A design equation is presented for packed-bed reactors containing immobilized enzymes in spherical porous particles with internal diffusion effects and obeying reversible one-intermediate Michaelis-Menten kinetics. The equation is also able to explain irreversible and competitive product inhibition kinetics. It allows the axial substrate profiles to be calculated and the dependence of the effectiveness factor along the reactor length to be continuously evaluated. The design equation was applied to explain the behavior of naringinase immobilized in Glycophase-coated porous glass operating in a packed-bed reactor and hydrolyzing both p-nitrophenyl-alpha-L-rhamnoside and naringin. The theoretically predicted results were found to fit well with experimentally measured values.  相似文献

Enzyme immobilization in the form of fiber and paper was easily achieved by wet spinning of aqueous admixture of sodium alginate and enzymes into divalent metallic ion solution as a coagulating bath, followed by paper making of resultant shortly cut fibers. Entrapment yields of enzymes used, e.g., glucoamylase, cyclodextrin glucanotransferase, endo-polygalacturonase, and protease, were always higher in calcium alginate fibers and their papers than those in corresponding beads. It was found that the yields increased with an increase of the discharge rate through the spinning nozzle because the higher discharge rate could provide more highly oriented metal-chelate linear polymer molecules along the fiber axis for preventing leakage of entrapped enzymes. Divalent metallic ions affected greatly the entrapment of glucoamylase in alginate fibers, the order of which followed roughly the ionotropic series of Thiele. Entrapment of glucoamylase in bicomponent systems comprising alginate and other water-soluble polymers was also investigated.  相似文献

The growth inhibitory and lethal effects of ethanol on Escherichia coli BB were investigated in batch cultures, by measuring total cell number, viable cell number, and cell mass concentration. Ethanol below ca. 50 g/L allowed exponential growth but depressed the specific growth rate. The effect of ethanol on the specific growth rate appeared to follow noncompetitive inhibition kinetics with apparently cooperative binding with a Hill coefficient of 2.5. The Hill coefficient and the inhibition constant were temperature independent over the range tested. Ethanol at 30 g/L decreased the growth yield. Ethanol enhanced the specific death rate in an experimental way. Stationary cell populations were more resistant than exponential ones but the degree of enhancement by ethanol was the same in both populations. Isopropanol and propanol also enhanced the specific death rate exponentially and the degree of enhancement was correlatedwith their membrane-buffer partition coefficients.  相似文献

The effects of growing Clostridium acetobutylicum NCIB 8052 in a chemostat under conditions of glucose, NH(4) (+), PO(4) (3-), Mg(2+), and Fe(2+) limitation were examined. It was noted that limitation of any major nutrient resulted in the same fermentation pattern. Conditions where minor nutrient levels were reduced appeared to stimulate solvent production. Under conditions of Mg(2+) restriction, a productive solventogenic culture was produced, with a yield of total solvents on glucose of 35.5%.  相似文献

Three anaerobic fluidized bed reactors at 37 degrees C were utilized to observe the effects of startup and operational procedures on biomass attachment. Using a meat-based synthetic waste and stepped-loading regime, the influences of synthetic polymer addition and maintenance of anaerobiosis during startup were investigated. Subsequently, increasing bed expansions were applied to assess shear effects. Synthetic polymer addition enhanced biomass retention but did not improve process performance. Maintenance of a reduced environment ameliorated fluctuating process parameters during start up and aided biomass retention and substrate removal. A bed expansion of 5% was detrimental to biomass attachment and COD removal but system stability was maintained at expansions between 10% and 30%. Startup was achieved in 56 days. Anaerobiosis appeared to enhance the initial evolution of a stable, well-adapted microbial population, whereas polymer addition interfered with this. Moderate bed expansions had negligible effects on attachment and performance.  相似文献

Deficiencies encompassing part or all of the X heterochromatin of Drosophila melanogaster have been linked to three abnormalities in male meiosis and spermatogenesis: X-Y nondisjunction, skewed sperm recovery ratios favoring sperm with reduced chromatin content, and sterility in males carrying either a Y-autosome translocation or mal +Y. In this study, 18 X heterochromatic deficiencies of varying sizes were tested in XY males for their spermatogenic phenotypes. All 18 proved to be either mutant for all three phenotypes or wild type for all three. Although variable among mutant deficiencies, expression levels of all three phenotypes were strongly correlated. Deficiencies that cause high levels of nondisjunction also cause severe recovery ratio distortion and are completely sterile in conjunction with mal+ Y. Low nondisjunction deficiencies cause comparable mild effects for the other phenotypes. The same deficiencies were also tested in males carrying a large heterochromatic free X duplication Dp(1; f)3. For all deficiencies which induce nondisjunction in XY males, the Y and free duplication pair regularly and the X fails to pair in XYDp males. Drive levels are constant across deficiencies in these males. Thus elimination of variability in the pairing phenotype also eliminates variability in sperm recovery ratios.  相似文献

