Slow anion channels (SLAC/SLAH) are efflux channels previously shown to be critical for stomatal regulation. However, detailed analysis using the β‐glucuronidase reporter gene showed that members of the SLAC/SLAH gene family are predominantly expressed in roots, in addition to stomatal guard cells, implicating distinct function(s) of SLAC/SLAH in the roots. Comprehensive mutant analyses of all slac/slah mutants indicated that slah3 plants showed a greater growth defect than wild‐type plants when ammonium was supplied as the sole nitrogen source. Ammonium toxicity was mimicked by acidic pH in nitrogen‐free external medium, suggesting that medium acidification by ammonium‐fed plants may underlie ammonium toxicity. Interestingly, such toxicity was more severe in slah3 mutants and, particularly in wild‐type plants, was alleviated by supplementing the media with micromolar levels of nitrate. These data thus provide evidence that SLAH3, a nitrate efflux channel, plays a role in nitrate‐dependent alleviation of ammonium toxicity in plants.  相似文献

We measured the Δ Ψ and ΔpH components of the transthylakoid proton motive force ( pmf ) in light-adapted, intact tobacco leaves in response to moderate heat. The Δ Ψ causes an electrochromic shift (ECS) in carotenoid absorbance spectra. The light–dark difference spectrum has a peak at 518 nm and the two components of the pmf were separated by following the ECS for 25 s after turning the light off. The ECS signal was deconvoluted by subtracting the effects of zeaxanthin formation (peak at 505 nm) and the qE-related absorbance changes (peak at 535 nm) from a signal measured at 520 nm. Heat reduced ΔpH while Δ Ψ slightly increased. Elevated temperature accelerated ECS decay kinetics likely reflecting heat-induced increases in proton conductance and ion movement. Energy-dependent quenching (qE) was reduced by heat. However, the reduction of qE was less than expected given the loss of ΔpH. Zeaxanthin did not increase with heat in light-adapted leaves but it was higher than would be predicted given the reduced ΔpH found at high temperature. The results indicate that moderate heat stress can have very large effects on thylakoid reactions.  相似文献

ABSTRACT. The genus Hyperamoeba Alexeieff, 1923 was established to accommodate an aerobic amoeba exhibiting three life stages—amoeba, flagellate, and cyst. As more species/strains were isolated, it became increasingly evident from small subunit (SSU) gene phylogenies and ultrastructure that Hyperamoeba is polyphyletic and its species occupy different positions within the class Myxogastria. To pinpoint Hyperamoeba strains within other myxogastrid genera we aligned numerous myxogastrid sequences: whole small subunit ribosomal (SSU or 18S rRNA) gene for 50 dark‐spored (i.e. Stemonitida and Physarida) Myxogastria (including a new “Hyperamoeba/Didymium sequence) and a ~400‐bp SSU fragment for 147 isolates assigned to 10 genera of the order Physarida. Phylogenetic analyses show unambiguously that the type species Hyperamoeba flagellata is a Physarum (Physarum flagellatum comb. nov.) as it nests among other Physarum species as robust sister to Physarum didermoides. Our trees also allow the following allocations: five Hyperamoeba strains to the genus Stemonitis; Hyperamoeba dachnaya, Pseudodidymium cryptomastigophorum, and three other Hyperamoeba strains to the genus Didymium; and two further Hyperamoeba strains to the family Physaridae. We therefore abandon the polyphyletic and redundant genus Hyperamoeba. We discuss the implications for the ecology and evolution of Myxogastria, whose amoeboflagellates are more widespread than previous inventories supposed, being now found in freshwater and even marine environments.  相似文献

Abstract The herbicides DCMU, bentazon, amitrole, and SAN 6706 were tested for their ability to influence the carotenoid and pro-tochlorophyll(ide) composition as well as the protochloro-phyll(ide) phototransformation and the Shibata shift in dark-grown radish seedlings (Raphanus sativus L. cv. Saxa Treib). Bentazon enhanced the formation of lutein and carotenes, while SAN 6706 suppressed the biosynthesis of carotenoids. Amitrole led to a reduced accumulation of phototransformable pro-tochlorophyll(ide). The phototransformation of pro-tochlorophyll(ide) and the Shibata shift were not affected by any of the tested herbicides, irrespective of the presence or absence of activated phytochrome. From this we conclude that herbicides inhibiting photosystem II or producing chlorosis partly affect, but do not block, carotenoid and chlorophyll biosynthesis in dark-grown plants. The main herbicide effect becomes visible only after prolonged illumination.  相似文献

The recent spread of Salvinia molesta , a free floating weed in tropical and sub-tropical regions has attracted attention the world over. In India, many states face serious problems on account of Salvinia infestation. Several methods of controlling the weed have been tried, but with limited success. Biological control by snails, especially Pila globosa , a common tropical snail found in Kerala and other parts of India is suggested. It was found to feed voraciously on Salvinia , but not on paddy which forms a major crop in Kerala and other parts of India. The snail has efficient adaptations for a tropical environment in aquatic, semi-aquatic and even terrestrial habitats and its flesh is edible. Further studies on the performance of the snails in the field are, however, required.  相似文献

