Establishment of human tumor strains from cell lines transplanted into athymic mice and rats

The authors describe human cancer strains established from 6 cell lines transplanted to nude mice and rats: cancer of the liver, colon, lung, bladder and Burkitt's lymphoma. In spite of a long history (for several years) of cell line transfers in vitro, on transplantation of tumor cells, nude animals established tumors histologically identical to the primary cancer.     

Antibiotics in surgical treatment of acute abscesses.

A four-way, double-blind, prospective trial of treatment of abscesses by incision, curettage, and primary closure with and without antibiotic cover (clindamycin injection before operation or capsules after operation, or both) was conducted. There was no appreciable difference in mean healing time between the patients given both the antibiotic injection and the antibiotic capsules and those given the injection and placebo capsules, whereas healing times in those given the placebo injection and antibiotic capsules or placebo only were appreciably longer. Four of the patients who were not given the antibiotic injection developed bacteraemia; one patient who was given the antibiotic injection also developed a bacteraemia, but this was caused by clindamycin-resistant bacteria. These results show that a single injection of an effective antibiotic before operation is sufficient to protect the patient against bacteraemia and permit optimum healing.     

Developmental patterns of galactosyltransferase activity in various regions of rat brain

Abstract: The developmental pattern of glycoprotein-galactosyltransferase activity was determined in the microsomal fractions of three regions of the embryonic rat brain and in parts of the visual system and the cerebellum postnatally. It could be shown that the enzyme activity was highest in the embryonic brain, where regional differences were apparent, and decreased progressively after birth. The enzyme profile in the cerebellum showed no marked postnatal changes.     

Effects of Patch-Burn Management on Dickcissel Nest Success in a Tallgrass Prairie

Abstract Grassland birds have declined more than any other North American habitat-associated bird community. Because most species of grassland birds evolved within heterogeneous landscapes created by the interaction of fire and grazing, traditional rangeland management that promotes homogeneity, including annual dormant-season burning combined with early-intensive grazing, might be partly responsible for these declines, especially in some regions of the Great Plains, USA. Recently, an alternative grassland management practice known as patch-burning has been promoted as a means of restoring heterogeneity to grasslands by mimicking the grazing-fire interaction that once occurred on the prairie before European settlement. From 2003 to 2004, we examined effects of patch-burning and traditional management (annual burning followed by early-intensive grazing) on the reproductive success of dickcissels (Spiza americana) in tallgrass prairie in Oklahoma. We monitored 296 dickcissel nests and found that dickcissel nesting phenology differed between traditional and patch-burned pastures. Specifically, dickcissels tended to initiate their nests later in the traditional pasture. Mean number of eggs laid and fledglings produced were similar between the treatments, but nest densities were higher in traditional pastures. Predation was the predominant cause of nest failure and was higher in traditional pastures than in patch-burned pastures. Brown-headed cowbird (Molothrus ater) parasitism was higher in traditional pastures than in patch-burned pastures. Overall, dickcissel nest success was higher in patch-burned pastures than in traditional pastures. The positive response of dickcissel nest success to patch-burn management provides further evidence that this practice can be a useful tool for grassland bird conservation. By creating a mosaic of different stature vegetation, patch-burn management enhances productivity of grassland bird species by providing a refuge area in the unburned patches that affords dickcissels and other nesting grassland birds some protection from the direct (e.g., trampling) and indirect (e.g., cowbird parasitism and predation) effects of grazing, which are not available under traditional management. Patch-burn management should be encouraged as a conservation strategy for grassland birds throughout the Great Plains.     

Observing the Temperature Dependent Transition of the GP2 Peptide Using Terahertz Spectroscopy

The GP2 peptide is derived from the Human Epidermal growth factor Receptor 2 (HER2/nue), a marker protein for breast cancer present in saliva. In this paper we study the temperature dependent behavior of hydrated GP2 at terahertz frequencies and find that the peptide undergoes a dynamic transition between 200 and 220 K. By fitting suitable molecular models to the frequency response we determine the molecular processes involved above and below the transition temperature (T _D). In particular, we show that below T _D the dynamic transition is dominated by a simple harmonic vibration with a slow and temperature dependent relaxation time constant and that above T _D, the dynamic behavior is governed by two oscillators, one of which has a fast and temperature independent relaxation time constant and the other of which is a heavily damped oscillator with a slow and temperature dependent time constant. Furthermore a red shifting of the characteristic frequency of the damped oscillator was observed, confirming the presence of a non-harmonic vibration potential. Our measurements and modeling of GP2 highlight the unique capabilities of THz spectroscopy for protein characterization.     

Precise identification of gene products in hearts after in vivo gene transfection, using Sendai virus-coated proteoliposomes.

Both efficient gene transfer and the exact identification of gene product are required for gene therapy. Gene transfection of green fluorescence protein (GFP) might be useful for the reporter. After in vivo cotransfection of GFP and beta-galactosidase (beta-Gal) genes in Sendai virus-coated proteoliposomes to rat hearts, we compared the sensitivity and specificity of three methods: GFP detection, histochemical staining (HC) of beta-Gal activity, and immunostaining (IS) of the beta-Gal protein. Fluorescence microscopy and double staining of HC and IS revealed that both GFP and IS were equally sensitive and fourfold superior to HC at the peak of gene expression. However, different from skeletal muscle, the GFP of transfected cardiomyocytes showed two demerits: the fluorescence quenching due to the intense staining of beta-Gal activity, and nonspecific autofluorescence from myocardium. Thus, specific IS would be so far the most reliable to identify the gene product in heart.
