Identification of N5-methyl-N5-formyl-2,5,6-triamino-4-hydroxypyrimidine as a major adduct in rat liver DNA after treatment with the carcinogens, N,N-dimethylnitrosamine or 1,2-dimethylhydrazine

Human Tamm-Horsfall urinary glycoprotein from an individual of the blood group Sd(a+) phenotype was tritium-labelled by treatment with galactose oxidase and sodium boro[3H]hydride and was then digested with endo-beta-galactosidase. A series of dialysable, labelled fragments was released from which a pentasaccharide was isolated that strongly inhibited the agglutination of Sd(a+) red cells by human anti-Sda serum and hence contained the Sda determinant structure. Reduction, methylation analysis and sequential exo-glycosidase digestion established the structure of the pentasaccharide as: GalNAc beta(1 leads to 4)[NeuAc(2 leads to 3)]Gal beta(1 leads to 4)GlcNAc beta(1 leads to 3)Gal     

Effect of specific antibodies on active immunity development in those inoculated with an antirabies vaccine

The study of the effect of gamma globulin introduced in different doses (0.5 and 0.25 ml/mg) in combination with Fermi rabies vaccine (observations on humans were made) and with cerebral rabies vaccine inactivated by UV irradiation (in animal experiments) demonstrated that the injection of the higher doses of gamma globulin resulted in lower geometrical mean of antibody titers. Therefore, in combined administration of rabies vaccine and gamma globulin for postexposure rabies prevention it is advisable to reduce the dose of gamma globulin by one-half.     

Assignment of the human homologue of Pim-1, a mouse gene implicated in leukemogenesis,to the pter-q12 region of chromosome 6

Summary Viral leukemogenesis in mice is frequently initiated by proviral activation of a highly conserved cellular gene called Pim-1. Here we report the chromosomal localization of the human homologue by Southern blot analyses of DNAs obtained from human-rodent somatic cell hybrids. The single copy human homologue was assigned to the 6pter-q12 segment.     

Oxidative degradation of thymine with O2 promoted by L-ascorbic acid and Cu(II) ion

Oxidation of thymine with O2 was promoted by copper(I) ion generated from reaction of L-ascorbic acid (AA) with copper (II) ion. The main oxidation products were thymine glycol (TG) and N-formyl-N'-pyruvylurea (FPU). At higher concentration of O2, formation of FPU was favored, while TG was dominant at higher Cu(II) ion and lower O2 concentrations. Reaction mechanism involving hydroxy thyminyl radical was proposed.     

A numerical taxonomic study of Flavobacterium-Cytophaga strains from dairy sources

Phenotypic data on 203 Gram-negative non-fermentative bacteria of the Flavobacterium-Cytophaga group isolated from milk and butter were analyzed by numerical taxonomic techniques. Twenty reference strains including species of Flavobacterium, Cytophaga and strains of Pseudomonas paucimobilis were included in the study. Using the matching coefficient of Sokal & Michener with antibiotic susceptibility data included, 139 isolates were recovered in nine clusters. Six of these clusters were linked at or above the 85% S level while three were linked at or above the 79% S level. The largest cluster, representing 46.3% of the isolates, could be equated with Flavobacterium sp. Group IIb. Other clusters could be equated with Flavobacterium sp. L 16/1 (22.7% of isolates), F. balustinum (10.8% of isolates), F. breve (4.4%), F. multivorum (3.5%) and Cytophaga johnsonae (1.5%). The cluster resembling Flavobacterium sp. L 16/1 and a smaller unclassified cluster, were exceptional in being susceptible to the antibiotics cephalothin and penicillin G.     

Electron microscope study of the structures of the Bacillus subtilis prophages, SPO2 and phi105

Circular duplex structures of the correct length are observed in the electron microscope in hybridization mixtures of lysogen DNA and mature phage DNA for the case of the temperate Bacillus subtilis bacteriophage SPO2. This result shows that the sequence order of the prophage is a circular permutation of that of the mature phage. By making heteroduplexes of prophage DNA with that of the SPO2 deletion mutants, R90 and S25, the att site of the phage has been mapped at 61.2 ± 0.6% from one end of the mature phage DNA, which has a length of 38,600 base pairs. In the same co-ordinate system, the R90 deletion extends from 58.9 ± 0.7 to 66.8 ± 0.8% on the SPO2 chromosome, whereas the S25 deletion extends from 63.2 ± 0.6 to 66.9 ± 0.7%. In similar experiments with lysogen and mature phage DNA's of the temperate B. subtilis phage, φ105, no circular structures were seen. This result shows that the sequence order in the prophage and the phage are colinear, without circular permutation.
