The duration of the cell cycle and its component phases in cell cultures of Haplopappus gracilis was estimated by means of pulse labelling with tritiated thymidine and subsequent autoradiographic techniques. The total duration of the mitotic cycle was found to be 22.0 hours. The average durations of the following component phases were: the synthetic period (S) 6.4 hours, the postsynthetic period (G2) 4.86 hours, prophase (P) 0.64 hours, metaphase (M) 0.40 hours, anaphase + early telophase (AT) 0.36 hours, the presynthetic period (G1) 9.34 hours. The results indicate that G1 and G2 are the phases, which are most prolonged in populations of cultivated cells when compared to the same phases in root lip cells from the same species.  相似文献

Plant regeneration ability of ginseng (t Panax ginseng C.A. Meyer) via organogenesis was studied. Compact callus was induced from four different types of explants-leaf, petiole, flower stalk, and root of t in vitro-grown plantlets. Petioles were found to be the best material for callus induction. Calli induced on Murashige and Skoog (MS) medium supplemented with 2,4-dichlorophenoxyacetic acid (4.5 μM) and kinetin (0.46 μM) were conditioned for two weeks on the same medium. These calli differentiated into adventitious shoots when cultured on 1/2MS basal medium plus kinetin 4.7 μM and silver thiosulphate 10 μM. An addition of GA3 (2.9 μM) and BA (4.4 μM) to MS basal medium, however, induced high frequency t in vitro flowering (86.1%) and multiple shoot budding which affected the normal complete development of plantlets. Plantlets with a well-developed root system were obtained six weeks after regenerated shoots had been transplanted to 1/2 MS20 medium containing IBA 1.2 μM. Nuclear DNA content was measured to check the stability of their ploidy level. Based on DNA flow cytometric analysis, all the regenerants were typically diploids as the mother plants were, indicating that nuclear DNA content remained stable during cell differentiation. Abstract: Because of a revival in the controversy surrounding 'aerobic denitrification', especially in relation to Thiosphaera pantotropha , activity in aerobic batch cultures was evaluated using gas chromatography and mass spectrometry after the addition of 15N-labelled NH4+ and NO2. Aerobic denitrifying activity in T. pantotropha was present, but only at about 10% of the originally-reported levels. The activity of ' Pseudomonas denitrificans ' was similar to previously-reported values. Alcaligenes faecalis showed significant aerobic denitrifying activity, producing almost equivalent amounts of N2 and N2O. An unidentified pseudomonad, isolate G4, presumably requires anoxia for enzyme activity as it did not denitrify aerobically, even though it has a constitutive denitrifying pathway.  相似文献

Background

Genomic selection can be implemented by a multi-step procedure, which requires a response variable and a statistical method. For pure-bred pigs, it was hypothesised that deregressed estimated breeding values (EBV) with the parent average removed as the response variable generate higher reliabilities of genomic breeding values than EBV, and that the normal, thick-tailed and mixture-distribution models yield similar reliabilities.

Methods

Reliabilities of genomic breeding values were estimated with EBV and deregressed EBV as response variables and under the three statistical methods, genomic BLUP, Bayesian Lasso and MIXTURE. The methods were examined by splitting data into a reference data set of 1375 genotyped animals that were performance tested before October 2008, and 536 genotyped validation animals that were performance tested after October 2008. The traits examined were daily gain and feed conversion ratio.

Results

Using deregressed EBV as the response variable yielded 18 to 39% higher reliabilities of the genomic breeding values than using EBV as the response variable. For daily gain, the increase in reliability due to deregression was significant and approximately 35%, whereas for feed conversion ratio it ranged between 18 and 39% and was significant only when MIXTURE was used. Genomic BLUP, Bayesian Lasso and MIXTURE had similar reliabilities.

Conclusions

Deregressed EBV is the preferred response variable, whereas the choice of statistical method is less critical for pure-bred pigs. The increase of 18 to 39% in reliability is worthwhile, since the reliabilities of the genomic breeding values directly affect the returns from genomic selection.  相似文献

Background: Climate warming has been causing an increase in tall shrub cover around the Arctic, however, mammalian herbivory has been shown to inhibit shrub expansion. Though the effect of reindeer (Rangifer tarandus) and many other mammals has been widely studied in this context, the role of the mountain hare (Lepus timidus) in subarctic Scandinavia remains unknown.

