Sieve tubes are comprised of sieve elements, enucleated cells that are incapable of RNA and protein synthesis. The proteins in sieve elements are supplied from the neighboring companion cells through plasmodesmata. In rice plants, it was unclear whether or not all proteins produced in companion cells had the same distribution pattern in the sieve element-companion cell complex. In this study, the distribution pattern of four proteins, beta-glucuronidase (GUS), green fluorescent protein (GFP), thioredoxin h (TRXh) and glutathione S-transferase (GST) were analyzed. The foreign proteins GUS and GFP were expressed in transgenic rice plants under the control of the TRXh gene promoter (PTRXh), a companion cell-specific promoter. Analysis of leaf cross-sections of PTRXh-GUS and PTRXh-GFP plants indicated high accumulation of GUS and GFP, respectively, in companion cells rather than in sieve elements. GUS and GFP were also detected in phloem sap collected from leaf sheaths of the transgenic rice plants, suggesting these proteins could enter sieve elements. Relative amounts of GFP and endogenous phloem proteins, TRXh and GST, in phloem sap and total leaf extracts were compared. Compared to TRXh and GST, GFP content was higher in total leaf extracts, but lower in phloem sap, suggesting that GFP accumulated mainly in companion cells rather than in sieve elements. On the other hand, TRXh and GST appeared to accumulate in sieve elements rather than in companion cells. These results indicate the evidence for differential distribution of proteins between sieve elements and companion cells in rice plants.  相似文献

Bacteroids retaining high acetylene reduction activity (nitrogenase activity) were prepared anaerobically from soybean nodules. Addition of succinate (or of both leghemoglobin and succinate) to the acetylene reduction assay system greatly increased the activity of the isolated bacteroids.

When various organic acids were incubated with the bacteroids at 2% oxygen concentration, an optimum condition for bacteroid acetylene reduction, the organic acid degradation by bacteroids was very slow, and both lactate and acetate were accumulated in the incubation system, suggesting the operation of fermentative pathway in bacteroids under such low oxygen conditions.

With 20% oxygen, the added organic acids were degraded rapidly by bacteroids without addition of leghemoglobin to the incubation system.

With leghemoglobin in the incubation system, the organic acid degradation by bacteroids was accelerated extensively even at 2% oxygen, and the formation of lactate and acetate were negligible. No significant difference in the organic acid degradation rate was observed between the 2% and 20% oxygen concentrations when the leghemoglobin was present in the incubation system. Addition of acetylene to the assay system slightly inhibited the organic acid degradation.

This data suggests that bacteroids are unable to oxidize organic acid in low oxygen concentration and that the leghemoglobin allows the rapid organic acid dagradation by bacteroids even in such low oxygen concentrations.  相似文献

From our research of nonsecosteroidal vitamin D3 derivatives with gamma hydroxy carboxylic acid, we identified compound 6, with two CF3 groups in the side chain, as a most potent vitamin D receptor (VDR) agonist that shows superagonistic activity in VDRE reporter gene assay, MG-63 osteocalcin production assay and HL-60 cell differentiation assay. Compound 6 demonstrated that fluorination is as effective in the case of our nonsecosteroidal scaffold as in the case of secosteroidal VD3 analogs. X-ray analysis of the VDR with compound 6 revealed all of the six fluorine atoms of the hexafluoropropanol (HFP) moiety in the side chain effectively interacting with the VDR by both steric (van der Waals) and electrostatic (hydrogen bond, NH–F and CH–F) interactions. The HFP moiety of 6 effectively interacts with helix 12 (H12) of the VDR and stabilizes the position and the orientation of H12, which could result in stabilizing the coactivator and enhancing the VDR agonistic activity.  相似文献

Illumination-induced greening in dark-grown plants is one of the most dramatic developmental processes known in plants. In our current study, we characterized the greening process of rice seedlings using comparative proteome analysis. We identified 886 different proteins in both whole cell lysates of illuminated and nonilluminated rice shoots and performed comparative proteome analysis based on the MS spectral intensities obtained for unique peptides from respective proteins. Furthermore, the changes in the levels of individual proteins were then compared with those of the corresponding mRNAs. The results revealed well-coordinated increases in the enzymes involved in the Calvin cycle at both the protein and mRNA levels during greening, and that the changes at the mRNA level precede those at the protein level. Although a much lower effect of illumination was found on the enzymes associated with glycolysis and the TCA cycle, coordinated increases during greening were evident for the enzymes involved in photorespiration and nitrogen assimilation as well as the components of the chloroplastic translational machinery. These results thus define the differential regulation of distinct biological systems during greening in rice and demonstrate the usefulness of comprehensive and comparative proteome analysis for the characterization of biological processes in plant cells.  相似文献

