Summary The evaluation of germplasm to identify its potential as a source of new favorable alleles is a time-consuming phase of maize hybrid breeding programs. The objective of this paper was to study the relationship between allozyme diversity and quantitative estimators of the relative number of new favorable alleles for grain yield, present in donor lines but not present in the elite hybrid. Twenty-two maize inbred lines representing heterotic groups from the United States (US) and Yugoslavia (YU) were used as donors to estimate the presence of new favorable alleles for grain yield improvement for the hybrid B73 x Mo17. In a second experiment, a 15-line diallel was grown, and 13 single crosses differing in allozyme relatedness measure (ARM) and heterotic grouping were considered as targets to be improved by the remaining 13 lines. Minimally biased estimates of new favorable alleles for grain yield (G) and ARM values were made for all donor lines within each target hybrid. Donor lines were grouped in four allozyme-pedigree classes for each target hybrid to compare the effect of allozyme diversity with pedigree diversity. Pedigree dissimilarities had significant effects on G estimates. Dissimilar pedigree classes had higher G estimates than similar pedigree classes. Allozyme differences between donor inbred lines and target hybrids had inconsistent effects on G estimates. Significant differences in G estimates among allozyme classes were found for 31% of the target hybrids. Classes with similar allozymes had higher G estimates more frequently than classes with disimilar allozymes. Considerable epidemiological evidence exists to link thyroid disease with differing patterns of dietary consumption, in particular, cruciferous vegetables. We have been studying the anti-thyroid cancer (TCa) activity of indole-3-carbinol (I3C) found in cruciferous vegetables and its acid catalyzed dimer, 3,3'-diindolylmethane (DIM). There are no studies as yet to elucidate the effect of these compounds on the altered proliferative patterns in goiter or thyroid neoplasia. In this study, we tested the anti-proliferative effects of I3C and DIM on four different thyroid cancer cell lines representative of papillary (B-CPAP and 8505-C) and follicular carcinoma of the thyroid (CGTH-W-1 and ML-1), and primary human goiter cells. Cell survival and IC50 values for I3C and DIM were calculated by the XTT assay and cell cycle distribution analysis was done by flow cytometry. DIM was found to be a better anti-proliferative agent than I3C in both papillary and follicular TCa resulting in a greater cytotoxic effect at a concentration over three fold lower than predicted by the molar ratio of DIM and I3C. The anti-proliferative activity of DIM in follicular TCa was mediated by a G1 arrest followed by induction of apoptosis. DIM also inhibited the growth of primary goiter cells by 70% compared to untreated controls. Contrary to traditional belief that cruciferous vegetables are "goitrogenic", DIM has anti-proliferative effects in glandular thyroid proliferative disease. Our preclinical studies provide a strong rationale for the clinical exploration of DIM as an adjuvant to surgery in thyroid proliferative disease.  相似文献

Purpose

To report piloting and initial validation of the VQoL_CYP, a novel age-appropriate vision-related quality of life (VQoL) instrument for self-reporting by children with visual impairment (VI).

Methods

Participants were a random patient sample of children with VI aged 10–15 years. 69 patients, drawn from patient databases at Great Ormond Street Hospital and Moorfields Eye Hospital, United Kingdom, participated in piloting of the draft 47-item VQoL instrument, which enabled preliminary item reduction. Subsequent administration of the instrument, alongside functional vision (FV) and generic health-related quality of life (HRQoL) self-report measures, to 101 children with VI comprising a nationally representative sample enabled further item reduction and evaluation of psychometric properties using Rasch analysis. Construct validity was assessed through Pearson correlation coefficients.

Results

Item reduction through piloting (8 items removed for skewness and individual item response pattern) and validation (1 item removed for skewness and 3 for misfit in Rasch) produced a 35-item scale, with fit values within acceptable limits, no notable differential item functioning, good measurement precision, ordered response categories and acceptable targeting in Rasch. The VQoL_CYP showed good construct validity, correlating strongly with HRQoL scores, moderately with FV scores but not with acuity.

