A previously identified Kd restricted epitope of influenza A virus nucleoprotein (147-161) was modified, resulting in recognition by Kd restricted cytotoxic T cells at significantly lower concentrations than the natural peptide sequence. This was achieved by first refining the epitope to the minimum determinant 147-158. Deletion of arginine 156 resulted in a peptide that was shown to be greatly superior in both dose response titrations and in its rate of association with cells to form targets. Analog peptides were tested to determine the important amino acid changes. These data suggest that T cell epitopes can be modified to result in improved immunological recognition.  相似文献

Twenty-four university students received differential Pavlovian conditioning with two colored stimuli separately accompanied by shock, and two other colored stimuli separately presented without shock. The reinforced and nonreinforced pairs of stimuli both contained complementary elements. After differentiation between the reinforced and nonreinforced elements was established, the complementary pairs were each additively mixed, (i.e., presented at the same time and in the same locus), producing two identical white compounds (established by pilot study). The subjects' skin-conductance responses to the two compounds showed that their different conditioning histories did not result in different responses. Rather, a simple declining function was obtained, resembling habituation or extinction. It was concluded that the definition of the conditional stimulus as a physical event is inappropriate in studies in which physically different stimuli may result in identical internal processes (or phenomenologic experiences)--for example, in additive color mixture.  相似文献

The histological structure of ruminant (family: Bovidae) placentomes in eight antelope species was compared to that of domestic cattle and sheep. The chorioallantoic villi differed in degree of branching, surface corrugation, and complexity of utero-placental junction. All species had the epitheliochorial type of placenta, with the epithelial lining of maternal caruncular crypts varying between cellular and syncytial types.  相似文献

Abstract The phototropic response in stems of higher plants is brought about by blue/UV light. The problem studied here is to what extent long-wavelength light, which is absorbed by phytochrome, affects the phototropic response. A refined measurement of phototropism — a curvature index — was applied to the hypocotyl of the sesame seedling (Sesamum indicum L.). The time course of the phototropic response was followed in continuous unilateral weak blue light (B, 460 nm, 8 mW m?2). Long term red light (R) pretreatments, operating through phytochrome, strongly increase the rate and extent of the phototropic response once it is elicited by unilateral B, while the pretreatments decrease the sensitivity towards B. If a R pulse is given immediately prior to the onset of unilateral B, the rate of the response is strongly reduced compared to the time course of curvature observed when the pretreatment was terminated with a long wavelength far-red light (FR) pulse. R and FR were then applied simultaneously with unilateral B to manipulate the status of the phytochrome system during actual curvature. It was found that a low Pfr/P ratio (established by FR) stimulates the phototropic response far above the control (B alone), while a high Pfr/P ratio (established by R) reduces the response below the control. During bending a positive effect of phytochrome on the rate and extent of the phototropic response, which is saturated at a low level of Pfr, appears to be counteracted by an inhibitory effect which dominates at higher levels of Pfr, such as established by omnilateral R. However, if R is applied unilaterally from the same direction as B, R increases the rate of curvature. Apparently the sesame seedling is capable of detecting the direction of R relative to the direction of B. While a mechanistic explanation of these effects cannot be advanced at present, it is clear that the seedling is capable of super-imposing information about the actual light conditions during bending on a ‘memory’ of the light conditions prior to the onset of bending. Thus, the previous as well as the actual light conditions determine its phototropic responsiveness.  相似文献

Summary Enzymic saccharification of Eucalyptus regnans pulps pretreated by autohydrolysis-steam explosion resulted in low cellulose conversions into glucose when using trichodermal cellulase preparations. The reduced levels of glucose were attributable to the production of compounds during enzymic hydrolysis which were inhibitory to -d-glucosidase of Trichoderma reesei C-30 and in Meicelase, but not to the cellulases. Aspergillus niger -glucosidase was not inhibited, nor were -d-xylosidase(s) and 1,4--d-xylanase(s). The inhibitory compound(s) could be extracted from the enzymic hydrolyzates with ethyl acetate. The ethyl acetate extractives inhibited -glucosidase in a competitive manner, and inhibitory action was not affected by pH. Addition of the inhibitory compound(s) to trichodermal cellulase digests of cellulose resulted in reduced glucose yields compared to a control. The inhibitory effects could be overcome when cellulase digests were supplemented with A. niger -glucosidase resulting in higher cellulose-to-glucose conversions. The inhibitory compound(s) were localized mainly in the heartwood of E. regnans. An inhibitor compound of this type has not hitherto been reported. The presence of inhibitory compound(s) in the autohydrolysis liquor fraction is also reported.  相似文献

The effects of pH (3.5-7.5) on the brain uptake of histidine by the blood-brain barrier (BBB) carriers for neutral and cationic amino acids were tested, in competition with unlabeled histidine, arginine, or phenylalanine, with the single-pass carotid injection technique. Cationic amino acid ( [14C]arginine) uptake was increasingly inhibited by unlabeled histidine as the pH of the injection solution decreased. In contrast, the inhibitory effect of unlabeled histidine on neutral amino acid ( [14C]phenylalanine) uptake decreased with decreasing pH. Brain uptake indices with varying histidine concentrations indicated that the neutral form of histidine inhibited phenylalanine uptake whereas the cationic form competed with arginine uptake. Since phenylalanine decreased [14C]histidine uptake at all pH values whereas arginine did not, it was concluded that the cationic form of histidine had an affinity for the cationic carrier, but was not transported by it. We propose that the saturable entry of histidine into brain is, under normal physiological circumstances, mediated solely by the carrier for neutral amino acids.  相似文献

Guineapig antibodies to Streptococcus pneumoniae (SPN) serotype 19F were detected by an enzyme-linked immunosorbent assay using a simple procedure. In experimentally infected hosts, antibody was detectable as early as 2 to 3 weeks after infection, and high titres were maintained for a long period. Antibodies higher than 1:64 were regarded as specific. In a field study, high antibody titres were shown in SPN enzootic colonies in contrast to negative or low antibody titres in a majority of the animals from non-enzootic and SPF colonies.  相似文献