Aims: To quantify browsing from mountain hare and reindeer on tall shrubs in different vegetation types and to investigate differences in shrub preference between the two.

Methods: In the summers of 2013 and 2014, we counted signs of browsing by hare and reindeer on tall shrub species in 31 study plots at three alpine locations in the Scandes range, Sweden.

Results: Hare browsing was significantly more frequent than that by reindeer in two (dry-mesic heath and dry meadow) out of seven vegetation types studied. Reindeer browsing was significantly higher in the low herb meadow and Långfjället shrub heath. Two shrub species, Betula nana and Salix hastata, were significantly more browsed by hare, while reindeer browsing was significantly higher on S. phylicifolia and S. lapponum.

Conclusions: Our results show that mountain hares can cause extensive damage to tall shrubs in the subarctic and may have a stronger impact on shrub communities than previously recognised.  相似文献

Growth and differentiation in shoot cultures of five willow clones on media of different BA concentrations were compared. The tendency of axillary shoots to develop on shoot cultures depended on the genotype, the type of shoot and the number of previous subcultures. The optimum concentration for shoot multiplication was either 5×10-7 M or 10-6 M. On BA concentrations of 10-5 M or higher, browning and death of shoots occurred. Depending on the genotype, shoot elongation was best on media containing 0–5×10-7 M BA. Rooting ability was also genotype dependent. Prolonged culture in vitro improved the rooting ability of the two most reluctant clones. BA concentrations of 5×10-7 M or higher inhibited rooting almost completely, but this was not a permanent effect. All clones could be rooted on medium containing 10-6 M NAA. Shoots were transferred to greenhouse conditions and rooted with varying degrees of success depending on shoot size and genotype.  相似文献

Binding of [3H](R,S)-alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid ([3H]AMPA) to quisqualate receptors in the presence of SCN- ions produced curvilinear Scatchard plots. Kinetic investigations of [3H]AMPA binding showed that the curvilinearity cannot be explained by assuming binding to two separate binding sites or by considering it due to cooperative interaction. A more likely explanation is that the quisqualate receptors exist in two states, one with high and one with low affinity for [3H]AMPA. Chaotropic ions change the relaxation constant between the two states.  相似文献

An improved system involving a modification of the bead culture system was developed for culturing pea protoplasts. Using this method, sustained divisions and callus growth could be obtained in all 10 cultivars tested. In the best responding cultivar division frequency could be raised from 17% in liquid culture to 80% in the bead system. Binding of [3H]AMPA to rat brain membranes was investigated. The binding was saturable and reversible at physiological pH. Computer-aided Scatchard analysis of the binding data, as determined by using L-glutamic acid (L-GLU) to define nonspecific binding, suggested the presence of two independent binding sites, with KDS of 9 and 2440 nM, respectively. Additional freezing, thawing and washing sequences gave membranes with only one binding site, with a KD of 278 nM. [3H]AMPA binding exhibited the highest level in striatal membranes. A series of analogues of GLU and aspartic acid (ASP) were tested as inhibitors of [3H]AMPA binding. L-ASP and compounds which interact predominantly with N-methyl-D-aspartic acid (NMDA) receptor sites were inactive as inhibitors of [3H]AMPA binding, whereas L-GLU and compounds which interact predominantly with glutamic acid diethyl ester receptor sites were inhibitors with the same order of potency as that shown by the excitatory action in vivo. The result suggests that [3H]AMPA might represent binding to an excitatory GLU receptor.  相似文献

Summary Protoplasts of Panax ginseng were isolated from embryos obtained from the 4-year old embryogenic cell line KCTC PCL 49031 which was derived from a zygotic embryo. High protoplast yields of 22–25 × 106 protoplast / g tissue were obtained following 5–6 h digestion with 2% Cellulysin, 1% Pectinase and 1% Macerasae in half strength Murashige and Skoog's medium containing 12% mannitol. A plating density of 1×105 protoplasts /ml was found optimal for protoplast culture. An initial division frequency of 10% was obtained in an agarosegelled defined medium. Myo-inositol (6%) was found to be the most suitable osmoticum.
