A series of 2,1,3- and 1,2,4-benzothiadiazine derivatives (BTDs) were synthesized and evaluated for their inhibitory activity versus enzymatic isoforms PDE3, PDE4 and PDE7. The compounds characterized by the 3,5-di-tert-butyl-4-hydroxybenzyl moiety at N1 position of 2,1,3-benzothiadiazine core (8, 13, 18), were found active and selective at micromolar level versus PDE4 and could be studied as new leads for the treatment of asthma and COPD (Chronic Obstructive Pulmonary Disease). The antioxidant activity evaluation on the same compounds highlighted 13 as the most significative. Molecular modelling studies gave further support to biological results and suggested targeted modifications so as to improve their potency.  相似文献

The recently discovered Nesfatin-1 plays a role in appetite regulation as a satiety factor through hypothalamic leptin-independent mechanisms. Nesfatin-1 is co-expressed with Melanin-Concentrating Hormone (MCH) in neurons from the tuberal hypothalamic area (THA) which are recruited during sleep states, especially paradoxical sleep (PS). To help decipher the contribution of this contingent of THA neurons to sleep regulatory mechanisms, we thus investigated in rats whether the co-factor Nesfatin-1 is also endowed with sleep-modulating properties. Here, we found that the disruption of the brain Nesfatin-1 signaling achieved by icv administration of Nesfatin-1 antiserum or antisense against the nucleobindin2 (NUCB2) prohormone suppressed PS with little, if any alteration of slow wave sleep (SWS). Further, the infusion of Nesfatin-1 antiserum after a selective PS deprivation, designed for elevating PS needs, severely prevented the ensuing expected PS recovery. Strengthening these pharmacological data, we finally demonstrated by using c-Fos as an index of neuronal activation that the recruitment of Nesfatin-1-immunoreactive neurons within THA is positively correlated to PS but not to SWS amounts experienced by rats prior to sacrifice. In conclusion, this work supports a functional contribution of the Nesfatin-1 signaling, operated by THA neurons, to PS regulatory mechanisms. We propose that these neurons, likely releasing MCH as a synergistic factor, constitute an appropriate lever by which the hypothalamus may integrate endogenous signals to adapt the ultradian rhythm and maintenance of PS in a manner dictated by homeostatic needs. This could be done through the inhibition of downstream targets comprised primarily of the local hypothalamic wake-active orexin- and histamine-containing neurons.  相似文献

AtHAK5是拟南芥高亲和性钾转运体,其基因表达受低钾条件强烈诱导,编码蛋白在低钾条件下可以整合到质膜.生物信息学分析发现其氨基酸序列含有多处潜在的磷酸化位点.本研究假设这些位点对于AtHAK5的功能至关重要,为探讨AtHAK5的功能位点,分别将AtHAK5 cDNA和带有13种不同点突变位点的AtHAK5转化到athak5突变体中,获得14种稳定表达的转基因植株.利用athak5突变体根对Cs敏感的表型,最终确定T549A和T666A为非核心磷酸化位点.如下11个位点为AtHAK5功能必需位点:F85L,T86A,T311A,T359A,P551S,D552N,S603A,S604A,K668E,S698A和V713L.  相似文献

中华穿山甲(Manis pentadactyla)属于全球极度濒危物种,也是我国一级保护动物。对中华穿山甲的非法捕杀曾导致其种群数量锐减。但是,近年来相关研究报道较少,穿山甲分布状况不明,极大地制约了对该物种的有效保护。搜集了近年来国内中华穿山甲的救护记录和救护新闻,甄别出67个记录分布点,利用最大熵模型软件(MaxEnt)进行因子筛选,结果表明最冷季度降水量、人口密度、年降水量、坡度、坡向、海拔等6个环境变量是与中华穿山甲分布显著相关的影响因子。基于6个主导环境变量构建的MaxEnt模型AUC平均值为0.961±0.014,预测结果达到极好标准。刀切法(Jackknife)表明,其中最冷季度降水量、年降水量、人口密度和海拔是影响中华穿山甲分布的主要因素。中华穿山甲适宜生境(出现概率大于0.498)具有以下特点:最冷季度降水量141.22-439.46 mm,年降水量1471.67-2386.56 mm,人口密度≥390人/km2,海拔<316.98 m。该模型预测中华穿山甲在我国的潜在分布适宜区主要位于我国长江以南地区,总面积约为74.27×104 km2,占国土面积的7.73%,主要集中在江西、广东、湖南和广西省,面积分别占该区域的97.58%,89.65%,76.90%和73.08%;其次是浙江、福建、台湾和安徽省。湖北、江苏、四川、云南、贵州等省份也有中华穿山甲的零星分布。湖北东南部、江苏南部、浙江西南部和福建西北部等与江西接壤的区域也是中华穿山甲的重要潜在分布适宜区。明确中华穿山甲的潜在分布适宜区,可为该物种的种群保护和栖息地管理提供科技支撑。  相似文献

