Rice plants (Oryzae sativa L.) grown in a nutrient solutionwere fed with (15NH4)2SO4 during the 5 days of their young panicleformation. At the end of that time in the youngest leaf blade, which hadstarted to emerge during the labelling, absorbed-nitrogen accountedfor 37% of the increased nitrogen of the tissue; in the nextdeveloping leaf blade it accounted for 55%. Thus, remobilized-nitrogenoriginating from older patrs of the plant made up 63 and 45%,respectively, of their total nitrogen. The important contributionof the remobilized-nitrogen to the development of a leaf isevident. The remobilization of nitrogen in the 12th leaf blade on themain stem was examined in detail after labelling during itsdeveloping stage. The 15N level started to decrease soon afterthe end of the labelling period and continued to decrease untilfull senescence, although the total nitrogen in the same leafincreased until just after its complete expansion, suggestingthat even a young leaf plays a role as a supplier of remobilized-nitrogen. During the rapid decrease in the total nitrogen after its peakat full expansion of the leaf, the actual proportion of labelledabsorbed nitrogen remained nearly the same, indicating thatinflux of new nitrogen into a senescing leaf is very limited. Summary We transformed B. subtilis 168 with DNA from B. subtilis IAM1231, IAM1192 and ATCC6633. When we examined the restriction activities of the transformants in vivo and in vitro using phage 105C we found the following: (1) Cells of either IAM1231 or IAM1192 have two modification and restriction systems (Bsu1231(1)-system and Bsu1231(II)-system in IAM1231, and Bsu1192(I)-system and Bsu1192(II)-systems in IAM1192), and cells of ATCC6633 have only one system (Bsu6633-system). (2) The restriction enzymes of all of these five systems are site-specific endonucleases. (3) The nucleotide sequence specificities of the enzymes involved in Bsu1231(I)-system, Bsu1192(I)-system and Bsu6633-system are the same; and those of Bsu1231(II)-system and Bsu1192(II)-system are the same. The sequence specificities of these two groups are different from each other and also different from those of the Bsu168-system of B. subtilis 168, the BsuR-system of B. subtilis R and the Bsu1247(I)-and Bsu1247(II)-systems which are systems of B. subtilis IAM1247. (4) Transformants possessing four different modification and restriction systems (Bsu1231(I)-, Bsu1247(I)-, BsuR- and Bsu168-systems) were constructed. (5) Transformation of two derivatives of 168 that were m R + r R + by DNA from IAM1231 produced 16 transformants that had the Bsu1231(II) restriction system, but had lost the BsuR system. Transformation of a derivative of 168 that was m 1247(II) + r 1247(II) + by DNA from m 1231(II) + r 1231(II) + -or m R + r R + -derivative of 168 produced about 100 each of transformants that had the Bsu1231(II)-restriction system or the BsuR-restriction system. But all these transformants lost the Bsu1247(II)-system.  相似文献

Contact guidance was studied in cultures of chick heart fibroblasts and kidney epithelium by observing the relation of these cells to fine grooves ruled in plastic culture dishes, and also to ridges or grooves in plastic replicas moulded from rulings made in metal. The relation of the cells to the regularly arranged collagen fibers of fish scales was also studied by scanning and transmission electron microscopy (SEM and TEM). On the rulings with groove periodicity in the range of 5 μm about 75% of the cells were aligned, but on grooves separated about 30 μm only 60% of cells were aligned. Cytoplasmic components of the cells such as microfilaments maintained a constant relation to the axis of the cell as a whole, but they, and also any cytoplasmic extensions, such as filopodia, bore no consistent relation to any features of the substratum, whether or not the cells were aligned. The cells were not guided to become aligned by filopodia or lamellipodia. The most remarkable and consistent finding was that cells bridged over grooves without contacting their surfaces, whether the grooves were 2 or 10 μm wide. The bridging was a characteristic of cells growing on any of the substrates, including those with grooves or ridges, and also of collagen substrates made from fish scales. A hypothesis is proposed to explain the contact guidance seen on ridged or grooved substrata and on the orientated collagen fibers involving the observed cell bridging and the fact that linear cell-to-substrate contacts (focal contacts) are known to be vital for cell movement. The cell is considered to be stiff so that as it bridges over much of the substratum there is only a limited area available for contact. Assuming that focal contacts need to be of a certain length to provide adhesion, a cell orientation that presents the maximum linear contact would be favoured. An examination of the results of this study and of the reports in the literature shows that cells on these types of substrata take on an orientation such that linear contacts would be expected to predominate.  相似文献

Responses to heat and exercise were studied in 9 male Japanese subjects who walked on a treadmill at a speed of 4.4 – 4.8 km/h at 0 grade for 2 hours in a climatic chamber in July 1973, in Nagoya Japan. The results were compared with those obtained in a similar study made in July 1966 in Cincinnati, Ohio. The following results were obtained: (1) Japanese showed a 1.8 times higher rate of sweating than Caucasians. Total sweat from the whole body during 2 hours walk was also higher in Japanese. (2) Japanese exhibited lower chloride concentration in local sweat than Caucasians in spite of their higher dietary salt intake, higher serum chloride concentration and higher rate of sweating. While in Caucasians the sweat chloride concentration showed a tendency to continue to rise during the later period of the walk in spite of decreasing sweat rate after sweat suppression occurred, in Japanese it tended to fall in parallel with the sweat rate. No difference was observed in the length of the latent time of sweat suppression. (3) There were no differences in rectal temperature or heart rate, both at the period of equilibrium rectal temperature and at the end of the walk. (4) Mean skin temperature during the walk was significantly higher in Japanese than in Caucasians. It was concluded that the Japanese group was better heat acclimatized than Caucasians, though the two groups were considered to have been naturally heat exposed by season to the same extent.  相似文献

