Electrospray ionization sources, used with triple quadrupole mass spectrometers from PE/Sciex (API III+), Micromass (Quattro II), and Finnigan (TSQ 7000), were modified with a 35-gauge stainless steel needle. The dimensions of the needle were 63 μm i.d. by 145 μm o.d. with variable length, depending on the specific instrument. This modification led to enhanced signal stability, improved signal/noise ratios, and lowered sample consumption for a wide range of peptides. Stable baselines were observed with flow rates in the range of 50 nL/min to 5 μL/min. An alternative design, based on a metal wire housed within a fused silica capillary, led to the most stable signals of all during infusion, but caused excessive peak broadening with capillary chromatography. The Finnigan interface was further modified with an external postcolumn addition tee, used in conjunction with capillary liquid chromatography columns of 30 and 50 μm internal diameter. The best results with the modified Finnigan interface were acquired using the 50-μm column at a flow rate of 150 to 200 nL/min. 相似文献

We have constructed a partial linkage map in tetraploid potato which integrates simplex, duplex and double-simplex AFLP markers.
The map consists of 231 maternal and 106 paternal markers with total map lengths of 990.9 cM and 484.6 cM. The longer of the
two cumulative map lengths represents approximately 25% coverage of the genome. In tetraploids, much of the polymorphism between
parental clones is masked by `dosage' which significantly reduces the number of individual markers that can be scored in a
population. Consequently, the major advantage of using AFLPs – their high multiplex ratio – is reduced to the point where
the use of alternative multi-allelic marker types would be significantly more efficient. The segregation data and map information
have been used in a QTL analysis of late blight resistance, and a multi-allelic locus at the proximal end of chromosome VIII
has been identified which contributes significantly to the expression of resistance. No late blight resistance genes or QTLs
have previously been mapped to this location.

Removal of young leaves and application of the cytokinin, N-benzyla-α-(tetrahydro-2H-pyran-2yl)-adenine promote inflorescence development in Bougainvillea “San Diego Red”. Defoliation greatly increased the amount of assimilate accumulated at the shoot tip 1 to 2 days after treatment. Cytokinin applications further increased the amount accumulated and this increase was apparent 4 days before morphological changes could be detected at the inflorescence axes. Short days promoted inflorescence development and also increased assimilate accumulation at the reproductive axes; thus, it is suggested that the role of short day induction in bougainvillea may be that of redirecting the flow of assimilates, perhaps by its influence on cytokinin synthesis and distribution. 相似文献

Auditory information is relayed from the ventral nucleus of the medial geniculate complex to a core of three primary or primary-like areas of auditory cortex that are cochleotopically organized and highly responsive to pure tones. Auditory information is then distributed from the core areas to a surrounding belt of about seven areas that are less precisely cochleotopic and generally more responsive to complex stimuli than tones. Recent studies indicate that the belt areas relay to the rostral and caudal divisions of a parabelt region at a third level of processing in the cortex lateral to the belt. The parabelt and belt regions have additional inputs from dorsal and magnocellular divisions of the medial geniculate complex and other parts of the thalamus. The belt and parabelt regions appear to be concerned with integrative and associative functions involved in pattern perception and object recognition. The parabelt fields connect with regions of temporal, parietal, and frontal cortex that mediate additional auditory functions, including space perception and auditory memory. 相似文献

Angel is the first miniature inverted-repeat transposable element (MITE) isolated from fish. Angel elements are imperfect palindromes with the potential to form stem-loop structures in vitro. Despite sequence divergence
of elements of up to 55% within and between species, their inverted repeat structures have been maintained, implying functional
importance. We estimate that there are about 103–104
Angels scattered throughout the zebrafish genome, evidence that this family of transposable elements has been significantly amplified
over the course of evolution. Angel elements and Xenopus MITEs carry common sequence motifs at their termini, indicating common origin and/or related mechanisms of transposition.
We present a model in which MITEs take advantage of the basic cellular mechanism of DNA replication for their amplification,
which is dependent on the characteristic inverted repeat structures of these elements. We propose that MITEs are genomic parasites
that transpose via a DNA intermediate, which forms by a folding-back of a single strand of DNA, that borrow all of the necessary
factors for their amplification from products encoded in the genomes in which they reside. DNA polymorphisms in different
lines of zebrafish were detected by PCR using Angel-specific primers, indicating that such elements, combined with other transposons in vertebrate genomes, will be useful molecular
tools for genome mapping and genetic analyses of mutations.

