Brachypodium distachyon (Brachypodium) has been proposed as a model temperate grass because its physical, genetic, and genome attributes (small stature, simple growth requirements, small genome size, availability of diploid ecotypes, annual lifecycle and self fertility) are suitable for a model plant system. Two additional requirements that are necessary before Brachypodium can be widely accepted as a model system are an efficient transformation system and homogeneous inbred reference genotypes. Here we describe the development of inbred lines from 27 accessions of Brachypodium. Determination of c-values indicated that five of the source accessions were diploid. These diploid lines exhibit variation for a variety of morphological traits. Conditions were identified that allow generation times as fast as two months in the diploids. An Agrobacterium-mediated transformation protocol was developed and used to successfully transform 10 of the 19 lines tested with efficiencies ranging from 0.4% to 15%. The diploid accession Bd21 was readily transformed. Segregation of transgenes in the T 1 generation indicated that most of the lines contained an insertion at a single genetic locus. The new resources and methodologies reported here will advance the development and utilization of Brachypodium as a new model system for grass genomics.  相似文献

Potassium channels are one of the fundamental requirements for the generation of action potentials in the nervous system, and their characteristics shape the output of neurons in response to synaptic input. We review here the distribution and function of a high-threshold potassium channel (Kv3.3) in the electrosensory lateral line lobe of the weakly electric fish Apteronotus leptorhynchus, with particular focus on the pyramidal cells in this brain structure. These cells contain both high-threshold Kv3.3 channels, as well as low-threshold potassium channels of unknown molecular identity. Kv3.3 potassium channels regulate burst discharge in pyramidal cells and enable sustained high frequency firing through their ability to reduce an accumulation of low-threshold potassium current.W. Infrared, far-infrared and Raman spectra of Re2(O2CCH3)4X2 (X = Cl, Br) and Re2(O2CCD3)4Cl2 have been recorded. Assignments of the vibrational spectra of Os2(O2CCH3)4Cl2 and its deuterated derivative have been completed together with the Re complexes on the basis of normal-coordinate analysis. Force constant calculation was made for the acetate ion as well as for a four-atomic unit (with the CH3 and CD3 groups considered as point masses) using optimized masses of 16.7, 17.8, 20.5 and 21.6 for 12CH3, 13CH3, 12CD3 and 13CD3 groups, respectively. The force constants of the acetate ion have been adopted to the starting force field of the M2(O2CCH3)4X2 type complexes. The metal-halide (0.889, 0.997 and 1.286 N cm−1) and metal-metal stretching (3.32, 3.34 and 3.57 N cm−1) force constants were obtained for Re2(O2CCH3)4Cl2Re2(O2CCH3)4Br2 and Os2(O2CCH3)4Cl2 complexes, respectively. It was shown that the so-called diatomic approximation in most cases overestimates the M-M stretching force constants by 30-40%. Much better correlation has been obtained to fit these force constants, which produced values very close to those obtained by full normal-coordinate calculations. The Re-Re stretching force constants showed a reasonable correlation with the Re-Re bond distances for 18 rhenium complexes.  相似文献

A stem-girdling experiment was carried out on an evergreen conifer, the Korean pine (Pinus koraiensis Sieb. et Zucc.), in mid summer in Northeast China. A 50 % higher respiration rate at the upper part of the stem was observed 3 d after stem girdling, and a stable higher rate (1.2–2.8 times) one week later. However, no higher soluble sugar or starch contents were found in the upper bark of the girdled stems in measurements over three weeks. These findings indicate that most of the newly-formed photosynthates were consumed by the high respiratory activity; this is also implied by the strong correlation between the photosynthetic photon flux over the canopy (PPF) and respiration at the upper parts of girdled stems. Moreover, the maximum PPF and cumulative PPF one day before measurement (PPFmax-Y and CPPF-Y, respectively) were closely correlated with the respiratory difference between the upper and the lower parts, but no such correlation was found with the instantaneous PPF (PPF-I) and cumulative PPF on the current day from sunrise to measured time point (CPPF-C). This shows that photosynthates newly formed by canopy needles need at least one day for transportation in order to increase the stem respiration at tree breast height.  相似文献

