During endochondral ossification, growth plate chondrocytes release plasma membrane (PM) derived matrix vesicles (MV), which are the site of initial hydroxyapatite crystal formation. MV constituents which facilitate the mineralization process include the integral membrane ectoenzymes alkaline phosphatase (ALPase) and nucleotide pyrophosphatase phosphodiesterase (NPP1/PC-1), along with a phosphatidylserine- (PS-) rich membrane surface that binds annexins and calcium, resulting in enhanced calcium entry into MV. In this study, we determined that chick growth plate MV were highly enriched in membrane raft microdomains containing high levels of cholesterol, glycophosphatidylinositol- (GPI-) anchored ALPase, and phosphatidylserine (PS) localized to the external leaflet of the bilayer. To determine how such membrane microdomains arise during chondrocyte maturation, we explored the role of PM cholesterol-dependent lipid assemblies in regulating the activities of lipid translocators involved in the externalization of PS. We first isolated and determined the composition of detergent-resistant membranes (DRMs) from chondrocyte PM. DRMs isolated from chondrocyte PM were enhanced in ganglioside 1 (GM1) and cholesterol as well as GPI-anchored ALPase. Furthermore, these membrane domains were enriched in PS (localized to the external leaflet of the bilayer) and had significantly higher ALPase activity than non-cholesterol-enriched domains. To understand the role of cholesterol-dependent lipid assemblies in the externalization of PS, we measured the activities of two lipid transporters involved in PS externalization, aminophospholipid translocase (APLT) and phospholipid scramblase (PLSCR1), during maturation of a murine chondrocytic cell line, N1511. In this report, we provide the first evidence that maturing chondrocytes express PLSCR1 and have scramblase activity. We propose that redistribution of PS is dependent on an increase in phospholipid scramblase activity and a decrease in APLT activity. Lastly, we show that translocator activity is most likely to be modulated by membrane cholesterol levels through a membrane raft microdomain.  相似文献

We have investigated the interactions between synthetic amphipathic peptides and zwitterionic model membranes. Peptides with 14 and 21 amino acids composed of leucines and phenylalanines modified by the addition of crown ethers have been synthesized. The 14-mer and 21-mer peptides both possess a helical amphipathic structure as revealed by circular dichroism. To shed light on their mechanism of membrane interaction, different complementary biophysical techniques have been used such as circular dichroism, fluorescence, membrane conductivity measurement and NMR spectroscopy. Results obtained by these different techniques show that the 14-mer peptide is a membrane perturbator that facilitate the leakage of species such as calcein and Na ions, while the 21-mer peptide acts as an ion channel. (31)P solid-state NMR experiments on multilamellar vesicles reveal that the dynamics and/or orientation of the polar headgroups are greatly affected by the presence of the peptides. Similar results have also been obtained in mechanically oriented DLPC and DMPC bilayers where different acyl chain lengths seem to play a role in the interaction. On the other hand, (2)H NMR experiments on multilamellar vesicles demonstrate that the acyl chain order is affected differently by the two peptides. Based on these studies, mechanisms of action are proposed for the 14-mer and 21-mer peptides with zwitterionic membranes.  相似文献

Background  

The phylum Chlorophyta contains the majority of the green algae and is divided into four classes. While the basal position of the Prasinophyceae is well established, the divergence order of the Ulvophyceae, Trebouxiophyceae and Chlorophyceae (UTC) remains uncertain. The five complete chloroplast DNA (cpDNA) sequences currently available for representatives of these classes display considerable variability in overall structure, gene content, gene density, intron content and gene order. Among these genomes, that of the chlorophycean green alga Chlamydomonas reinhardtii has retained the least ancestral features. The two single-copy regions, which are separated from one another by the large inverted repeat (IR), have similar sizes, rather than unequal sizes, and differ radically in both gene contents and gene organizations relative to the single-copy regions of prasinophyte and ulvophyte cpDNAs. To gain insights into the various changes that underwent the chloroplast genome during the evolution of chlorophycean green algae, we have sequenced the cpDNA of Scenedesmus obliquus, a member of a distinct chlorophycean lineage.  相似文献