Vascular endothelial growth factor (VEGF) is a hypoxia-induced protein that produces vascular permeability, and limited evidence suggests a possible role for VEGF in the pathophysiology of acute mountain sickness (AMS) and/or high-altitude cerebral edema (HACE). Previous studies demonstrated that plasma VEGF alone does not correlate with AMS; however, soluble VEGF receptor (sFlt-1), not accounted for in previous studies, can bind VEGF in the circulation, reducing VEGF activity. In the present study, we hypothesized that free VEGF is greater and sFlt-1 less in subjects with AMS compared with well individuals at high altitude. Subjects were exposed to 4,300 m for 19-20 h (baseline 1,600 m). The incidence of AMS was determined by using a modified Lake Louise symptom score and the Environmental Symptoms Questionnaire for cerebral effects. Plasma was collected at low altitude and after 24 h at high altitude, or at time of illness, and then analyzed by ELISA for VEGF and for soluble VEGF receptor, sFlt-1. AMS subjects had lower sFlt-1 at both low and high altitude compared with well subjects and a significant rise in free plasma VEGF on ascent to altitude compared with well subjects. We conclude that increased free plasma VEGF on ascent to altitude is associated with AMS and may play a role in pathophysiology of AMS. 相似文献

Inspiratory muscle weakness in patients with COPD is of major clinical relevance. For instance, maximum inspiratory pressure generation is an independent determinant of survival in severe COPD. Traditionally, inspiratory muscle weakness has been ascribed to hyperinflation-induced diaphragm shortening. However, more recently, invasive evaluation of diaphragm contractile function, structure, and biochemistry demonstrated that cellular and molecular alterations occur, of which several can be considered pathologic of nature. Whereas the fiber type shift towards oxidative type I fibers in COPD diaphragm is regarded beneficial, rendering the overloaded diaphragm more resistant to fatigue, the reduction of diaphragm fiber force generation in vitro likely contributes to diaphragm weakness. The reduced diaphragm force generation at single fiber level is associated with loss of myosin content in these fibers. Moreover, the diaphragm in COPD is exposed to oxidative stress and sarcomeric injury. This review postulates that the oxidative stress and sarcomeric injury activate proteolytic machinery, leading to contractile protein wasting and, consequently, loss of force generating capacity of diaphragm fibers in patients with COPD. Interestingly, several of these presumed pathologic alterations are already present early in the course of the disease (GOLD I/II), although these patients appear not limited in their daily life activities. Treatment of diaphragm dysfunction in COPD is complex since its etiology is unclear, but recent findings indicate the ubiquitin-proteasome pathway as a prime target to attenuate diaphragm wasting in COPD. 相似文献

Hard coral (Scleractinia and Milleporina) cover data were examined from 37 sites surveyed annually from 1996 to 2003 in the
Florida reef tract, USA. Analyses of species numbers and total cover showed that site-to-site differences were generally very
much greater than differences among times within sites. There were no significant differences among different geographical
areas within the reef tract (Upper, Middle and Lower Keys). Large-scale changes documented included a reduction in species
numbers and total cover on both deep and shallow offshore reefs between 1997 and 1999 followed by no recovery in cover, and
only scant evidence of any recovery in species numbers by 2003. These changes coincided with bleaching events in 1997 and
1998, and the passage of Hurricane Georges through the Lower Keys in 1998. The lack of recovery among offshore reefs suggests
that they were no longer resilient. Multivariate analyses revealed that some sites showed relatively little temporal variation
in community composition, essentially random in direction, while others showed relatively large year-on-year changes. There
was little evidence of any major region-wide changes affecting assemblage composition, or of any events that had impacted
all of the sampling sites in any single year. Instead, different sites exhibited differing patterns of temporal variation,
with certain sites displaying greater variation than others. Changes in community composition at some sites are interpreted
in the light of knowledge of events at those sites and the relative sensitivities of species to various stressors, such as
changes in cover of Acropora palmata and Millepora complanata at Sand Key following the bleaching events and hurricane in 1998, and declines in Montastraea annularis at Smith Shoal following a harmful algal bloom in 2002. For most sites, however, it is impossible to determine the causes
