Six strains of denitrifying bacteria isolated from various oxic and anoxic habitats on different monocyclic aromatic substrates were characterized by sequencing 16S rRNA genes, determining physiological and morphological traits, and DNA-DNA hybridization. According to these criteria, strains S100, SP and LG356 were identified as members of Thauera aromatica. Strains B5-1 and B5-2 were tentatively affiliated to the species Azoarcus tolulyticus. Strains B4P and S2 were only distantly related to each other and to other described Thauera species. These two strains are proposed as the type strains of two new species, Thauera phenylacetica sp. nov. and Thauera aminoaromaticasp. nov., respectively. By 16S rRNA gene analysis, strain U120 was highly related to the type strains of Azoarcus evansii and Azoarcus anaerobius, whereas corresponding DNA-DNA reassociation values indicated only a low degree of genomic relatedness. Based upon a low DNA similarity value and the presence of distinguishing physiological properties, strain U120 is proposed as the type strain of a new species, Azoarcus buckelii sp. nov. Almost all of the new isolates were obtained with different substrates. The highly varied substrate spectra of the isolates indicates that an even higher diversity of denitrifying bacteria degrading aromatic compounds would be discovered in the different habitats by using a larger spectrum of aromatic substrates for enrichment and isolation.  相似文献

Five male camels dwelling in the Algerian Sahara were studied for circadian rhythmicity in plasma aldosterone concentration and its seasonal modulation. Blood was sampled at a frequency of 1 h or less for a span of 27 h during each season of the year. The mean plasma aldosterone concentration exhibited a significant circadian rhythmicity in every season of the year. Plasma aldosterone concentration was lowest in the morning, increased in the afternoon, and generally highest in the late evening. The peak of the circadian rhythm exhibited seasonal variation; it occurred at 20:04h in October, 16:41h in December, 20:40h in March, and 24:16h in June. The rhythm's 24h mean also exhibited seasonal variability, being significantly higher in March and June compared to October.  相似文献

Superficial bladder cancer shows a high frequency of total or partial chromosome 9 losses. Loss of heterozygosity at position 9q22.3 is one of the most frequent and is associated with highly recurrent tumours. The PATCHED gene, ortholog of a gene first described in the drosophila as a segment polarity gene, is located at 9q22.3. It is a member of a signal transduction pathway and a tumour suppressor gene (TSG), involved in basal cell carcinoma. We propose PATCHED as a TSG candidate in superficial bladder cancer.  相似文献

The effects of Cd2+ and NaCl, applied together or separately, on growth and uptake of Cd2+ were determined for the halophyte Sesuvium portulacastrum L. Seedlings were cultivated in the presence of 50 or 100 μmol L−1 Cd2+ alone or combined with 100 or 400 mmol L−1 NaCl. Data showed that alone, Cd2+ induced chlorosis, necrosis, and inhibited growth. Addition of NaCl to Cd2+-containing medium restored growth and alleviated the toxicity, however. NaCl also enhanced the amounts of Cd2+ accumulated in the shoots. All Cd2+ treatment reduced K+ and Ca2+ uptake and transport to the shoots. Accumulation of Na+ in the shoots was not affected by Cd2+, however. Thus S. portulacastrum maintained its halophytic characteristics in the presence of Cd2+. We suggest this halophyte could be used for phytoextraction of Cd2+ from salt-contaminated sites.  相似文献

Obesity is associated with adipose tissue remodeling, characterized by adipocyte hypertrophy and macrophage infiltration. Previously, we have shown that very low density lipoprotein receptor (VLDLR) is virtually absent in preadipocytes but is strongly induced during adipogenesis and actively participates in adipocyte hypertrophy. In this study, we investigated the role of VLDLR in adipose tissue inflammation and adipocyte-macrophage interactions in wild type and VLDLR-deficient mice fed a high fat diet. The results show that VLDLR deficiency reduced high fat diet-induced inflammation and endoplasmic reticulum (ER) stress in adipose tissue in conjunction with reduced macrophage infiltration, especially those expressing pro-inflammatory markers. In adipocyte culture, VLDLR deficiency prevented adipocyte hypertrophy and strongly reduced VLDL-induced ER stress and inflammation. Likewise, cultures of primary peritoneal macrophages show that VLDLR deficiency reduced lipid accumulation and inflammation but did not alter chemotactic response of macrophages to adipocyte signals. Moreover, VLDLR deficiency tempered the synergistic inflammatory interactions between adipocytes and macrophages in a co-culture system. Collectively, these results show that VLDLR contributes to adipose tissue inflammation and mediates VLDL-induced lipid accumulation and induction of inflammation and ER stress in adipocytes and macrophages.  相似文献