Conclusions

Robust child-appropriate self-report VQoL measures for children with VI are necessary for understanding the broader impacts of living with a visual disability, distinguishing these from limited functioning per se. Future planned use in larger patient samples will allow further psychometric development of the VQoL_CYP as an adjunct to objective outcomes assessment.  相似文献

To investigate the characteristics of motor, sensory and sensory-evoked potentials (SEPs) of thoracic and lumbar roots, and demonstrate the feasibility of assessing axonal regrowth after the neurotization procedure in a sheep model. Six adult sheep were anaesthetized and placed in a sternal position. The thoracic and lumbar roots from T11 to L5 were identified at their emergence from the vertebral foramen and stimulated. Motor and sensory responses were monitored. Thoracic and lumbar roots were easily identified in all cases. Motor potentials were detected for each stimulated nerve without difficulty. The amplitudes were quite variable, ranging from 100 to 5300 microV. Sensory and SEPs were satisfactorily recorded in only three of the six animals. Sensory amplitudes also varied greatly, ranging from 25 to 120 microV. In three cases, SEPs could not be identified due to motor artefacts. The motor pathway after axonal regrowth in neurotized lumbar roots might easily be explored by proximal electric stimulation of the root, close to the sutured area. Detection of sensory and spinal cord evoked potentials might be improved by the use of curve summation techniques. Specific axonal tracing holds promise of being a useful technique for examining sensory and motor pathway recovery after neurotization in the sheep model.  相似文献

Objective

Tonic-clonic activity (TCA) at onset complicates 3% to 21% of cases of subarachnoid hemorrhage (SAH). The impact of onset TCA on in-hospital complications, including seizures, remains unclear. One study associated onset TCA with poor clinical outcome at 6 weeks after SAH, but to our knowledge no other studies have confirmed this relationship. This study aims to assess the impact of onset TCA on in-hospital complications, poor functional outcome, mortality, and epilepsy at 3 months.

Methods

Analysis of a prospective study cohort of 1479 SAH patients admitted to Columbia University Medical Center between 1996 and 2012. TCA within 6 hours of hemorrhage onset was identified based on accounts of emergency care providers or family witnesses.

Results

TCA at onset was described in 170 patients (11%). Patients with onset TCA were younger (P = 0.002), presented more often with poor clinical grade (55% vs. 26%, P<0.001) and had larger amounts of cisternal, intraventricular, and intracerebral blood than those without onset TCA (all, P<0.001). After adjusting for known confounders, onset TCA was significantly associated with in-hospital seizures (OR 3.80, 95%-CI: 2.43–5.96, P<0.001), in-hospital pneumonia (OR 1.56, 95%-CI: 1.06–2.31, p = 0.02), and delayed cerebral ischemia (OR 1.77, 95%-CI: 1.21–2.58, P = 0.003). At 3 months, however, onset TCA was not associated with poor functional outcome, mortality, and epilepsy after adjusting for age, admission clinical grade, and cisternal blood volume.

Conclusions

Onset TCA is not a rare event as it complicates 11% of cases of SAH. New and clinically relevant findings are the association of onset TCA with in-hospital seizures, pneumonia and delayed cerebral ischemia. Despite the increased risk of in-hospital complications, onset TCA is not associated with disability, mortality, and epilepsy at 3 months.  相似文献

Rhabdoviridae, characterized by bullet-shaped viruses, is known for its diverse host range, which includes plants, arthropods, fishes and humans. Understanding the viral–host interactions of this family can prove beneficial in developing effective therapeutic strategies. The host proteins interacting with animal rhabdoviruses have been reviewed in this report. Several important host proteins commonly interacting with animal rhabdoviruses are being reported, some of which, interestingly, have molecular features, which can serve as potential antiviral targets. This review not only provides the generalized importance of the functions of animal rhabdovirus-associated host proteins for the first time but also compares them among the two most studied viruses, i.e. Rabies virus (RV) and Vesicular Stomatitis virus (VSV). The comparative data can be used for studying emerging viruses such as Chandipura virus (CHPV) and the lesser studied viruses such as Piry virus (PIRYV) and Isfahan virus (ISFV) of the Rhabdoviridae family.  相似文献

