Chenopodium rubrum L. ecotype 184 is a qualitative short-day plant with critical length of the night of eight hours that must be exceeded in order to flower: Five days after sowing, the plants were exposed to a various number of inductive cycles (14/10 h of däy/night cycle) to test the optimal photoperiodic conditions for flowering. In our experimental conditions the plants flowered with high percentage after more than four received inductive cycles, but there was no flowering below that. The plants grown on the herbicide Norflurazon (photobleached plants) showed different photoperiodic characteristics. There was negligible flowering of photobleached plants in the same experimental conditions as for the green ones.  相似文献

Molecular studies highlighted a strong genetic affinity between the remnant and isolated population of the Apennine brown bear and other southern European populations. Despite this genetic closeness a recent morphometric study revealed a marked phenotypic distinctiveness of the Apennine population, supporting the reinstatement of a distinct taxon (Ursus arctos marsicanus). By building upon previous morphological analyses, we adopted geometric morphometrics to better investigate the skull morphology of the Apennine brown bear with reference to the other, closely related southern European populations. Both skull shape and size differences confirmed the strong divergence of U. arctos marsicanus. In particular, the Apennine bears are characterized by an enlargement of the supraorbital apophysis and a larger distance across the zygomatic arches. Furthermore, our analyses highlighted significant shape differences of the first upper molar in the Apennine bears. Our results suggest that the Apennine bears underwent a rapid morphological change, possibly driven by genetic drift and local selective pressures. Because the greatest morphological differentiation is likely to be related to the muscles involved in mastication, we hypothesize that local selective pressures might be related to a shift in food habits, with highly reduced depredation and feeding on large carcasses in favour of vegetation and hard mast (beech nuts and acorns). These results suggest an adaptive distinctiveness of the Apennine bears, which should be carefully considered in any management and conservation action addressed to this highly endangered population. Although more in‐depth molecular studies are required to better assess the taxonomic and genetic status of the Apennine brown bear population, our study emphasizes the importance of morphological analyses as a complementary tool for a more thorough characterization of variation and divergence in endangered taxa. © 2012 The Linnean Society of London, Biological Journal of the Linnean Society, 2012, ?? , ??–??.  相似文献

In anesthetized Wistar rats, we studied the effect of electrical stimulation of the locus coeruleus (LC) on the firing rates of Purkinje cells using spectral analysis. The frequency of extracellularly recorded activity of Purkinje cells was measured before and during the 1st, 5th, 6th, and 11th min after cessation of 10-sec-long LC stimulations. Spectral analysis of the Purkinje cell firing rates (imp./bin, the bin duration was 2-8 sec) for 60- to 120-sec-long intervals was performed using fast Fourier transformation after digital conversion of unitary spikes. Mean power spectra of the Purkinje cell firing rates (derived from 8-sec-long consecutive epochs at a sampling rate of 256 sec-1) showed an increase in the slow frequency range (0.1-1.0 Hz) after LC stimulation, particularly due to the slowest components (below 0.5 Hz). This effect lasted more than 1 min and usually less than 6 min after cessation of LC stimulation and could be interpreted as the development of slow oscillations in the Purkinje cell firing. Our results suggest that slow oscillations of the firing rate of cerebellar output neurons, induced by LC stimulation, reflect a specific coordination of the cerebellar neuronal activities (important for a central norepinephrine influence) in regulation of different pathological states.  相似文献

Polycystin-2 (PC2) is a Ca2+-permeable transient receptor potential channel activated and regulated by changes in cytoplasmic Ca2+. PC2 mutations are responsible for ∼15% of autosomal dominant polycystic kidney disease. Although the C-terminal cytoplasmic tail of PC2 has been shown to contain a Ca2+-binding EF-hand domain, the molecular basis of PC2 channel gating by Ca2+ remains unknown. We propose that the PC2 EF-hand is a Ca2+ sensor required for channel gating. Consistent with this, Ca2+ binding causes a dramatic decrease in the radius of gyration (Rg) of the PC2 EF-hand by small angle x-ray scattering and significant conformational changes by NMR. Furthermore, increasing Ca2+ concentrations cause the C-terminal cytoplasmic tail to transition from a mixture of extended oligomers to a single compact dimer by analytical ultracentrifugation, coupled with a >30 Å decrease in maximum interatomic distance (Dmax) by small angle x-ray scattering. Mutant PC2 channels unable to bind Ca2+ via the EF-hand are inactive in single-channel planar lipid bilayers and inhibit Ca2+ release from ER stores upon overexpression in cells, suggesting dominant negative properties. Our results support a model where PC2 channels are gated by discrete conformational changes in the C-terminal cytoplasmic tail in response to changes in cytoplasmic Ca2+ levels. These properties of PC2 are lost in autosomal dominant polycystic kidney disease, emphasizing the importance of PC2 to kidney cell function. We speculate that PC2 and the Ca2+-dependent transient receptor potential channels in general are regulated by similar conformational changes in their cytoplasmic domains that are propagated to the channel pore.  相似文献

In the present study, developmental stability of leaf traits was examined in three natural populations of Plantago major L. (ssp. major), representing two polluted environments (Karaburma and Zemun) and an unpolluted area (Crni Lug). Developmental stability was assessed as fluctuating asymmetry (FA). The magnitude of FA is believed to reflect differences in the ability of individuals to buffer their development in natural populations. We hypothesized that there are differences within characters and among characters in response to environmental conditions. Significant patterns of asymmetry correlations and asymmetry changes were detected both within characters and between characters. The manova results revealed a significant effect of individual and a significant individual × environment interaction on actual asymmetry (logLi ? logRi) and on the amount of asymmetry |(logLi ? logRi)| for leaf width and vein distances within a leaf. Over time, statistically significant and positive correlations of the FA values were detected for each trait separately per sample (population). For both leaf traits, there were differences for (logLi ? logRi) and |(logLi ? logRi)| asymmetry values among individuals within samples in response to yearly variations. Statistically significant and negative correlations for (logLi ? logRi) versus |(logLi ? logRi)| asymmetries were detected for both leaf traits. In summary, our results highlight the importance of differences in the ability of individuals to buffer their development under different environmental conditions and point to the concept that developmental stability is character specific.  相似文献