An enzyme activity specific for UV-DNA1 was found in the extract of Bacillus subtilis(Marburg 168). The enzyme preparation obtained from the extract by ammonium sulfate precipitation acts on UV-DNA endonucleolytically and induces single strand breaks. Genetic data suggesting the absence of crossover in Saccharomycodes ludwigii have been described. Tetrad data obtained from 888 asci from 60 pairs of genes with 22 genetic markers showed the absence of tetratype asci, except for 5 asci in which a single pair of alleles showed tetratype segregation to the other genetic markers in each ascus. Spore arrays in the linear asci showed that the + - + - and + - - + (or - + + -) asci occurred at almost equal frequencies. The two coherent spores at each end of an ascus were always marked with different alleles of a gene.  相似文献

1. The relationship between thermal salivation (TS) and thermoregulation was studied in anesthetized rats. 2. Of the 6 anesthetics used, ketamine-anesthetized rats secreted the largest amount of saliva. Salivation, however, was thermal and not induced by ketamine itself. 3. Ketamine-anesthetized rats readily secreted saliva at core temperatures less than 40 degrees C but TS was remarkably enhanced by hyperthermia of 40-42.5 degrees C. 4. The equilibrium phase in the triphasic heat response of core temperature was a consequence of equilibrium between heat gain and heat loss by salivation.  相似文献

Although fatigue is a common and distressing symptom in cancer survivors, the mechanism of fatigue is not fully understood. Therefore, this study aims to investigate the relation between the fatigue and mindfulness of breast cancer survivors using anxiety, depression, pain, loneliness, and sleep disturbance as mediators. Path analysis was performed to examine direct and indirect associations between mindfulness and fatigue. Participants were breast cancer survivors who visited a breast surgery department at a university hospital in Japan for hormonal therapy or regular check-ups after treatment. The questionnaire measured cancer-related-fatigue, mindfulness, anxiety, depression, pain, loneliness, and sleep disturbance. Demographic and clinical characteristics were collected from medical records. Two-hundred and seventy-nine breast cancer survivors were registered, of which 259 answered the questionnaire. Ten respondents with incomplete questionnaire data were excluded, resulting in 249 participants for the analyses. Our final model fit the data well (goodness of fit index = .993; adjusted goodness of fit index = .966; comparative fit index = .999; root mean square error of approximation = .016). Mindfulness, anxiety, depression, pain, loneliness, and sleep disturbance were related to fatigue, and mindfulness had the most influence on fatigue (β = − .52). Mindfulness affected fatigue not only directly but also indirectly through anxiety, depression, pain, loneliness, and sleep disturbance. The study model helps to explain the process by which mindfulness affects fatigue. Our results suggest that mindfulness has both direct and indirect effects on the fatigue of breast cancer survivors and that mindfulness can be used to more effectively reduce their fatigue. It also suggests that health care professionals should be aware of factors such as anxiety, depression, pain, loneliness, and sleep disturbance in their care for fatigue of breast cancer survivors. Although females suffer twice as much as males from stress-related disorders, sex-specific participating and pathogenic cellular stress mechanisms remain uncharacterized. Using corticotropin-releasing factor receptor 2–deficient (Crhr2−/− ) and wild-type (WT) mice, we show that CRF receptor type 2 (CRF2) and its high-affinity ligand, urocortin 1 (Ucn1), are key mediators of the endoplasmic reticulum (ER) stress response in a murine model of acute pancreatic inflammation. Ucn1 was expressed de novo in acinar cells of male, but not female WT mice during acute inflammation. Upon insult, acinar Ucn1 induction was markedly attenuated in male but not female Crhr2−/− mice. Crhr2−/− mice of both sexes show exacerbated acinar cell inflammation and necrosis. Electron microscopy showed mild ER damage in WT male mice and markedly distorted ER structure in Crhr2−/− male mice during pancreatitis. WT and Crhr2−/− female mice showed similarly distorted ER ultrastructure that was less severe than distortion seen in Crhr2−/− male mice. Damage in ER structure was accompanied by increased ubiquitination, peIF2, and mistargeted localization of vimentin in WT mice that was further exacerbated in Crhr2−/− mice of both sexes during pancreatitis. Exogenous Ucn1 rescued many aspects of histological damage and cellular stress response, including restoration of ER structure in male WT and Crhr2−/−mice, but not in females. Instead, females often showed increased damage. Thus, specific cellular pathways involved in coping and resolution seem to be distinct to each sex. Our results demonstrate the importance of identifying sex-specific pathogenic mechanisms and their value in designing effective therapeutics.  相似文献
