Light-shade adaptation of the chlorophyll a/b containing procaryote Prochlorothrix hollandica was studied in semicontinuous cultures adapted to 8, 80 and 200 μmole quanta per square meter per second. Chlorophyll a contents based on dry weight differed by a factor of 6 and chlorophyll b by a factor of 2.5 between the two extreme light conditions. Light utilization efficiencies determined from photosynthesis response curves were found to decrease in low light grown cultures due to lower light harvesting efficiencies; quantum requirements were constant at limiting and saturating irradiances for growth. At saturating growth irradiances, changes in light saturated oxygen evolution rate originated from changes in chlorophyll a antenna relative to the number of reaction centers II. At light-limiting conditions both the number of reaction centers II and the antenna size changed. The amount of chlorophyll b relative to reaction center II remained constant. As in cyanobacteria, the ratio of reaction center I to reaction center II was modulated during light-shade adaptation. On the other hand, time constants for photosynthetic electron transport (4 milliseconds) were low as observed in green algae and diatoms. The occurrence of state one to two and state two to one transitions is reported here. Another feature linking photosynthetic electron transport in P. hollandica to that in the eucaryotic photosynthetic apparatus was blockage of the state one to two transition by 3-(3,4-dichlorophenyl)-1,1-dimethylurea. Although chlorophyll b was reported in association with photosystem I, the 630 nanometer light effect does not exclude that chlorophyll b is the photoreceptor for the state one to two transition.  相似文献

The effects of the stress of dystocia on the adrenocortical function post partum in cows (n = 6) requiring a cesarean section were assessed by an adrenocorticotropic hormone (ACTH) stimulation test. Nine cows that calved normally were used as controls. The plasma glucocorticoid levels, before and 60 min after an intramuscular injection of 25 IU ACTH, were 4.4 +/- 0.5 (mean +/- SD) and 21.5 +/- 2.4 ng/ml 1 d post partum, 1.9 +/- 0.3 and 18.4 +/- 2.9 ng/ml 4 d post partum and 2.7 +/- 1.1 and 14.6 +/- 3.3 ng/ml 8 d post partum, respectively. Corresponding values of glucocorticoids in cows with normal calving were 4.5 +/- 3.6 and 18.1 +/- 5.2 ng/ml 1 d post partum, 1.7 +/- 1.3 and 13.2 +/- 5.5 ng/ml 4 d post partum and 1.9 +/- 1.7 and 14.6 +/- 3.3 ng/ml 8 d post partum. There were no statistically significant differences of the values between cows with dystocia requiring a cesarean section and cows with normal calving. The results indicate that dystocia requiring a cesarean section like normal calving does not lead to significant depression of the adrenocortical function post partum.  相似文献

An adaptive state estimator for detecting contaminants in bioreactors   总被引:2,自引:0,他引:2  
An algorithm is presented for detecting the appearance of contaminants during batch or fed-batch fermentations, using only presently available on-line measurements. Its adaptive nature enables it to rely on almost no prior knowledge of the real process. The necessary on-line measurements are total biomass and its production rate; it is also shown how a physical variable such as oxygen uptake can be used alone instead. The algorithm's properties are studied theoretically and through simulations. These were confirmed by on-line experimental results, obtained with a Yeast culture, both pure and contaminated by a Bacteria. The algorithm does not detect contaminants when none are there, and it also provides a convergent estimate of a pure culture's specific growth rate. Contaminated cultures are recognized by the algorithm, and this detection can be made more or less conservative. After detection, the various estimates may diverge, due to general observability difficulties, though this divergence can itself be monitored. Moreover, the algorithm is easy to tune and its qualitative behavior is quite insensitive to its adjustable parameters. A practical criterion and scheme for implementation are proposed. The generality of the approach, which far exceeds the experimental system used, is finally discussed.  相似文献

The objective of this article is to propose an algorithm for the on-line estimation of the specific growth rate in a batch or a fed-batch fermentation process. The algorithm shows the practical procedure for the estimation method utilizing the macroscopic balance and the extended Kalman filter. A number of studies of the on line estimation have been presented. However, there are few studies discussing about the selection of the observed variables and for the tuning of some parameters of the extended Kalman filter, such as covariance matrix and initial values of the state.The beginning of this article is devoted to explain the selection of the observed variable. This information is very important in terms of the practical know-how for using technique. It is discovered that the condition number is a practically useful and valid criterion for number is a practically useful and valid criterion for choosing the variable to be observed.Next, when the extended Kalman filter in applied to the online estimation of the specific growth rate, which is directly unmeasurable, criteria for judging the validity of the estimated value from the observed data are proposed. Based on the proposed criterial, the system equation of the specific growth rate is selected and initial value of the state variable and covariance matrix of the system noises are adjusted. From many experiments, it is certified that the specific growth rate in the batch or fed -batch fermentation can be estimated accurately by means of the algorithm proposed here. In these experiments, that is, when the cell concentration is measured directly, the extended Kalman filter using the convariance matrix with a constant element can estimate more accurately values of the specific growth rate than the adaptive extended Kalman filter does.  相似文献