Several insect taxa have evolved symbioses with actinobacteria that protect the host or its nutritional resources against pathogens. Digger wasps of the genus Philanthus (‘beewolves’; Hymenoptera, Crabronidae) cultivate ‘Candidatus Streptomyces philanthi’ (Ca. S. philanthi) in specialized antennal glands and transfer them to subterranean brood cells, where the symbionts provide protection for the beewolf larva against pathogens by producing a cocktail of antibiotic substances. The present study investigates the occurrence of antennal symbionts in two species of the genus Trachypus, which is the closest relative to Philanthus. Cross sections of the antennae from females of both Trachypus denticollis and Trachypus boharti reveal bacteria‐containing gland reservoirs that are morphologically very similar to those found in Philanthus spp. Polymerase chain reaction‐based screens with specific primers, sequencing of the partial 16S rRNA gene, and fluorescence in situ hybridization confirm the presence of close relatives of ‘Ca. S. philanthi’ in T. denticollis and T. boharti. However, 16S rRNA sequence divergence between Trachypus spp. and Philanthus spp. symbionts is much higher for T. boharti than for T. denticollis, suggesting that horizontal transmission and/or de novo uptake of symbionts from the environment occurs occasionally. The results obtained indicate that the protective symbiosis with antennal actinobacteria is older and more widespread than previously recognized and occurs in at least two genera of digger wasps that comprise approximately 170 species.  相似文献

1. Although leaf morphology and chemistry show profound changes as trees age, the consequences of such changes to herbivory have received little attention, particularly late in the ontogeny of canopy trees. 2. Using a mobile aerial lift for canopy access, patterns of leaf damage were evaluated in canopy‐dominant mature sugar maple (Acer saccharum Marsh) trees ranging from ~20 to 70 cm in diameter, corresponding to an age range of ~40–180 years. 3. Herbivore damage patterns varied in relation to both vertical canopy position (among upper‐, mid‐, and lower‐canopy positions) and with tree size. Damage types attributable to herbivores active on leaf surfaces, including leaf skeletonizers and leaf cutters (both principally Lepidoptera), and leaf stippling inducers (Hemiptera) showed decreases with tree size, and with increasing height in the canopy. In contrast, leaf damage from the most abundant gall‐forming arthropod in the system, the eriophyid mite Vasates aceriscrumena, increased markedly with tree size. 4. The results indicate that herbivory patterns vary with both canopy stratum and with tree size in sugar maple, and that the relative strength of vertical stratification and tree ontogeny effects are similar in magnitude. The predominant patterns are of a decrease in herbivory with increasing height in the canopy and with tree size, but certain galling arthropods exhibit the reverse trends.  相似文献

Toothed mysticetes of the family Aetiocetidae from Oligocene rocks of the North Pacific play a key role in interpretations of cetacean evolution because they are transitional in grade between dorudontine archaeocetes and edentulous mysticetes. The holotype skull of Aetiocetus weltoni from the late Oligocene (28–24 Ma) of Oregon, USA, has been further prepared, revealing additional morphological features of the basicranium, rostrum and dentary that have important implications for mysticete evolution and functional anatomy. The palate of Aetiocetus weltoni preserves diminutive lateral palatal foramina and associated delicate sulci which appear to be homologous with the prominent palatal foramina and sulci that occur along the lateral portion of the palate in extant mysticetes. In modern baleen whales these foramina allow passage of branches of the superior alveolar artery, which supplies blood to the epithelia of the developing baleen racks. As homologous structures, the lateral palatal foramina of A. weltoni suggest that baleen was present in this Oligocene toothed mysticete. Cladistic analysis of 46 cranial and dental characters supports monophyly of the Aetiocetidae, with toothed mysticetes Janjucetus and Mammalodon positioned as successive sister taxa. The Nymphaeales (water-lilies and relatives) represent one of the earliest branching lineages of angiosperms and comprise about 70 aquatic species. Here, we present a comprehensive study of phylogenetic relationships within the Nymphaeales from a dataset containing 24 representatives of the order, including all currently recognized genera and all subgenera of the genus Nymphaea , plus 5 outgroup taxa. Nine different regions of the chloroplast genome − comprising spacers, group II introns, a group I intron, and a protein coding gene − were analysed. This resulted in a character matrix of 6597 positions and an additional 369 characters obtained from coded length mutations. Maximum parsimony and Bayesian analyses of the complete dataset yielded congruent, fully resolved and well-supported trees. Our data confirm the monophyly of the Cabombaceae but do not provide convincing support for the monophyly of Nymphaeaceae with respect to Nuphar . Moreover, the genus Nymphaea is inferred to be paraphyletic with respect to Ondinea , Victoria and Euryale . In fact, the Australian endemic Ondinea forms a highly supported clade with members of the Australian Nymphaea subgenus Anecphya . In addition, Victoria and Euryale are inferred to be closely related to a clade comprising all night-blooming water-lilies ( Nymphaea subgenera Hydrocallis and Lotos ). An experimental approach showed taxon sampling to be of influence on the nodes reconstructed in core Nymphaeaceae.