The objective of this study was to analyze the immunogenicity and antigenicity of the V3 domain (Cys313–Cys346) of the external envelope glycoprotein gp125 of SIVmac251. The corresponding peptide was synthesized and characterized as linear and cyclic peptides. Our results showed that this region, as for HIV‐1, contained an immunodominant epitope. The antigenicity was similar for the linear and cyclic peptides when tested against a panel of 15 sera from SIV infected macaques. Similarly, both peptide structures presented similar immunogenicity as shown by the characterization of the anti‐peptide antibodies produced in rabbits against the cyclic and linear forms. But, unexpectedly, the antibodies produced against linear peptides recognized with a relatively higher intensity the native envelope gp140 than those produced against the cyclic structure. Furthermore, we showed that these antibodies recognized better the deglycosylated form of the glycoprotein. But, in contrast to the neutralizing activity obtained with anti‐V3 peptides from HIV‐1, no antiviral activity was obtained with antibodies generated against linear or cyclic SIVmac V3 peptides. Multimeric catalase from Aspergillus niger was immobilized on CNBr activated agarose, increasing the enzyme stability. However, it was found that some enzyme subunits could be desorbed to the supernatant after boiling the enzyme preparation in the presence of SDS or during thermal inactivation. Moreover, a positive enzyme concentration-enzyme stability correlation was detected in the immobilized preparation. This suggested the existence of some dissociation mechanism as a first step in the enzyme inactivation. The treatment of the immobilized enzyme with aldehyde–dextran permitted to fully stabilize its multimeric structure, but even this preparation exhibited an enzyme concentration-stability correlation. The presence of EDTA reduced the enzyme stability, suggesting that some cation could be involved in enzyme stability. It was found that 10 mM Zn2+ increased the enzyme stability of this immobilized–stabilized preparation. Now, the dilution of the biocatalyst did not produce a reduction in the enzyme stability.Thus, we have prepared an immobilized enzyme that does not release any subunit to the medium even after inactivation, and found that Zn2+ has a very positive effect on the stability of this immobilized–stabilized enzyme.  相似文献

Derivative spectrophotometry (graphical method) and partial least-squares regression (numerical method) methods were developed for the spectrophotometric multi-component analysis of post-haemodialysis fluids and synthetic mixtures containing Al(III) and Fe(III) without any chemical separation. The complexes of these metal ions with chrome azurol S were formed immediately at pH 5.5 and were stable for at least 3h. The graphical method is based on the use of first-derivative spectra for evaluation because working wavelength determination was more precise and spectral overlap was less than in the ordinary spectra. Two wavelengths at which the complexes exhibited maximum absorption values for Fe(III) and Al(III) were selected as analytical wavelengths, i.e., 675 and 623.5nm, respectively. Lambert-Beer's law is obeyed between 0.0896-8.064mug/mL Fe(III) and 0.054-0.486mug/mL Al(III). Limits of detection for Fe(III) and Al(III) were 0.056 and 0.044mug/mL, respectively. The reproducibility, expressed as variation coefficients, for two sets of 10 standard mixtures containing 3.584mug/mL Fe(III) and 0.27mug/mL Al(III) were 1.9% and 2% for iron and aluminium, respectively. In the numerical method, a training set was randomly prepared by using 14 samples. The concentration of each component has been varied in the linear range of the analytical signal. The spectral regions between 510 and 720nm were selected for the analysis of the binary mixture of Fe(III)/Al(III). The proposed methods were validated by using synthetic binary mixtures and applied to the simultaneous determination of Fe(III) and Al(III) in post-haemodialysis samples. The obtained results were compared with each other; in general, both multi-component methods gave rise to similar recovery results for laboratory-prepared mixtures and real samples.  相似文献