Several on-line optimizing control strategies were proposed and tested by computer simulation for the efficient operation of bioreactors. The control task was divided into two, one of which was to search for the optimal operating point and passed the set point to the lower layer of which task was to make the process output follow the set point as soon as possible. It was shown to be effective for the upper layer to express the objective function as a polynomial with respect to the measurement variable and to make use of it for finding the optimum point. Noting that the major dynamic characteristics of bioreactor system is the time-varying and nonlinear nature, the adaptive type control system is in evitable. It was shown to be quite effective to use discrete type self-tuning PID controller and the optimal controller compensated for the interaction between the control loops.Application was made to the cell recycle system for the production of lactic acid and baker's yeast cultivation. I was found from the former application that the control quality can be significantly improved by incorporating the decoupling strategy into the lower layer closed-loop system. It was also found from the latter application that the initial startup period can be significantly reduced by making use of the rough mathematical model.  相似文献

In this article a novel approach to the control of fermentation processes is introduced. A "physiological state control approach" has been developed using the concept of representing fermentation processes through the current physiological state of the cell culture. No conventional mathematical model is required for the synthesis of such a control system.The main idea is based on the fact that during batch, feed-batch, or even continuous cultivation the physiological characteristics of the cell population, jointly expressed by the term "physiological state", are not constant but rather variable, which is reflected in expected or unexpected changes in the behavior of the control plant, and which requires flexible alteration of the current control strategy. The proposed approach involves decomposition of the physiological state space into several subspaces called "physiological situations." In every physiological situation the cell population expresses stable characteristics, and therefore an invariant control strategy can be effectively applied. The on-line functions of the physiological state control system consist of the calculation of physiological state variables, determination of the current physiological situation as an element of a previously defined set of known physiological situations, switching of the relevant control strategy, and calculation of the control action. Attention is focused on the synthesis of the novel and nonstandard part of the control system - the algorithm for online recognition of the current physiological state. To this end an effective approach, based on artificial intelligence methods, particularly fuzzy sets theory and pattern recognition theory, was developed. Its practical realization is demonstrated using data from a continuous fermentation process for single cell protein production.  相似文献

The stability of immobilized maltotetraose (G(4))-forming amylase (1,4-alpha-D-glucan maltoteraohydrolase, EC 3.2.1.60) from Pseudomonas stutzeri was investigated in both batch and continous processes. The inactivation process of the immobilized enzyme seemed to obey first-order kinetics, and the immobilized enzyme became more stable when coexisting with 20-30 wt % substrate and calcium ions. From intensive studies on the operational stability in the continuous process, the apparent half-life of G(4) productivity in a constant-flow system was mainly affected by the reaction temperature, substrate concentration, and initial immobilized enzyme activity. A new factor, immobilized enzyme stability factor f(s), was proposed to evaluate the half-life of the immobilized enzyme system.  相似文献

Rat renal glomerular epithelial cells (SGE1 cell line) can be maintained and grown continuously in serum-free medium supplemented with insulin, iron-saturated transferrin (Tr), selenium, bovine serum albumin (BSA), linoleic acid, and epidermal growth factor (EGF). Of the growth supplements used, Tr is essential for proliferation of the cells. In the present study, we describe the use of a unique iron-chelate complex, ferric cacodylate (Fe-Cac), positively charged molecules in neutral buffer, that could almost replace Tr in serum-free culture. It even stimulated the growth of SGE1 cells more efficiently than ferric chloride (FeCl3) and other iron-chelate complexes, such as ferric nitrilotriacetate (Fe-NTA) and ferric citrate (Fe-Cit). The growth-stimulatory activity of Fe-Cac was exerted at iron concentrations of more than 0.01 g/ml, whereas a 10-fold excess of iron concentration was required with FeCl3, Fe-NTA and Fe-Cit. We observed that SGE1 cells grew until confluent, then formed hemicysts (domes) in serum-free medium containing Fe-Cac, suggesting that Fe-Cac did not merely permit cell growth but also supported polarization and organization of the cells into a functional epithelial architecture. Moreover, since the stimulatory activity of Fe-Cac was completely abolished by desferrioxamine, a strong iron chelator, it is suggested that iron is crucial for growth of SGE1 cells. When the cells were treated with suramin, an inhibitor of cellular pinocytosis and endocytosis of a large spectrum of ligands including receptor-bound growth factors, growth-stimulatory activity of Tr was inhibited, whereas the activity of Fe-Cac was not affected. These results, taken together, strongly suggest that the growth-stimulatory activity of Fe-Cac is associated with iron delivery into the cells through the cell membrane by diffusion, which is different from Tr receptor-mediated endocytosis. The use of Fe-Cac for investigating iron-regulated cell proliferation is suggested.  相似文献

Glucose was combined covalently with the epsilon-amino groups of lysyl residues of bovine casein in the presence of sodium cyanoborohydride as a reducing reagent by reductive alkylation, forming stable secondary amine linkages. Solubility characteristics and nutritional values of the neoglycoprotein were examined. The degree of modification (%) of the glucosylated casein was 82.5. Solubility of the modified casein was increased by the attachment of glucose. The modification did not disturb the digestion of casein by pepsin or trypsin. Rat feeding experiments using 10% protein diets demonstrated that the protein efficiency ratio (PER) of the modified casein was 0.35 +/- 0.33 compared with 2.99 +/- 0.29 for the unmodified casein. When the modified casein was supplemented with L-lysine to equal the level of total lysine of unmodified casein, the PER value was increased to 2.21 +/- 0.29. Nitrogen balance experiments showed that the modified casein was digested completely. On the other hand, biological value and net protein utilization of the modified protein were shown to be considerably lower than those of the unmodified casein.  相似文献