目的:了解铅锌镉联合染毒对大鼠血液系统的影响及营养干预对其损伤的修复作用。方法:选择SPF级初断乳Wistar大鼠72只,随机分为对照组、染毒组和干预组,分别采用生理盐水、铅锌镉联合染毒液及染毒后以营养干预液灌胃28天和56天之后,检测其血液系统中五元素和血细胞的指标。结果:染毒组较对照组大鼠血铜、血锌含量高,血钙含量低于对照组,差异均有统计学意义(P〈0.05);染毒组血铜含量高于干预组,血钙含量低于干预组,差异均有统计学意义(P〈0.05);干预组红细胞(RBC)计数、血红蛋白(Hb)、血细胞比容(HCT)均高于染毒组,差异均有统计学意义(P〈0.05);对照组白细胞(WBC)计数高于染毒组、干预组,差异均有统计学意义(P〈0.05)。结论:铅镉对大鼠血铜、血钙、血锌水平有影响;综合营养干预对重金属元素造成的血液系统损伤有明显的拮抗作用,对血液系统有一定的保护及修复作用。  相似文献

Climate warming and habitat transformation are widely recognized as worrying threatening factors. Understanding the individual contribution of these two factors to the change of species distribution could be very important in order to effectively counteract the species range contraction, especially in mountains, where alpine species are strongly limited in finding new areas to be colonized at higher elevations. We proposed a method to disentangle the effects of the two drivers of range change for breeding birds in Italian Alps, in the case of co‐occurring climate warming and shrub and forest encroachment. For each species, from 1982 to 2017, we related the estimated yearly elevational distribution of birds to the correspondent overall average of the daily minimum temperatures during the breeding season and the estimated amount of shrubs and forest cover. Using a hierarchical partitioning approach, we assessed the net contribution (i.e., without the shared effect) of each driver. Both temperature and shrub and forest cover showed a positive trend along the time series and resulted the most likely causes of the significant elevational displacement for 21 of the 29 investigated birds. While shrub and forest cover was found to be an important driver of the expansion of forest bird range toward higher elevations, the effect of temperature on favouring the colonization of previously climatically unsuitable forests at higher elevations was not negligible. Shrub and forest expansion resulted the main driver of the range contraction for edge and open habitat species, which suffered a distribution shrinkage at their lower elevational boundary. In light of climate warming, these results highlighted how the net range loss for edge and open habitat species, caused by shrub and forest encroachment consequent to land abandonment, should be counteracted by implementing proper conservation management strategies and promoting sustainable economic activities in rangeland areas.  相似文献

The aim of this work was to develop and characterize chitosan/gelatin films as innovative mucoadhesive system for buccal delivery of propranolol hydrochloride. FT-IR and TGA analysis confirmed the interaction between chitosan and gelatin. The presence of higher chitosan amounts in chitosan/gelatin films allowed the lowest percent water-uptake ability (235.1 ± 5.3%) and the highest in vivo residence time in the buccal cavity (240 ± 13 min). Moreover, the presence of mannitol in the formulation allowed 80% drug permeation through porcine buccal mucosa in 5 h. This behaviour suggests that the application of four and two films containing 5 mg of propranolol hydrochloride could be suitable for achieving the proposed daily dose for hypertension and atrial fibrillation treatment, respectively. Another interesting aspect of chitosan/gelatin films was their compatibility with buccal microflora in the absence of drug and their ability to determine growth inhibition for pathogen bacteria, but not for probiotic species, when loaded with drug.  相似文献

In this work we evaluate the potential grazing impact of the invasive reef-forming polychaete Ficopomatus enigmaticus in a SW Atlantic coastal lagoon (Mar Chiquita coastal lagoon; 37° 40' S, 57° 23' W; Argentina). This gregarious species feeds on suspended detritus and phytoplankton. Given the large area dominated by reefs, suspension feeding by this species might reduce the lagoon phytoplankton concentration and even hinder local eutrophication. To evaluate this hypothesis in situ replicated mesocosm experiments were performed in spring 2005 and, summer and winter 2006. Mesocosms enclosing reefs and without reefs were installed and grazing intensity was measured as the difference in chlorophyll a concentration and turbidity between the reef-treatment and the treatment without reefs. Reefs of F. enigmaticus decreased the mean chlorophyll a concentration, more during summer (56% decrease) than spring (25% decrease) and winter (19% decrease). Reefs also decreased water turbidity during summer (54% decrease) and spring (23% decrease). While previous studies indicate that the physical structure of these reefs alters water flow increasing water turbidity, our evidences show that their suspension-feeding activity can counteract this effect. Water turbidity was positively correlated with chlorophyll a concentration, which suggests that phytoplankton grazing by F. enigmaticus decreased light attenuation through the water column, with a potential for enhancement of benthic primary productivity. Therefore, our results suggest that grazing by this polychaete affects overall estuarine primary production as well as the relative importance of planktonic and benthic carbon sources to higher trophic levels.  相似文献
