need powerful methods to rapidly assess drug effectiveness against the intracellular form of Leishmania in high throughput assays. Reporter gene technology has proven to be an excellent tool for drug screening in vitro. The effects of reporter proteins on parasite infectivity should be identified both in vitro and in vivo. In this research, we initially compared the infectivity rate of recombinant Leishmania major expressing stably enhanced green fluorescent protein (EGFP) alone or EGFP-luciferase (EGFP-LUC) with the wild-type strain. Next, we evaluated the sensitivity of these parasites to amphotericin B (AmB) as a standard drug in 2 parasitic phases, promastigote and amastigote. This comparison was made by MTT and nitric oxide (NO) assay and by quantifying the specific signals derived from reporter genes like EGFP intensity and luciferase activity. To study the amastigote form, both B10R and THP-1 macrophage cell lines were infected in the stationary phase and were exposed to AmB at different time points. Our results clearly revealed that the 3 parasite lines had similar in vitro infectivity rates with comparable parasite-induced levels of NO following interferon-γ/lipopolysaccharide induction. Based on our results we proposed the more reporter gene, the faster and more sensitive evaluation of the drug efficiency.  相似文献

A group of 2-alkoxy-5-methoxyallylbenzene were designed, synthesised and evaluated as potential inhibitors of the soybean 15-lipoxygenase (SLO) on the basis of the eugenol and esteragol structures. Compound 4d showed the best half maximal inhibitory concentration (IC??) for SLO inhibition (IC???=?5.9?±?0.6 μM). All the compounds were docked in the SLO active site retrieved from the Research Collaboratory for Structural Bioinformatics (RCSB) Protein Data Bank (PDB entry: 1IK3) and showed that the allyl group of the synthetic compounds similar to the linoleic acid double bond, were oriented toward the Fe3+-OH moiety in the active site of the enzyme and this conformation was especially fixed by the hydrophobic interaction of the 2-alkoxy group with Leu?1?, Trp?1?, Val??? and Ile??2. It was concluded that the molecular volume and shape of the alkoxy moiety was a major factor in the inhibitory potency variation of the synthetic compounds.  相似文献

Arginine kinase-1 (MrAK-1) was sequenced from the freshwater prawn Macrobrachium rosenbergii using Illumina Solexa Genome Analyzer Technique. MrAK-1 consisted of 1068 bp nucleotide encoded 355 polypeptide with an estimated molecular mass of 40 kDa. MrAK-1 sequence contains a potential ATP:guanido phosphotransferases active domain site. The deduced amino acid sequence of MrAK-1 was compared with other 7 homologous arginine kinase (AK) and showed the highest identity (96%) with AK-1 from cherry shrimp Neocaridina denticulate. The qRT-PCR analysis revealed a broad expression of MrAK-1 with the highest expression in the muscle and the lowest in the eyestalk. The expression of MrAK-1 after challenge with the infectious hypodermal and hematopoietic necrosis virus (IHHNV) was tested in muscle. In addition, MrAK-1 was expressed in Escherichia coli by prokaryotic expression plasmid pMAL-c2x. The optimum temperature (30 °C) and pH (8.5) was determined for the enzyme activity assay. MrAK-1 showed significant (P < 0.05) activity towards 10-50 mM ATP concentration. The enzyme activity was inhibited by α-ketoglutarate, glucose and ATP at the concentration of 10, 50 and 100 mM respectively. Conclusively, the findings of this study indicated that MrAK-1 might play an important role in the coupling of energy production and utilization and the immune response in shrimps.  相似文献

Leishmania major (L. major) signal peptidase type I (SPase I) is an endopeptidase encoded by a single-copy gene. In all organisms, SPase I is responsible for removing the signal peptide from secretory pre-proteins and releasing mature proteins to cellular or extra-cellular space. In this study, the role of SPase I in L. major is investigated by gene deletion using homologous recombination (HR). The null mutant of SPase I was not possible to create, suggesting that SPase I is an essential gene for parasite survival.The obtained heterozygote mutant by disrupting one allele of SPase I in L. major showed significantly reduced level of infectivity in bone marrow-derived macrophages. In addition, the heterozygote mutants are unable to cause cutaneous lesion in susceptible BALB/c mice. This is the first report showing that SPase I may have an important role in Leishmania infectivity, e.g. in differentiation and survival of amastigotes. Apparently, the SPase I expression is not essential for in vitro growth of the parasite.  相似文献