Sexual selection molds the morphology, physiology and behavior of males in many animals. At first glance, it seems reasonable to assume that females would use the same male traits and signals in mate choice as males do during male-male competition. However, intra- and intersexual competition may affect traits in the same or the opposite direction, with differing strength. We investigated which color, morphometric and performance traits are selected for through male-male competition and whether female mate preference is based on these same traits and/or dominance status in the three male color morphs of the lizard Podarcis melisellensis. Males with relatively bigger heads and relatively higher bite forces were more likely to win fights and orange males were always dominant over the other morphs. Females, however, preferred scents of bigger males that were in better body condition, and surprisingly had lower bite force capacities. They did not show a preference for scents of any particular color morph or for scents of the more dominant males. These results indicate that intra- and intersexual competition may result in selection for different secondary sexual traits in P. melisellensis.  相似文献

We describe polymerase chain reaction primers and amplification conditions for 13 highly polymorphic microsatellite DNA loci isolated from the Dalmatian wall lizard, Podarcis melisellensis. The number of alleles per locus ranged from 12 to 41, with levels of observed heterozygosity between 0.62 and 0.94. Most of these loci were successfully cross-amplified in the closely related species P. sicula, but levels of polymorphism were always lower.  相似文献

The receptor for advanced glycation end products (RAGE) plays an important role in host defense against bacterial infection. In the present experiments, we investigated the mechanisms by which RAGE contributes to the ability of neutrophils to eradicate bacteria. Wild-type (RAGE(+/+)) neutrophils demonstrated significantly greater ability to kill Escherichia coli compared with RAGE(-/-) neutrophils. After intraperitoneal injection of E. coli, increased numbers of bacteria were found in the peritoneal fluid from RAGE(-/-) as compared with RAGE(+/+) mice. Exposure of neutrophils to the protypical RAGE ligand AGE resulted in activation of nicotinamide adenine dinucleotide phosphate (NADPH) oxidase and enhanced killing of E. coli, and intraperitoneal injection of AGE enhanced bacterial clearance during peritonitis. However, incubation of neutrophils with high mobility group box 1 protein (HMGB1), which also binds to RAGE, diminished E. coli-induced activation of NADPH oxidase in neutrophils and bacterial killing both in vitro and in vivo. Deletion of the COOH-terminal tail of HMGB1, a region necessary for binding to RAGE, abrogated the ability of HMGB1 to inhibit bacterial killing. Incubation of neutrophils with HMGB1 diminished bacterial or AGE-dependent activation of NADPH oxidase. The increase in phosphorylation of the p40(phox) subunit of NADPH oxidase that occurred after culture of neutrophils with E. coli was inhibited by exposure of the cells to HMGB1. These results showing that HMGB1, through RAGE-dependent mechanisms, diminishes bacterial killing by neutrophils as well as NADPH oxidase activation provide a novel mechanism by which HMGB1 can potentiate sepsis-associated organ dysfunction and mortality.  相似文献

We investigated the effect of low temperature and gibberellic acid (GA3) treatment on dormancy in Fritillaria meleagris L. bulbs. Also, we studied the effect of dormancy breaking on the antioxidant enzymes activity. To overcome dormancy, bulbs require a period (4–8 weeks) of exposure to low temperature. Bulbs regenerated in vitro were grown in the dark on medium without growth regulators at the standard (24 °C) or at low temperatures (4 and 15 °C) for 4, 6, 8 and 10 weeks. Bulbs were collected after 3, 4 and 5 weeks of cooling at 4 °C. To investigate the influence of GA3 on dormancy, bulbs were treated for 24 h with GA3 solutions with 1, 2 and 3 mg l?1 concentrations. During the period of growth of bulbs at 4 °C, regeneration of bulbs was very weak, while at 15 °C the number of regenerated bulbs increased significantly. Improved bulb sprouting was achieved by a short treatment with gibberellin. Low temperature also represents a kind of oxidative stress for the plant. The activity of superoxide dismutase, catalase (CAT) and peroxidase (POX) in bulbs of F. meleagris L. grown in vitro and ex vitro increased with decreasing temperature in contrast to glutathione reductase. POX showed generally lower activity than CAT which indicates that major role in the breaking dormancy and preparing bulbs for sprouting have catalases.  相似文献
