Differentiation of embryonic stem cell (ESC)-derived embryoid bodies (EBs) is a heterogeneous process. ESCs can differentiate
in vitro into different cell types including beating cardiomyocytes. The main aim of the present study was to develop an improved
preparation method for scanning electron microscopic study of ESC-derived cardiac bundles and to investigate the fine structural
characteristics of mouse ESCs-derived cardiomyocytes using electron microscopy. The mouse ESCs differentiation was induced
by EBs’ development through hanging drop, suspension and plating stages. Cardiomyocytes appeared in the EBs’ outgrowth as
beating clusters that grew in size and formed thick branching bundles gradually. Cardiac bundles showed cross striation even
when they were observed under an inverted microscope. They showed a positive immunostaining for cardiac troponin I and α-actinin.
Transmission and scanning electron microscopy (TEM & SEM) were used to study the structural characteristics of ESC-derived
cardiomyocytes. Three weeks after plating, differentiated EBs showed a superficial layer of compact fibrous ECM that made
detailed observation of cardiac bundles impossible. We tried several preparation methods to remove unwanted cells and fibers,
and finally we revealed the branching bundles of cardiomyocytes. In TEM study, most cardiomyocytes showed parallel arrays
of myofibrils with a mature sarcomeric organization marked by H-bands, M-lines and numerous T-tubules. Cardiomyocytes were

The objective of this study is to determine the prevalence of expression and bilateralism of two dental morphological traits in contemporary Jordanians: The hypocone reduction trait on the maxillary second permanent molar and Carabelli's trait on maxillary permanent first and second molars. Furthermore, inter-trait correlation and the relationship of Carabelli's traits with upper first molar dimensions were investigated. Three hundred subjects of school children at their 10th grade and of an average age of 15.5 +/- 0.4 years were involved. Alginate impressions for the maxillary arch were taken, dental casts were reproduced. The selected accurate casts were of 132 male- and 155 female-students. The frequencies of hypocone reduction trait on the maxillary second molar and Carabelli's trait on the maxillary molars were examined. Buccolingual and mesiodistal diameters of the maxillary first molar were measured and recorded. Paired Sample t test and Nonparametric Correlation analysis were used for data analysis. Hypocone reduction trait on the maxillary second molar was found in 29.8% of the examined students. Positive forms of Carabelli's trait on first and second molars were observed in 65.0% and 3.8%, respectively. Nonparametric correlation analysis revealed positive association between Carabelli's trait on first molar and hypocone reduction trait on the maxillary second molar. The presence of Carabelli's trait on first molar was strongly associated with the increase of buccolingual, but not the mesiodistal, diameter. Bilateralism was found highly significant in the tested traits and both genders (p < 0.001). This finding might be a sign of relatively low environmental stresses in the living Jordanian population and/or great ability of its individuals to buffer the adverse effects of such stresses. 相似文献

Ascorbic acid (vitamin C) was given to 144-days-old layer chickens mixed with their ration at three concentrations: 30, 60 and 90 p.p.m. for 6 months. Cholesterol levels in blood serum (CS) and egg yolk (CE) were measured every 6 weeks (four periods); there was a marked decrease in CS in most treated birds, especially those receiving the highest concentration of vitamin C. There was also a slight decrease in CE in most treated birds. Epilepsy is a neurological disorder affecting more than 50 million people worldwide. It can be controlled by antiepileptic drugs
(AEDs) but more than 30% patients are still resistant to AEDs. To overcome this problem, researchers are trying to develop novel
approaches to treat epilepsy including the use of herbal medicines. The γ-amino butyric acid type-A receptor associated protein
(GABARAP) is ubiquitin-like modifier implicated in the intracellular trafficking of GABAAR. An in silico mutation was created at
116 amino acid position G116A, and an in silico study was carried out to identify the potential binding inhibitors (with antiepileptic
properties) against the active sites of GABARAP. Five different plant derived compounds namely (a) Aconitine (b) Berberine (c)
Montanine (d) Raubasine (e) Safranal were selected, and their quantitative structure-activity relationships (QSAR) have been
conducted to search the inhibitory activity of the selected compounds. The results have shown maximum number of hydrogen
bond (H-bond) interactions of Raubasine with highest interaction energy among all of the five compounds. So, Raubasine could be

The average inbreeding coefficients of the highly consanguineous Fur and Baggara tribes of Western Sudan were 0.04167 and 0.04450, respectively. Two hundred ninety-eight subjects from the two tribes were tested for polymorphism of hemoglobins, seven red cell enzymes, and four serum proteins. The Baggara showed a higher gene frequency of HbS and TfD and lower gene frequency of GdA and PC compared to the Fur. Both tribes showed a low gene frequency of PGM1 and high frequency of G6PD deficiency when compared to other Sudanese tribes. In spite of the high degree of inbreeding, no significant deviation from the Hardy-Weinburg equilibrium was observed in either tribe. The effects of inbreeding seem to be offset by mixing between the two tribes on Gabal Marra Plateau. The flow of the sickle gene from the Baggara into the Fur and other Sudanese tribes is discussed. 相似文献

We have determined the complete nucleotide sequence of the small- subunit
ribosomal RNA genes for the ciliate protozoans Stylonychia pustulata and
Oxytricha nova. The sequences are homologous and sufficiently similar that
these organisms must be closely related. In a phylogeny inferred from
comparisons of several eukaryotic small-subunit ribosomal RNAs, the
divergence of the ciliates from the eukaryotic line of descent is seen to
coincide with the radiation of the plants, the animals, and the fungi. This
radiation is preceded by the divergence of the slime mold, Dictyostelium
discoideum.

Molecular population genetics of ref(2)P, a locus which confers viral resistance in Drosophila 总被引:4,自引:0,他引:4
The ref(2)P locus (2-54.2) is polymorphic for two allelic forms in natural
populations of Drosophila melanogaster, ref(2)Po and ref(2)Pp. The latter
allele confers resistance to the rhabdovirus sigma infecting wild
populations. Previous work, based on a small sample of prescreened
restrictive (resistant) and permissive (susceptible) alleles, identified a
large number of amino acid replacement changes (7) relative to synonymous
changes (1). Such protein variability could be the result of
variation-enhancing selection. To further test the selection hypothesis, we
have examined the DNA sequences of ten randomly chosen lines of D.
melanogaster and one line of D. simulans. Nine of the ten lines are
permissive; D. simulans does not harbor the virus. The melanogaster alleles
contain 4 synonymous changes, 19 noncoding changes, and 13 amino acid
replacement changes, indicating a relatively high level of polymorphism.
Three sequenced restrictive alleles have nearly identical sequences,
indicating that they are relatively young. Compared to the permissive
alleles, they share only a complex deletion at codon 34, CAG-AAT to GGA,
which our analysis indicates to be the site conferring the restrictive
phenotype. Patterns of polymorphism and divergence differ from neutral
predictions by several criteria for the amino terminal region, which
contains the complex deletion (codons 1-91), but not the remainder of the
protein (codons 92-599). We find a higher rate of evolution on the D.
melanogaster lineage than on the D. simulans lineage. The relatively large
amount of both replacement and silent polymorphism in the permissive
alleles and the lack of divergence between permissive and restrictive
alleles suggests that the sigma virus and ref(2)P may be engaged in an
evolutionary race in which new restrictive alleles are continually arising
but are relatively short-lived.
