An imbalance between the phases of biotransformation systems, such as activation, detoxification, and release of toxic substances, is one of the causes of multifactor pathology. Therefore, it is important to examine the impact of the total contribution of the polymorphic variants of xenobiotic-metabolizing enzyme genes at all three phases on predisposition to lung cancer. The purposes of the present work were to study the relationship between polymorphic variants of xenobiotic-metabolizing enzyme genes and risk of lung cancer and to identify molecular genetic markers of predisposition to the disease. It was shown that GSTT1 null-genotype plays a dominant role in the development of lung cancer predisposition in the Belarusian population, while the polymorphic variants of other genes of xenobiotic-metabolizing enzymes render a modifying effect on predisposition to this disease. Combination 734AA CYP1A2/GSTT1(?)/GSTM1(+)/“slow” acetylator has the greatest risk significance, and combination GSTT1(?)/GSTM1(+)/“slow” acetylator exerts a protective effect.  相似文献

A regression analysis shows the direct linear relation between 137Cs accumulation in rodents and the level of aberrant cells. For 90Sr this trend was negative. The dose relationship was the same with 137Cs. The trends were negative 1 month later after feeding of animals with clean food. Correspondingly the dose relationship was also negative. The levels of cells with apoptosis features were different in animals from the control and the radiocontaminated sites. The analysis has revealed the correlation between the 90Sr content in animal body and the number of alveolar macrophages containing micronuclei. The relationship was revealed between the 137Cs content in the animal body and the number of intestinal epithelial cells with micronuclei.  相似文献

A new genus, Pseudolessonia, is proposed for the kelp Lessonia laminarioides Postels et Ruprecht (Laminariales, Phaeophyceae), which occurs on the northwest side of the Sea of Okhotsk, in the northwest Pacific Ocean. Pseudolessonia is monotypic and differs from Lessonia in its short primary stipes and its corrugated, unilaterally arranged blades with entire margins. This species is transferred on the basis of morphology and plastid gene sequence comparisons. We determined psaA and rbcL gene sequences from 17 taxa of Pseudolessonia, Lessonia, and putative relatives. Analyses of individual and combined data sets resulted in congruent trees showing a clear separation of Pseudolessonia laminarioides from Lessonia, but suggesting its sister relationships with the clade of Nereocystis, Macrocystis, Pelagophycus, and Postelsia in the North Pacific Ocean. On the other hand, Lessonia species from the South Pacific Ocean formed a strongly supported clade. The results indicate that the basal splitting of the blade, which has been considered a diagnostic character for the family Lessoniaceae, is a result of convergent evolution.  相似文献

The dynamics of the activity of cysteine-dependent enzymes involved in intracellular proteolysis was studied in freshwater salmon Salmo salar L. during its early development: in the period from the blastodisc formation to hatching and in eggs before fertilization. The Ca2+-activated proteolytic activity of calpains, the activity of acidic lysosomal protease cathepsin B, and the activity of several peptidases that use the chromogenic component p-nitroaniline-containing peptides (L-Ala-pNA, L-Pro-pNA, L-Arg-pNA, N-Bzl-L-Arg-pNA, L-Phe-pNA, N-Glu-L-Phe-pNA, and Z-Gly-Pro-pNA) as substrates were analyzed. It is shown that the activity of the enzymes of interest increases at the stage of late blastula and early gastrula. The next increase in the activity of cathepsin B and calpain is observed at the stage of eye pigmentation, and the activity of peptidases and cathepsin B increases in eggs before hatching. The functional role of changes in intracellular proteolytic enzymes in the processes of protein metabolism associated with the Atlantic salmon embryo is discussed.  相似文献

By Chromatographic separation of cytosol fraction of homogenates from gonads of females and males of green sea urchinStrongylocentrotus droebachiensis using an AcA-34 ultragel, protein fractions with mol. mass of 123 and 81 kDa are revealed, which have activity of calciumactivated proteinases. Dynamics of activity of the calcium-activated proteinases is studied during annual sexual cycles of the sea urchin. It is shown that the maximal activity of the studied enzyme in females and males of the sea urchin is present at the IV stage of gonad maturation, at the period of their trophic growth which is characterized by formation of stores of nutritient substances necessary for the subsequent development of embryos.  相似文献

The aggresome is an organelle that recruits aggregated proteins for storage and degradation. We performed an siRNA screen for proteins involved in aggresome formation and identified novel mammalian AAA+ protein disaggregases RuvbL1 and RuvbL2. Depletion of RuvbL1 or RuvbL2 suppressed aggresome formation and caused buildup of multiple cytoplasmic aggregates. Similarly, downregulation of RuvbL orthologs in yeast suppressed the formation of an aggresome‐like body and enhanced the aggregate toxicity. In contrast, their overproduction enhanced the resistance to proteotoxic stress independently of chaperone Hsp104. Mammalian RuvbL associated with the aggresome, and the aggresome substrate synphilin‐1 interacted directly with the RuvbL1 barrel‐like structure near the opening of the central channel. Importantly, polypeptides with unfolded structures and amyloid fibrils stimulated the ATPase activity of RuvbL. Finally, disassembly of protein aggregates was promoted by RuvbL. These data indicate that RuvbL complexes serve as chaperones in protein disaggregation.  相似文献

Nutritional attractiveness of the brown alga Laminaria japonica for the sea urchin Strongylocentrotus intermedius was studied. The composition of L. japonica was analyzed after one and two years of its life under natural conditions, in its seedlings, and in the alga partially degraded by natural factors. Substances extracted with various solvents were tested for the presence of inhibitors and activators of 1,3-β-D-glucanase, a digestive enzyme of the sea urchin. Ethanolic extract of freshly harvested L. japonica was found to suppress the enzyme activity. Substances present in ethanolic extracts of the alga after one or two years of its life cycle and in the alga, partly degraded by natural factors, activated the sea urchin enzyme. This fact is in agreement with earlier natural observations concerning the nutritional attractiveness of such L. japonica samples for Strongylocentrotus intermedius.  相似文献