The influence of P deficiency on the utilization of two sources of N, mineral N (exogenous N) and reserved N (endogenous N), for regrowth of Italian ryegrass (Lolium multiflorum) was studied. P-sufficient (+P) or P-free (−P) nutrition solution was applied from 7 days before defoliation to 24 days of regrowth and the N flows derived from two different N sources within the plant were quantified by 15N pulse-chase labeling. Shoot regrowth significantly reduced by 12 days of regrowth, while root growth was more in −P plants. Inorganic P (Pi) concentration was highly reduced by P deprivation more in the stubble and regrowing shoots and less in the roots. At defoliation, P deprivation had induced a higher accumulation for all N compounds in the stubble and for amino acids in the roots. The previously incorporated 15N in stubble and roots as nitrate and amino acids was much decreased in −P plants especially for the first 6 days of regrowth. Total N content in the regrowing leaves was not significantly different between +P and −P plants, but percentage contribution of remobilized N for total leaf N formation was significantly higher in −P plants (78%) than in +P plants (69%) at 6 days of regrowth. From day 12, the utilization of both endogenous and exogenous N was apparently inhibited in −P plants.  相似文献

1. 1. As part of “research on environmental comfort,” that is, research which aims to make people's living environment more comfortable, we attempted to clarify the relation between the living environment of elderly people and physiological and psychological factors. We carried out a comprehensive study with a view to establishing comfort standards for the residential thermal environment, and for creating evaluation and control systems.

2. 2. The comfort of the living environment is closely related to the thermal environment and the temperature-regulation ability of the human body. This ability of the body to regulate temperature develops during childhood and recedes as the adult ages.

3. 3. We therefore carried out: (a) experiments on body-temperature regulation and on special characteristics of the body-temperature regulation of elderly people, (b) a nationwide survey of the actual residential thermal environments of elderly people and (c) a survey of the daily activities of elderly people, and how elderly people's physiological and psychological conditions change when they are engaged in these activities.

4. 4. As a result of these experiments and surveys, we were able to: (d) formulate standards for evaluation of the residential thermal environment and (e) numerically express the results of systematic evaluation of residential thermal environments of elderly people, by means of a RTE-index.

Mast cell development from spleen cells was not triggered by prostaglandin E1 (PGE1) or dibutyryl cAMP (db-cAMP) during a 12 day culture when the spleen cells were obtained from C57BL/6N and DBA/1 mice, but mast cells did develop when the spleen cells were obtained from C3H/HeN, BALB/c and ICR mice. A lack of endogenous IFN-gamma in the initial 2 days of the culture period was responsible for the failure. This was confirmed by adding neutralizing anti-IFN-gamma antibody and rIFN-gamma to the cultures and by determining IFN-gamma levels in the spleen cell cultures. Th1 cells in the spleens of C57Bl and DBA/1 mice were much more sensitive to PGE1 and db-cAMP than Th1 cells from other inbred mice strains, and consequently, IFN-gamma production was inhibited in spleen cell cultures of C57BL and DBA/1 mice on addition of PGE1 or db-cAMP. Furthermore, the different sensitivities of Th1 cells to PGE and db-cAMP were dependent on the different levels of IL-12 p40 monomers or homodimers in the spleen cell cultures. As the endogenous specific inhibitors of IL-12 p70 (heterodimers of p40 and p35), large amounts of IL-12 p40 monomers or homodimers in the spleen cell cultures of C57BL and DBA/1 mice enhanced the ability of PGE1 and db-cAMP to inhibit IFN-gamma production by antagonizing the activity of IL-12 heterodimers. These results indicate that the strain-dependent development of mast cells from mouse splenocytes is related to endogenous IFN-gamma levels, which are regulated by PGE, db-cAMP, IL-12 p70 and IL-12 p40.  相似文献

In higher plants, the supply of metals such as Zn and Fe via phloem is important for the growth and physiology of young organs. However, little information is available on the speciation (chemical forms) of these metals in the phloem fluids. Because the pH of phloem fluids is slightly alkaline and the concentration of phosphate, which may bind to metals, is high, Zn and Fe in phloem fluids could be precipitated if these metals do not form complexes with some ligand compounds. In the present experiment, we examined the chemical forms of Zn and Fe in phloem sap collected from rice (Oryza sativa L.) by separating the phloem sap using size-exclusion and anion-exchange chromatography, and identifying the contents using electrospray ionization time-of-flight mass spectrometry. The low molecular weight chemical forms of Zn and Fe were identified as Zn-nicotianamine and Fe(III)-2'-deoxymugineic acid complexes, respectively. This report is the first to identify metal-chelate complexes in rice phloem sap.  相似文献

1. 1. 10 elderly and 10 college-aged females served as subjects in cold and heat environments. The subjects changed into the standard clothing (0.63 clo), and stayed in the neutral environment (25°C) for 23 min, thereafter they were exposed to the cold (10°C) or hot (35°C) environment for 49 min.

2. 2. Then they returned to the neutral environment, and stayed there for 47 min. Oral temperature, skin temperatures at 10 sites, blood pressure and thermal sensation were measured during the experiments.

3. 3. In the cold environment, the elderly could not reduce heat loss by vasoconstriction as did young people, and their blood pressures increased more rapidly than in young people. In the hot environment, the elderly could not promote heat loss by vasodilation as did young people. Moreover, there is a delayed sensitivity to cold for the elderly. Therefore, in the houses of the elderly, it is important to have heating and cooling systems which also includes the areas where the people do not stay for a long period of time (e.g. toilet, passageways).