Regulator of bithorax (Rg-bx)- [or trithorax (trx)-] lethal zygotes show anterior transformations of various cuticular features of the larval thorax and abdomen. The Rg-bx- lethal syndrome depends on the dosage of the bithorax gene complex (BX-C), and lack of Rg-bx+ function is antagonistic to posterior transformations displayed by Polycomb ( Pc)- embryos. Significantly, when the BX-C is deleted, the Rg-bx- embryos disclose homoeosis of mesothoracic to prothoracic cuticular structures. This homoeotic transformation is due to a reduction in Antennapedia (Antp)+ gene activity and is consequently dependent on the dosage of the Antennapedia gene complex (ANT-C), suggesting that the Rg-bx+ activity is necessary for proper expression of the Antp+ gene. However, the functional relationship between the Rg-bx and Sex combs reduced (Scr) loci in embryogenesis is still to be established.  相似文献

Perspective display techniques are applied to chemical and biochemical data sets. These represent spatially distributed gradients of reactive compounds that participate in pattern-formation processes due to reaction-diffusion or reaction-convection coupling. The patterns form in thin solution layers and are observed as chemical waves in the Belousov-Zhabotinskii reaction, as convection-induced stationary structures during oscillating glycolysis in yeast cytoplasm, and as the diffusive spreading of enzyme-catalyzed metabolic turnover in a substrate layer. The digital data are measured with a two-dimensional spectrophotometer based on a computerized video equipment with high spatial, temporal and intensity resolution. By application of three-dimensional procedures detailed structural properties of chemical and biochemical model systems will be presented yielding localization of reaction and transport events.  相似文献

For the first time, the 31P nuclear magnetic resonance technique has been used to study the properties of isolated vacuoles of plant cells, namely the vacuolar pH and the inorganic phosphate content. Catharanthus roseus cells incubated for 15 hours on a culture medium enriched with 10 millimolar inorganic phosphate accumulated large amounts of inorganic phosphate in their vacuoles. Vacuolar phosphate ions were largely retained in the vacuoles when protoplasts were prepared from the cells and vacuoles isolated from the protoplasts. Vacuolar inorganic phosphate concentrations up to 150 millimolar were routinely obtained. Suspensions prepared with 2 to 3 × 106 vacuoles per milliliter from the enriched C. roseus cells have an internal pH value of 5.50 ± 0.06 and a mean trans-tonoplast ΔpH of 1.56 ± 0.07. Reliable determinations of vacuolar and external pH could be made by using accumulation times as low as 2 minutes. These conditions are suitable to follow the kinetics of H+ exchanges at the tonoplast. The 31P nuclear magnetic resonance technique also offered the possibility of monitoring simultaneously the stability of the trans-tonoplast pH and phosphate gradients. Both appeared to be reasonably stable over several hours. The buffering capacity of the vacuolar sap around pH 5.5 has been estimated by several procedures to be 36 ± 2 microequivalents per milliliter per pH unit. The increase of the buffering capacity due to the accumulation of phosphate in the vacuoles is, in large part, compensated by a decrease of the intravacuolar malate content.  相似文献

The activity of superoxide dismutase was investigated in needles of spruce trees. To obtain maximum activity, needles were homogenized in the presence of Triton X-100 and polyvinylpyrrolidone. Superoxide dismutase activity was measured in dialyzed extracts with a modified epinephrine assay (HP Misra, I Fridovich [1972] J Biol Chem 247: 3170-3175) at pH 10.2. The extracts contained 70 to 120 units of superoxide dismutase per milligram protein. One unit of superoxide dismutase was completely inhibited in the presence of 20 micromolar NaCN. On native polyacrylamide gels three electromorphs were visualized after staining for activity. All three species were sensitive to CN and H2O2 and were therefore assumed to be Cu/Zn-superoxide dismutases. Superoxide dismutase activity was dependent on the age of the needles and declined by approximately 25% within 3 to 4 years.  相似文献
