Structure of low density lipoprotein in complexes formed with arterial matrix components

Temperature dependent techniques (differential scanning calorimetry, polarizing light microscopy and n.m.r. spectroscopy) were used to study the physico-chemical state of low density lipoprotein (LDL) in LDL-aorta glycosaminoglycan and LDL aorta proteoglycan complexes. Complex formation between LDL and glycosaminoglycans (GAGs) resulted in a reversible liquid-liquid crystalline reorganization of the core lipids within the LDL molecule. In the proteoglycan-LDL complexes, prepared by the addition of porcine arterial proteoglycans or of human ‘lipoprotein complexing proteoglycan’, the formation of liquid crystals was an irreversible process, suggesting that the protein moiety of the proteoglycans also participates in the interaction with LDL. Arterial specimens were also examined. In atheromatous intima samples, spherulites with the above characteristics could be identified at the sites of lipoprotein deposition. Since proteoglycans are present in the arterial intima media, the phenomena observed in the present model system may also take place in vivo. The formation of proteoglycan-LDL complexes may result in the establishment of a liquid crystalline LDL structure and may thus play a role in the immobilization of LDL and in the development of the atherosclerotic lesion.     

Fluctuations in the polymerization of sickle hemoglobin. A simple analytic model

Using the stochastic theory of chemical reactions and the theory of first passage times, a simple analytic expression is derived for the distribution of delay times that has been observed in studies of the polymerization kinetics of sickle hemoglobin under conditions where the polymerization progress curves exhibit stochastic variation. The rate of homogeneous nucleation can be readily extracted from such experiments using this expression. This work constitutes a significant addition to the rather limited number of examples where contact can be successfully made between the stochastic theory of chemical kinetics and experiment.     

Synthesis and characterization of (1-13C) Phe9 gramicidin A. Effects of side chain variations

The synthesis of (1-13C)-Phe9-gramicidin (90% enriched) was carried out by the solid phase method. The peptide was removed from the resin by treatment with ethanolamine, deblocked, formylated and purified by preparative t.l.c. to obtain the gramicidin analog in an overall yield of 24%. The peptide was verified and characterized by high pressure liquid chromatography, carbon-13 nuclear magnetic resonance, circular dichroism and single channel currents. Single channel conductances were found to be similar to those of (1-13C)-Phe11-GB but significantly lower than that of gramicidin A. When this gramicidin analog was incubated with phospholipid, the characteristic channel spectrum was not obtained and interaction with sodium ion was not observed. A possible explanation for this behavior is discussed.     

Solomon Islander skin pigmentation: Ultrastructural differences related to genetic variation in Melanesia

Genetic differences between Solomon Islander populations are distinguishable on the basis of melanosome packaging in epidermal keratinocytes. By electron microscopic study of skin pigmentation in various Melanesian populations, we have found distinct inter-island differences in the mode of melanosome packaging. Melanosomes are found as singlets in the skin of Bougainville Islanders but are found aggregated in melanosome complexes in Malaita Islanders and Ontong Javanese. This variation in melanosome packaging represents an important biologic difference found between Solomon Islanders and may reflect the genetic diversity existing in the original Melanesian founding population or populations.     

HuD, a paraneoplastic encephalomyelitis antigen, contains RNA-binding domains and is homologous to Elav and Sex-lethal.

A neuronal antigen (HuD) recognized by the sera of patients with antibody-associated paraneoplastic encephalomyelitis has been isolated by screening a lambda cerebellar expression library. The recombinant antigen provides an unambiguous assay for this rare condition associated with small cell lung cancer. The recombinant antigen has been used to identify specific infiltrating lymphocytes in tumors and affected brain tissues of patients with antibody-associated paraneoplastic encephalomyelitis and sensory neuronopathy. HuD mRNA is uniquely expressed in brain tissue. The HuD protein shows a remarkable homology to the Drosophila proteins Elav and Sex-lethal and is likely to play a role in neuron-specific RNA processing.
