Reversible binding of DIDS [4,4'-diisothiocyanato-2,2'-stilbenedisulphonate] to Band 3 protein, the anion exchanger located in erythrocyte plasma membrane, was studied in human erythrocytes. For this purpose, the tritiated form of DIDS ([3H]DIDS) has been synthesized and the filtering technique has been used to follow the kinetics of DIDS binding to the sites on Band 3 protein. The obtained results showed monophasic kinetics both for dissociation and association of the 'DIDS-Band 3' complex at 0° C in the presence of 165 mM KCl outside the cell (pH 7.3). A transient CO2 burst from seedlings of some plant species was observed after a rapid temperature decrease. The magnitude of the CO2 release depended on initial temperature, oxygen concentration and light intensity. To obtain a maximal value of CO2 release, the temperature had to decrease by more than 8°C. The phenomenon was detected only in the light, and was confined to C3 species. It was inhibited by low oxygen concentration, indicating its possible connection with photorespiration.  相似文献

The hypothesis that peripheral, skeletal muscle tissue contains a trophic factor supporting central neurons has recently been investigated in vitro by supplementing the culture medium of spinal cord neurons with muscle extracts and fractions of extract. We extended these studies asking whether or not a trophic factor is present in peripheral nerves, the connecting link between muscle and central neurons via which factors may be translocated from muscle to neurons by the retrograde transport system. Lumbar, 8-day-old chick spinal cords were dissociated into single cells and then cultured in the presence of peripheral nerve extract. Cytosine arabinoside was added to inhibit proliferation of nonneuronal cells. In the presence of nerve extract, spinal cord neurons survived for more than a month, extended numerous neurites, and showed activity of choline acetyltransferase. In the absence of extract, neurons attached and survived for a few days but then died subsequently in less than 10 days. Neurite outgrowth did not occur in the absence of extract. Withdrawal of extract from the medium of established neuronal cultures caused progressive loss of both cells and neurites. Other tissues also contained neuron supporting activity but less than that found in nerve extract. These studies indicate that peripheral nerves contain relatively high levels of spinal cord neuron-directed trophic activity, suggesting translocation of neurotrophic factor from muscle to central target neurons. The neurotrophic factor has long-term (weeks) effects, whereas short-term (days) survival is factor independent.  相似文献

Ca2+-dependent influence of excess Cu2+ on the photosynthetic akpparatus monitored through chlorophyll fluorescence measurements was investigated in runner bean plants (Phaseolus coccineus L. cv. Pie kny Ja?) at three different growth stages. It was observed that the toxic effect of excess Cu2+ on plants depends both on their growth stages and the Ca2+ content in the medium. Increased Ca2+ content limits Cu2+ action on plants at their initial growth stage (I) through: stabilization of the PSII complex (increase of the ratio of variable to minimal fluorescence [Fv/F0]), improved electron flow and reoxidative processes of the quinone primary electron acceptor of PSII (QA) (increase of quantum yield of PSII electron transport [φe] and photochemical quenching of fluorescence [qP] values) and elimination of nonphotochemical energy dissipation (decrease of nonphotochemical fluorescence quenching from the Stern-Volmer equation [NPQ] and fraction of the absorbed light energy not used for photochemistry [LNU] values). At this growth stage excess Cu2+ decreases the rates of QA reduction as a result of decreased PSII activity at its donor side only at lower Ca2+ level. At the intermediate growth stage (II) the plants were less sensitive to Cu2+ treatment and also to changed Ca2+ content. A weakening of some photochemical processes by excess Cu2+ could be observed only at a higher Ca2+ dose. At the final growth stage of plants (III) Ca2+ ions exerted a decisively different effect on the mechanism of excess Cu2+ action on bean plants, visualized by decreased PSII stabilization and utilization of absorbed light energy at increased Ca2+ content in the medium.  相似文献

Seasonal and longitudinal metabolic trends in benthic communities were studied in the five orders of a natural mountain river (the Stradomka) in southern Poland. The method of oxygen change of rocky substrata in chambers was applied. Metabolic data were compared with abiotic (light regime, channel geomorphology and stream hydrology), and water chemical variables. A tendency of downstream increase in gross production and community respiration was observed between 1st and 4th stream orders. The highest values of GP (0.94 g C m-2 24 h -1) and CR (1.12 g C m-2 24 h -1) were observed in the middle Stradomka course (3–4 orders) in spring. A distinct negative NDM (heterotrophy) of the benthic community was measured in autumn and spring. During winter NDM was almost constant and oscillated around zero. The P/R ratio changed according to season and fluctuated between heterotrophy (P/R < 1) and autotrophy (P/R > 1). In general, heterotrophy prevailed in the study system (except 5th order). A marked shift in P/R between 4th and 5th stream orders confirms the prediction included in the RCC. Water temperature and PAR accounted for 56% in GP variability, whereas CR and NDM were not related to these variables. The combined variables (PAR and PO4–P) attributed to 69% in GP variability. The set of chemical factors was less related to Stradomka metabolic data. These results suggest that metabolic processes in this mountain river remarkably depend on physical variables (solar radiation, altitude, water temperature and conductivity). Metabolic parameters were similar to other geographical mountain systems. This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

Pathogens are one of the main forces driving the evolution and maintenance of the highly polymorphic genes of the vertebrate major histocompatibility complex (MHC). Although MHC proteins are crucial in pathogen recognition, it is still poorly understood how pathogen‐mediated selection promotes and maintains MHC diversity, and especially so in host species with highly duplicated MHC genes. Sedge warblers (Acrocephalus schoenobaenus) have highly duplicated MHC genes, and using data from high‐throughput MHC genotyping, we were able to investigate to what extent avian malaria parasites explain temporal MHC class I supertype fluctuations in a long‐term study population. We investigated infection status and infection intensities of two different strains of Haemoproteus, that is avian malaria parasites that are known to have significant fitness consequences in sedge warblers. We found that prevalence of avian malaria in carriers of specific MHC class I supertypes was a significant predictor of their frequency changes between years. This finding suggests that avian malaria infections partly drive the temporal fluctuations of the MHC class I supertypes. Furthermore, we found that individuals with a large number of different supertypes had higher resistance to avian malaria, but there was no evidence for an optimal MHC class I diversity. Thus, the two studied malaria parasite strains appear to select for a high MHC class I supertype diversity. Such selection may explain the maintenance of the extremely high number of MHC class I gene copies in sedge warblers and possibly also in other passerines where avian malaria is a common disease.  相似文献
