The N-terminal protected and unprotected peptides MAHHEEQHG-NH2, Ac-MAHHEEQHG-NH2 from Hpn (Helicobacter histidine-rich protein) and MAHHEQQQQQQA-NH2, Ac-MAHHEQQQQQQA-NH2 from Hpn-like protein, respectively, were synthesized and their interactions with Cu2+ and Ni2+ ions were studied by potentiometric, UV-visible, CD, and EPR techniques. The studies have shown that because of their albumin-like sequence, unprotected peptides are very effective chelating agents for both studied metals. The presence of the hexa-glutamine sequence has very distinct impact on the stability of the complexes formed even if direct interactions with metal ions were not found. The much more effective Ni2+ binding by Hpn-like N-terminal domain when compared to Hpn protein could be critical for different biological functions played by both proteins.  相似文献

The coordination properties of cyclic octapeptides with multi-His motif: c(His-Gly-His-Xaa-His-Gly-His-Xaa) where Xaa = Asp or Lys, were investigated. The binding abilities of this peptides towards Cu(II) ions were studied by using different analytic methods as: potentiometry, spectroscopy and mass spectrometry. The obtained results show that the studied peptides in physiological related pH prefer formation of the species with the {4NIm} binding mode. The efficiency of Cu(II) binding depends on additional side chain groups Asp or Lys. Additionally the analysis of results for His containing cyclopeptides with different numbers of amino acid residues in cyclopeptide ring e.g. four, eight shows that in higher pH in both cases the binding by four amide nitrogens is not observed in the case of α-amino acid peptides.  相似文献

Pseudomonas aeruginosa rods are one of the most common isolated opportunistic nosocomial pathogens. Strains usually are capable to secret a capsule-like polysaccharide called alginate important for evasion of host defenses, especially during chronic pulmonary disease of patients with cystic fibrosis. Most genes for alginate biosynthesis and lysis are encoded by the operon. The aim of our study was to evaluate the incidence of algD sequence, generally use for alginate-coding gene detection, in 120 P. aeruginosa strains resistant to carbapenems. All isolates were obtained in the Department of Clinical Microbiology University Hospital no. 1 of dr A. Jurasz Collegium Medicum of L. Rydygier in Bydgoszcz Nicolaus Copernicus University in Toruń. Examined strains demonstrated resistance to carbenicillin (90,0%), ticarcillin (89,2%) and ticarcillin clavulanate (86,7%). All strains were susceptible to colistin. The majority of examined strains was susceptible to ceftazidime and cefepime (40,8% each) and norfloxacin (37,5%). Presence of algD gene - noted in 112 (93,3%) strains proves that not every strain is capable to produce alginate. It was also found out that differences in algD genes incidence in case of different clinical material that strains were isolated from were not statistically important.  相似文献

There are significant differences between acetyl‐CoA and ATP levels, enzymes of acetyl‐CoA metabolism, and toll‐like receptor 4 contents in non‐activated microglial N9 and non‐differentiated cholinergic SN56 neuroblastoma cells. Exposition of N9 cells to lipopolysaccharide caused concentration‐dependent several‐fold increases of nitrogen oxide synthesis, accompanied by inhibition of pyruvate dehydrogenase complex, aconitase, and α‐ketoglutarate dehydrogenase complex activities, and by nearly proportional depletion of acetyl‐CoA, but by relatively smaller losses in ATP content and cell viability (about 5%). On the contrary, SN56 cells appeared to be insensitive to direct exposition to high concentration of lipopolysaccharide. However, exogenous nitric oxide resulted in marked inhibition pyruvate dehydrogenase and aconitase activities, depletion of acetyl‐CoA, along with respective loss of SN56 cells viability. These data indicate that these two common neurodegenerative signals may differentially affect energy‐acetyl‐CoA metabolism in microglial and cholinergic neuronal cell compartments in the brain. Moreover, microglial cells appeared to be more resistant than neuronal cells to acetyl‐CoA and ATP depletion evoked by these neurodegenerative conditions. Together, these data indicate that differential susceptibility of microglia and cholinergic neuronal cells to neurotoxic signals may result from differences in densities of toll‐like receptors and degree of disequilibrium between acetyl‐CoA provision in mitochondria and its utilization for energy production and acetylation reactions in each particular group of cells.

Ovine β‐lactoglobulin has been isolated from whey fraction of sheep milk and crystallized. The high‐resolution structures of two crystal forms (triclinic and trigonal) obtained at pH 7.0 have been determined revealing that ovine protein, similarly to its bovine analog, is dimeric. Access to the binding site located in the eight‐stranded antiparallel β‐barrel in both structures is blocked by the EF loop that has been found in closed conformation. Similarly to bovine lactoglobulin (BLG), conformation of the EF loop is stabilized by hydrogen bond between Glu89 and Ser116 indicating that Tanford transition might occur with the same mechanism. The substitution at six positions in relation to the most abundant isoform B of BLG also affects the distribution of electrostatic potentials and the total charge. © 2014 Wiley Periodicals, Inc.
