The adult female of the freshwater ostracod Cyprinotus uenoi Brehm, 1936 (Podocopida: Cypridoidea) has a pair of long, sac-like ovaries separately lying in the posterior part of the left and the right carapace valves. Oogonia and very early previtellogenic oocytes are located in the terminal germarium of each ovary. In the germarium, the oogonia occur in the most terminal region, and the very early previtellogenic oocytes are located in the remainder, arranged in order of size, the larger ones nearer the ovarian lumen. Most of the growing oocytes, previtellogenic and vitellogenic, are found in the ovarian lumen, the larger ones farther from the germarium. In the germarium, a cytoplasmic bridge connects a pair of adjoining germ cells, resulting from an incomplete cytokinesis of oogonial division. Among the previtellogenic and early vitellogenic oocytes in the ovarian lumen, "nurse cells" are found as small, spherical cells in mostly the same number as these oocytes. A cytoplasmic bridge connects each "nurse cell" to an adjoining oocyte. Based on the manner of connection and some morphological features, we consider that each "nurse cell" originates from one of each pair of adjoining germ cells connected by a cytoplasmic bridge in the germarium, as in the true nurse cells of several branchiopod crustaceans and insects with meroistic ovarioles.  相似文献

Cyclodextrans (CIs) are cyclic isomaltooligosaccharides and only CI-7, CI-8, and CI-9 were known. CI-7, CI-8, and CI-9, consisting of seven, eight, and nine glucoses, respectively, bound by alpha-(1-->6) linkages, are known to be produced by T-3040 strain of Bacillus circulans. However, we have found, using 13C NMR and mass spectrometry, that this strain also produces CI-10, CI-11 and CI-12. These large CIs are very soluble in water and inhibit the glucan synthesis of glucansucrases to the same degree as do the smaller CIs. The CIs were thought to be poor at forming inclusion complexes with chemical compounds, due to their flexible alpha-(1-->6)-glucosidic structure. Among these six CIs, CI-10 was much better at forming an inclusion complex, and it ability to do so was as good as cyclodextrins, as determined by its ability to stabilize the color of Victoria blue B. Therefore, CI-10 may be the most commercially useful CI.  相似文献

Human kallikrein 8 (hK8), whose gene was originally cloned as the human ortholog of a mouse brain protease, is known to be associated with diseases such as ovarian cancer and Alzheimer's disease. Recombinant human pro-kallikrein 8 was activated with lysyl endopeptidase-conjugated beads. Amino-terminal sequencing of the activated enzyme demonstrated the cleavage of a 9-aa propeptide from the pro-enzyme. The substrate specificity of activated hK8 was characterized using synthetic fluorescent substrates. hK8 showed trypsin-like specificity, as predicted from sequence analysis and enzymatic characterization of the mouse ortholog. All synthetic substrates tested containing either arginine or lysine at P1 position were cleaved by hK8. The highest kcat/Km value of 20x10(3)M-1 s-1 was observed with Boc-Val-Pro-Arg-7-amido-4-methylcoumarin. The activity of hK8 was inhibited by antipain, chymostatin, and leupeptin. The concentration for 50% inhibition by the best inhibitor, antipain, was 0.46 microM. The effect of different metal ions on the enzyme activity was analyzed. Whereas Na+ had no effect on hK8 activity, Ni2+ and Zn2+ decreased the activity and Ca2+, Mg2+, and K+ had a stimulatory effect. Ca2+ was the best activator, with an optimal concentration of approximately 10 microM.  相似文献

The IAN (immune-associated nucleotide-binding protein) family is a family of functionally uncharacterized GTP-binding proteins expressed in vertebrate immune cells and in plant cells during antibacterial responses. Here we show that all eight IAN family genes encoded in a single cluster of mouse genome are predominantly expressed in lymphocytes, and that the expression of IAN1, IAN4, and IAN5 is significantly elevated upon thymic selection of T lymphocytes. Gain-of-function experiments show that the premature overexpression of IAN1 kills immature thymocytes, whereas short hairpin RNA-mediated loss-of-function studies show that IAN4 supports positive selection. The knockdown of IAN5 perturbs the optimal generation of CD4/CD8 double-positive thymocytes and reduces the survival of mature T lymphocytes. We also show evidence suggesting that IAN4 and IAN5 are associated with anti-apoptotic proteins Bcl-2 and Bcl-xL, whereas IAN1 is associated with pro-apoptotic Bax. Thus, the IAN family is a novel family of T cell–receptor-responsive proteins that critically regulate thymic development and survival of T lymphocytes and that potentially exert regulatory functions through the association with Bcl-2 family proteins.  相似文献

Novel nucleotide analogues have been synthesized from morpholine subunits with thiocarbamate linkages. They indicated much stronger interaction with poly U or poly dT than the corresponding natural oligodeoxyribonucleotides. The complete primary structure of a base non-specific and adenylic acid preferential RNase (RNase Le2) from the fruit bodies of Lentinus edodes was analyzed. The sequence was mostly determined by analysis of the peptides generated by V8 protease digestion and BrCN cleavage (including alpha-chymotryptic, and V8 protease digest of BrCN fragments). It consists of 239 amino acid residues. The molecular weight is 25831. The location of 10 half cystine residues were almost superimposable on those of known fungal RNases of the RNase T2 family. The sequence homologies between RNase Le2 and four known fungal RNases of the RNase T2 family, RNase T2, RNase M, RNase Trv, and RNase Rh, are 102, 103, 109, and 74, respectively. The homologous sequences are concentrated around the three histidines, which are supposed to form the active site of RNase T2 family RNases.  相似文献

Morphology and cytochemical properties of “mycetomes” are described in the developing oocytes and eggs of an idolothripine thrips, Bactrothrips brevitubus (Thysanoptera). The “mycetome” is an aggregation of numerous granules of various sizes. We found no membrane encapsulating the aggregation of granules. Two types of granules are distinguishable: the smaller ones filled with electron-dense material and the larger ones with inclusion of myelin-like structures. Each of the granules has a limiting membrane. The limiting membrane is a simple unit membrane but shows no characteristics of cell walls. No nucleoid or nucleoplasm is detected in the granules. The “mycetome” takes up dyes whose specific incorporation into lysosomes has been demonstrated. In addition, a high activity of acid phosphatase is demonstrated in the “mycetome.” These characteristics apparently indicated that the “mycetome” of Bactrothrips brevitubus is an aggregation of lysosomes but not a clump of microorganisms. Thus we propose that the structure being regarded as the mycetome should be renamed the “lysosomal aggregation.” © 1994 Wiley-Liss, Inc.  相似文献