The Effects of Bacterial Leaching on Metal Partitioning in Sewage Sludge   总被引:2,自引:0,他引:2  
The partitioning of Mn, Al, Zn, Cu and Ti ions in municipal sewage sludge was investigated before and after bioleaching processes effectuated by Acidithiobacillus ferrooxidans and Acidithiobacillus thiooxidans. Oxidation–reduction potential increase and pH decrease were obtained as a result of bacterial activity. A less pronounced and constant decrease was obtained with A. ferrooxidans, whereas A. thiooxidans presented a lag phase before a steep pH decrease. Metal solubilization was accomplished in experimental systems supplemented with energy source, Fe2+ for A. ferrooxidans and S0 for A. thiooxidans. Solubilization efficiency differed for each metal except for Al, and was relatively similar for either organism. Metal partitioning was conducted using a five-step sequential extraction procedure before and after the bioleaching. The results indicated that Zn and Mn ions were mostly associated with the organic fraction, whereas Cu, Al and Ti ions with the sulphide/residue fraction. The bioleaching process caused prompt solubilization of metals mostly associated with the more labile fractions (exchangeable, adsorbed and organically bound metals), whereas those associated to the less labile ones (EDTA and sulphide/residue fractions) were exchanged towards more labile fractions.  相似文献

This paper presents a first account of the salão vegetation of southwestern Amazonia, a frequent but poorly known formation and unusual in that it is a lowland moist tropical environment in which virtually all of the plant diversity is accounted for by herbs. The salões occur along canalized portions of the Purus and Juruá river basins, and their substrates consist of exposed parent materials that characterize much of this sector of Amazonia but are usually overlain by a relatively thin (<1 m) layer of soil. They are constantly moist seeps or ‘seepage banks,’ and the lower portions are seasonally submerged. Strata are recognizable in the vegetation, and the physiognomy and composition of each vegetation band of the salões is likely determined by the slope, the parent material, and (for lower bands) the duration of flooding. To date the salão flora has registered 66 species: 11 bryophytes, 12 ferns, and 43 angiosperms. Five of the bryophyte species, two ferns, and one grass, Arundinella berteroniana, are found in most if not all salões and can be considered indicators of this vegetation type, while most of the pteridophytes and other angiosperms occur more sporadically. None of the species is endemic to the formation and only four to the region; most are associated generally with the margins of rivers, lakes, and streams, often on rocky or sandy substrates. The floristic and conservation significance of this formation lie in its apparent affinities with montane regions, in the fact that 21 species and eight genera are recorded for the Acre flora only from the salões, and in its local value as a source of filtered potable water.  相似文献

Many Raphidophytes are important algal bloom-forming species. Morphology-based identification of these species is often ambiguous, however, as many species are very similar in shape and size. To accurately detect the presence of these species in pre-bloom conditions, single-cell PCR is probably the most rapid and convenient method. However, direct single-cell PCRs with conserved primers are apparently not effective, probably due to the impermeability of the cell wall. We report here an effective detergent-based pre-PCR cell lysis method, which turned out to be a critical step for effective single-cell PCR of the Raphidophytes. Two PCR-based methods, nested SC-PCR and SC-RAPD, were evaluated. The nested SC-PCR involves two consecutive PCRs, the first of which is performed with the D1 and D2 primers (external primers) resulting in an amplification of a partial LSU rRNA gene. The second amplification is performed with primers targeting the LSU domain and specifically annealing to Chattonella ovata and Chattonella marina only. The SC-RAPD performed with the established random primers, RP1–RP4, produced unique haplotypes that could be exploited to differentiate the two Chattonella species. The assay was demonstrated to be sensitive, with the lowest detection limit of a single Raphidophyceae cell. The method developed is a valuable tool for the study of intra-specific variations of the Raphidophytes and represents a platform for further development of species-specific SC-RAPD for all members of the Raphidophyceae.  相似文献
