Erythrocyte membrane Ca2+ ATPase: reactivities of human A, AS, and S erythrocytes with steroid hormones

In a preliminary study we reported a significant enhancement of Ca2+ ATPase activity in sickle cell membranes in the presence of progesterone and testosterone. In this work the reactivity of various classes of steroid hormones with the membranes of hemoglobin variants was investigated. A consistent universal stimulation of Ca2+ ATPase activity in sickle cell membranes by the different classes of steroid hormones does not appear to correlate with any major structural differences of the hormones or the presence of reactive functional groups. The universal interaction of the hormones with sickle cell membranes probably enhances Ca2+ efflux through the Ca2+ ATPase without directly affecting the characteristics of the pump.     

Is prostaglandin E the neural mediator of the febrile response? The case against a proven obligatory role.

We have reviewed the evidence in favor of a prostaglandin mediator of the thermal responses in fever and found that PGE injected into the hypothalamus does not always cause fever, that cerebrospinal fluid concentrations of PGE are not reliable reflections of hypothalamic events, and that antipyretic drugs may act in ways other than inhibiting PGE synthesis. Fever is not blocked by prostaglandin antagonists, nor by ablation of PGE-sensitive areas of the brain. There is poor correlation between the effects of pyrogens and of PGE on cerebral neurons. There is evidence that at least one prostanoid other than prostaglandin is a mediator of fever, but the prostanoid has not been identified yet. We conclude that PGE may contribute to the neural responses in fever but is not essential.     

Effects of industrial respirators on breathing pattern at different work levels

The effects of a filtering device and an air-line apparatus on breathing pattern were studied in healthy men with different physical characteristics and work capacity. The subjects comprised nine construction workers aged 35-44, and nine firemen aged 21-35. The construction workers' mean maximal oxygen consumption (VO2max) was 34.5 ml min-1 kg-1, the firemen's 66.9 ml min-1 kg-1. Breathing pattern was analyzed for its components, inspiratory time, expiratory time, breathing frequency, tidal volume, and pulmonary ventilation at rest, during two submaximal treadmill walks when the subjects' absolute work load was equal, and during recovery. Neither the filtering device nor the air-line apparatus had a significant effect on breathing pattern when compared with the control values measured twice with a low-resistance breathing valve. A significantly longer expiratory time, lower breathing frequency, and smaller pulmonary ventilation were found for the firemen with the breathing valve and the industrial respirators. The breathing pattern of the construction workers and the firemen differed, but the alterations were not induced by the use of the filtering device or the air-line apparatus when studied at aerobic work levels up to 60% VO2max.     

Extracellular potassium concentration in normal and denervated hind limb muscles of the rat

K+-sensitive electrodes with a side pore were used for measuring extracellular potassium levels in control muscles and in the gastrocnemius, extensor digitorum and soleus muscles of the rat, one, 8 and 14 days after denervation. In all cases, the extracellular potassium level was close to 5 mmol.1-1. There were no substantial differences between muscles of different functional and morphological types and between control and denervated muscles. Elevated extracellular potassium cannot therefore be the reason for increased sensory outflow of impulses observed after motor denervation.     

Southern analysis of BT-R 1, the Manduca sexta gene encoding the receptor for the Cry1Ab toxin of Bacillus thuringiensis

Various subspecies of the gram-positive bacterium Bacillus thuringiensis are known to produce a wide array of insecticidal crystal proteins (ICPs) upon sporulation. These ICPs act primarily on the brush border of midgut epithelial cells of susceptible larvae. Recently, a protein of 210?kDa, isolated from the midgut of Manduca sexta, has been demonstrated to bind the Cry1Ab toxin produced by B. thuringiensis subsp. berliner and is therefore postulated to be involved in mediating the toxicity of Cry1Ab. The cDNA encoding the 210?kDa protein, termed BT-R₁ (Bacillus thuringiensis receptor-1), was recently cloned, and shows limited homology to the cadherin superfamily of proteins. Quite naturally, there is a great deal of interest in the characterization of BT-R ₁ , the gene encoding the 210?kDa Cry1Ab binding protein. The studies presented here involve the use of various restriction fragments prepared from the cDNA encoding BT-R₁ as probes of Southern blots bearing M. sexta genomic DNA cleaved with a variety of restriction endonucleases. These Southern blot data reveal that there are two discrete regions within the M. sexta genome which encode sequences homologous to BT-R₁. On the basis of the signal intensities seen on Southern blots, it appears that only one of these genes encodes BT-R₁, whereas the other is a closely related homologue.     

Stimulation of 32Pi transport into human erythrocyte ghosts and reconstituted vesicles by Mg2+ and hemoglobin

Human erythrocyte ghosts catalyze a low rate of 32Pi uptake. A severalfold stimulation of 32Pi uptake was observed after exposure of the membranes to an erythrocyte lysate or to hemoglobin in the presence of Mg2+. Ghosts prepared from erythrocytes that had been exposed to 10 microM 4,4'-diisothiocyanostilbene-2,2'-disulfonic acid showed a marked reduction in 32Pi uptake. Reconstitution of membranes with added phospholipids by freezing and thawing, by octylglucoside dilution or by cholate dialysis, yielded vesicles that catalyzed 32Pi uptake. When membranes were incubated with hemoglobin and Mg2+ prior to reconstitution, the rate of uptake was increased severalfold. The inhibition of hemoglobin and Mg2+-dependent uptake of 32Pi by chloride suggests that the transport in the reconstituted vesicles is catalyzed by the classical inorganic anion transporter.     

New data on small mammals in the Kazakh Uplands

Small mammals were studied in the Kazakh Uplands in the spring and fall of 2008. The trapping studies revealed 10 species. Abundances of the animals were low in the four main distinct characteristic biotopes of Bayanaul National Park, but those of biotope dominants were high. In the Kazakh Uplands, rodents and insectivores are clearly restricted to certain biotopes. Biodiversity indices for small mammal communities are low, indicating that the community structure is disturbed.     

Interferon and cytotoxic factor (cytotoxin) released in the blood of mice infected with Mycobacterium bovis BCG. III. Interferon and cytotoxin induced by the specific antigen as compared with those induced by bacterial lipopolysaccharide

The time course of development and decline of the ability of BCG-infected mice to produce interferon in the serum in response to the intravenous infection of purified protein derivative of tuberculin (PPD) was very similar to that of their systemic hypersensitivity to PPD. A cytotoxic factor (cytotoxin) was produced in parallel with interferon in the serum of BCG-infected mice after stimulation with PPD. The duration of the period in which cytotoxin-production responsiveness to PPD was definitely detectable was much shorter than that for interferon-production responsiveness although the periods for the maximum production of interferon and cytotoxin coincided. The kinetics of release of interferon in the serum of BCG-infected mice after stimulation with PPD did not parallel that of release of cytotoxin. The four kinds of activities, interferons and cytotoxins induced by PPD and lipopolysaccharide (LPS) in the serum of BCG-infected mice, were compared for their stability to heating at 56 C and to treatment at pH 2. The kinetics of inactivation of these four activities differed significantly, when the serum was either heated at 56 C or treated at pH 2. Interferon produced in response to LPS could be neutralized by anti-L cell(NDV) interferon rabbit serum as easily as L cell (NDV) interferon, 16 times as much antiserum was required to neutralize the same amount of interferon in response to PPD, but cytotoxins induced by PPD and LPS were not neutralized at all by the antiserum. From these findings it is thought likely that interferons and cytotoxins induced by PPD and LPS in the serum of BCG-infected mice are different substances, although the antigenic relationship between cytotoxins induced by PPD and LPS remains unknown.