Growing interest in the structure and dynamics of animal social networks has stimulated major advances [1-3], but recording reliable association data for wild populations has remained challenging. While animal-borne 'proximity' tags have been available for some time [4], earlier devices were comparatively heavy, had limited detection ranges and/or necessitated recovery for data retrieval. We have developed wireless digital transceiver technology ('Encounternet') that enables automated mapping of social networks in wild birds, yielding datasets of unprecedented size, quality and spatio-temporal resolution. Miniature, animal-borne tags record the proximity and duration of bird encounters, and periodically transfer logs to a grid of fixed receiver stations, from which datasets can be downloaded remotely for real-time analysis. We used our system to chart social associations in New Caledonian crows Corvus moneduloides[5,6]. Analysis of ca. 28,000 encounter logs for 34 crows over a 7-day period reveals a substantial degree of close-range association between non-family birds, demonstrating the potential for horizontal and oblique information exchange.  相似文献

Observations in the field indicate that monarch butterflies will oviposit on dog‐strangler vine, an invasive introduced species in the same family as milkweed (Asclepias spp.), the principal larval host of monarchs. The potential impact of this behaviour depends on the strength of the preference of monarch adults to oviposit on these two hosts and the relative ability of larvae to survive on each. We determined the preference for milkweed vs. dog‐strangler vine of ovipositing adults and first instar larvae in choice and no‐choice tests. We also compared the ability of larvae to consume, develop, and survive on either host. In the presence of both hosts, adults exhibited a strong preference to oviposit on milkweed over dog‐strangler vine (mean 80.7 eggs compared to 0.4 eggs over 48 h, respectively). In the absence of milkweed, adults ceased oviposition (mean 0.9 eggs in 48 h), but resumed oviposition when the dog‐strangler vine was replaced with milkweed (mean 99.1 eggs in 48 h). Given a choice between hosts over 24 h, 92% of larvae moved to milkweed leaves and consumed 3.94 cm2 of milkweed leaves compared to 2% of larvae that moved to dog‐strangler vine and consumed negligible amounts of leaf material (0.01 cm2). Without a choice, larvae on dog‐strangler vine never consumed more than mean 0.02 cm2 larva?1 in a 24‐h period, did not develop beyond the first instar, and died within 96 h. We obtained no data in support of an effect of the presence of dog‐strangler vine on monarch butterfly populations.  相似文献

To gain insights into the phylogeny of charophytes and into their relationships with other green algae and bryophytes, we analyzed the chloroplast small and large subunit rRNA sequences of charophytes belonging to five orders (Charales, Coleochaetales, Desmidiales, Klebsormidiales, and Zygnematales), of chlorophytes from the four remaining classes of green algae, and of bryophytes representing the three classes reported in this group of land plants. We also probed the gene organization and intron content of the chloroplast rDNA operon in charophytes and bryophytes. The organization of this operon proved to be highly conserved, except in members of the Desmidiales and Zygnematales. Homologous group II introns were identified in the trnA(UGC) gene of all charophyte groups examined and in the trnI(GAU) gene of charophytes from all orders except the Desmidiales and Zygnematales. Phylogenetic analyses of concatenated rDNA sequences consistently placed the prasinophyte Mesostigma viride Lauterborn at the base of the Streptophyta and Chlorophyta, although alternative topologies positioning Mesostigma within the Streptophyta could not be rejected. A sister group relationship was unambiguously established between Chaetosphaeridium globosum (Nordstedt) Klebahn and members of the Coleochaetales. The Charales, Coleochaetales, Desmidiales, and Zygnematales were found to be monophyletic, and a sister group relationship was observed for the Desmidiales and Zygnematales. Although our analyses failed to resolve the branching order of the Coleochaetales, Charales, Desmidiales/Zygnematales, and bryophytes, they revealed that the problematic charophyte taxon Entransia fimbriata Hughes strongly clusters with Klebsormidium flaccidum (Kützing) Silva, Mattox et Blackwell to form a basal lineage relative to the other charophyte orders examined.  相似文献
