Foam separation on BSA-DNA (bovine serum albumine/deoxyribonucleic acid) and Lysozyme-DNA systems is performed. The separation of the total protein from DNA is evaluated for dissociated chromatin solution. Foam separation for the same systems is done also by a new method of creating a pressure gradient in the Plateau-Gibbs borders in the foam and obtaining a "dry" foam. It is shown that the effectiveness of the foam separation can be improved significantly by the application of the latter method. Some factors (pH, initial concentration of the solution, expansion factor of the foam) influencing the separation of proteins from DNA in the foam and in the residual solution are studied as well.  相似文献

A new subspecies,Lathyrus nissolia L. subsp.futakii Chrtková subsp. nova is described from East Slovakia. The diacritical characters are: high and rich branched stems, 50–110 cm long and larger, 10–12 mm long, bright orange-red flowers. It differs also in ecology, growing in wet lowland forests with the level of the ground water up to 15 cm over the earth also in summer.  相似文献

The fungicide oxafun decreased the production of antibiotic substances active against Phoma betae, by Bacillus subtillis. This was confirmed by reduced effectiveness of antibiotic substances against Phoma infection in micropot experiment. Besides, the bacterial metabolites effected cell membrane permeability of mycelium and resulted in some relationships of fungicide and antibiotic substances.  相似文献

Transforming DNA exposed to either diethyl sulfate (diES) or dimethyl sulfate (diMS) is inactivated. The rate of inactivation depends on the marker tested and on the chemical used: diMS is more active than diES. Cotransformation of linked markers is similarly depressed. In contrast, there is a transient increase in the cotransformation of distant, unlinked markers. These observations indicate that some of the intermolecular complexes of transforming DNA created in the test tube by the treatment with diES and diMS are biologically active. Radioactively labeled DNA treated with diES or diMS changes its patterns of interaction with cellular surfaces that are characteristic of untreated DNA. A possibility is considered that such alterations in DNA-protein interactions as well as the ability of these alkylating agents to transpose fragments of chromosomal material may play an important role in the processes of mutagenesis and, especially, carcinogenesis.  相似文献

The binding properties of 25 beta-lactam antibiotics to Bacillus megaterium membranes have been studied. The affinities of the antibiotics for the penicillin-binding proteins (PBPs) are also reported. We found that PBP 4 has the highest affinity for nearly all the antibiotics studied whereas PBP 5 has the lowest affinity. Both PBP 4 and PBP 5 appear to be dispensable for the maintenance of bacterial growth and survival and appear to be DD-carboxypeptidases. Only the beta-lactam cefmetazol bound preferentially to PBP 5 and has been used to study the inhibition of DD-carboxypeptidase. Comparative studies with beta-lactam that simultaneously result in (a) binding to PBPs 1 and 3, (b) inhibition of cell growth and (c) lysis, stressed the importance of PBPs 1 and 3 for cell growth and survival.  相似文献

We demonstrate that unstimulated platelets attach to immobilized fibrinogen in a selective process mediated by the membrane glycoprotein (GP) complex IIb-IIIa (alpha IIb beta 3). The initial attachment, independent of platelet activation, is followed by spreading and irreversible adhesion even in the presence of activation inhibitors. Using fibrinogen fragments derived from plasmin digestion, we found that unstimulated platelets do not attach to immobilized fragment E, which contains an Arg-Gly-Asp sequence at A alpha 95-97, and adhere to fragments X and D, both containing the gamma 400-411 dodecapeptide adhesion sequence, less efficiently than to intact fibrinogen. Thus, the carboxyl terminus of the A alpha chain, missing in the "early" fragment X used in these studies, appears to be involved in the interaction of fibrinogen with unstimulated platelets. In contrast, activated platelets adhere to immobilized fibrinogen and fragments X, D, and E in a time-dependent and equivalent manner. Although activated platelets adhere to immobilized vitronectin, fibronectin, and von Willebrand factor through GP IIb-IIIa, unstimulated platelets fail to adhere to vitronectin and have only a limited capacity to adhere to fibronectin and von Willebrand factor. These results demonstrate that GP IIb-IIIa on unstimulated platelets displays a recognition specificity for attachment to immobilized adhesive proteins that is distinct from that seen following platelet activation. Thus, unstimulated platelets selectively interact with fibrinogen, and the initial attachment is followed by spreading and irreversible adhesion in the absence of exogenous agonists. This process may be regulated by plasmin cleavage of the fibrinogen A alpha chain and may play an important role during normal hemostasis and during the pathological development of thrombotic vascular occlusions.  相似文献