The aim of this study was to investigate the effects of a static magnetic field on six behavioral parameters (travel distance, average speed while in motion, travel distance of the head, body rotations, time in movement, and immobility time) of the ground beetle Laemostenus (Pristonychus) punctatus (Dejean) (Coleoptera: Carabidae). Adults of this troglophilic and guanophilic beetle were randomly divided into two groups, the first exposed to a static magnetic field of 110 mT for 5 h, and the second a control group. Beetle behavior after these 5 h was monitored in an open‐field test for 12 min and analyzed using ANY‐maze software. Exposure to a static magnetic field of 110 mT increased motor activity (travel distance and average speed while in motion) in the first 4 min. After that there were no significant differences. We conclude that the applied static magnetic field affects motor activity of adult specimens of L. (P.) punctatus, and we discuss the mechanism, possibly through acting on the control centers responsible for orientation and movement.  相似文献

Results of previous morphometric and genetic analyses of grey wolf (Canis lupus L.) population from Serbia indicated different patterns of population subdivision. In order to explore population structure, level of genetic variability, genetic drift, inbreeding and signals of bottleneck for grey wolves from Serbia, we applied highly polymorphic genetic markers (microsatellites). Obtained data are valuable in determination of conservation units and creation of appropriate management plans. We have amplified 18 highly polymorphic microsatellites, in a total sample of 75 grey wolves, from different localities across Serbia and multilocus genotypes were analyzed using appropriate software. Observed values of the basic genetic parameters (HO = 0.69; HE = 0.75) indicated moderate level of genetic variability, similar to genetic variability in other populations belonging to the Dinaric-Balkan population of grey wolf. In STRUCTURE analysis, although ΔK was estimated to be at first peak K = 2, and second peak K = 4, CLUMPAK analyses showed that there’s no structuring for any of assumed K, and therefore the population of grey wolf from Serbia may be considered as one continuous population and treated as one conservation unit in future management plans. Signals of bottleneck haven’t been observed (Wilcoxon test two phase mutation model p = 0.247; and stepwise mutation model p = 0.815).  相似文献

In this work, the chemical composition, antimicrobial and cytotoxic activity of Heracleum verticillatum Pan?i? and H. ternatum Velen . root, leaf, and fruit essential oils were investigated. The composition was analyzed by GC and GC/MS. Heracleum verticillatum and H. ternatum root oils were dominated by monoterpenes, mostly β‐pinene (23.5% and 47.3%, respectively). Heracleum verticillatum leaf oil was characterized by monoterpenes, mainly limonene (20.3%), and sesquiterpenes, mostly (E)‐caryophyllene (19.1%), while H. ternatum leaf oil by the high percentage of phenylpropanoids, with (Z)‐isoelemicin (35.1%) being dominant constituent. Both fruit oils contained the majority of aliphatic esters, mostly octyl acetate (42.3% in Hverticillatum oil and 49.0% in Hternatum oil). The antimicrobial activity of the oils was determined by microdilution method against eight bacterial and eight fungal strains. The strongest effect was exhibited by H. verticillatum root oil, particularly against Staphylococcus aureus, Salmonella typhimurium (MICs = 0.14 mg/ml, MBCs = 0.28 mg/ml), and Trichoderma viride (MIC = 0.05 mg/ml, MFC = 0.11 mg/ml). Cytotoxic effect was determined by MTT test against malignant HeLa, LS174, and A549 cells (IC50 = 5.9 – 146.0 μg/ml), and against normal MRC‐5 cells (IC50 > 120.1 μg/ml). The best effect was exhibited by H. verticillatum root oil on A549 cells (IC50 = 5.9 μg/ml), and H. ternatum root oil against LS174 cells (IC50 = 6.7 μg/ml).  相似文献

The Streptomyces coelicolor A3(2) gene SCI11.14c was overexpressed and purified as a His-tagged protein from heterologous host, Streptomyces lividans. The purification procedure resulted in 34.1-fold increase in specific activity with an overall yield of 21.4%. Biochemical and physical properties of the purified enzyme were investigated and it was shown that it possesses (aryl)esterase and a true lipase activity. The enzyme was able to hydrolyze p-nitrophenyl-, α- and β-naphthyl esters and poly(oxyethylene) sorbitan monoesters (Tween 20–80). It showed pronounced activity towards p-nitrophenyl and α- and β-naphthyl esters of C12–C16. Higher activity was observed with α-naphthyl esters. The enzyme hydrolyzed triolein (specific activity: 91.9 U/mg) and a wide range of oils with a preference for those having higher content of linoleic or oleic acid (C18:2; C18:1, cis). The active-site serine specific inhibitor 3,4-dichloroisocoumarin (DCI) strongly inhibited the enzyme, while tetrahydrofurane and 1,4-dioxane significantly increased (2- and 4- fold, respectively) hydrolytic activity of lipase towards p-nitrophenyl caprylate. The enzyme exhibited relatively high temperature optimum (55 °C) and thermal stability. CD analysis revealed predominance of α-helical structure (54% α-helix, 21% β-sheet) and a Tm value at 66 °C.  相似文献
