Monoclonal antibodies to an artificial immunogen, specifically demonstrating phosphotyrosine-containing proteins

The importance of protein phosphorylation at tyrosyl hydroxy groups in the control of cell proliferation has recently been established. For identification of tyrosine-phosphorylated proteins, monoclonal antibodies (Mabs) against artificial immunogens containing O-phosphotyrosine (pTyr) or tripeptide pTyr-Gly-Gly as haptens were generated; the haptens were coupled to carrier proteins (bovine serum albumin, human immunoglobulin, keyhole limpet hemocyanin). After immunization of mice with pTyr coupled to keyhole limpet hemocyanin, Mabs were generated which were highly specific for pTyr and did not cross-react with O-phosphoserine, O-phosphothreonine, tyrosine or nucleoside-5'-monophosphates. The Mabs specifically react with tyrosinephosphorylated proteins in the Rous sarcoma virus-transformed rat XC-cell.     

Effects of histones on phagocytosis and phagocytic metabolism]

Enhanced phagocytosis and no intracellular glycogen breakdown was found in guinea-pig peritoneal polymorphonuclear leukocytes (PMN) incubated together with heat-killed staphylococci (Twort) treated with a calf thymus histone fraction (P II), for 1 hr at 37 degrees C. Glcose consumption and lactate production of these PMN's were as high as in phagocytosis of the same germ untreated with P II, and their oxygen consumption was depressed significantly.     

The antioxidant characteristics of medicinal plant extracts from Western Siberia

An antioxidant activity of the water-alcohol extracts of leaves of ten herbs from Western Siberia was studied. In vivo the capability of extracts to protect cells of Escherichia coli against the bacteriostatic action of H ₂O₂ and the influence of the extracts on the expression of the antioxidant gene katG coding catalase-hydroperoxidase I were investigated. In vitro the radical-binding activity with DPhPG^· (1,1-diphenyl-2-picrylhydrazyl radical), the chelating capability with ferrozine, and total composition of flavonoids and tannins were determined. The extracts of Filipendula stepposa and Limonium gmelinii were characterized by the highest antioxidant activity. According to data, the test extracts could have an antioxidant effect on bacteria in different ways at once including the direct inhibition of ROS (reactive oxygen species), iron ion chelation and antioxidant gene induction.     

Functional architecture of the ligamentum arteriosum in adults]

The ligamentum arteriosum was studied in eight normal adult human hearts obtained at autopsy. Four of these hearts were treated by the Zemper method and extensively dissected under a steromicroscope (from X 6 up to X 40); two were embedded in celloidin and sectioned at 100mugm; two were embedded in paraffin and sectioned one 50mugm and the other at 75 mugm. Section were stained by the axan and the resorcin-fuchsin methods. Unstained sections were examined under polarized light.The ligamentum arteriosum may be considered as a small smooth muscle, whose origin is in the arteria pulmonalis sinistra and the insertion in the arcus aortae. Therefore, the ligamentum arterisum may be considered as a myoelastic system, included in a collagenous stroma. Muscle, elastic and collagen fibers present a cross-spiral disposition. Being a myoelastic system with a collagen component, the ligamentum arteriosum cannot be considered as a simple arcus aortae support; it must play some functional role in controlling the arcus aortae curvature during the several different steps of the cardiac cycle.     

The use of an enzymatic kit to measure plasma creatinine in the mouse and three other species

Plasma creatinine concentrations were determined in mice, rats, dogs and humans using a kinetic alkaline picrate method and an enzymatic method with a centrifugal analyser. Lower creatinine values were obtained for all four species using the enzymatic method, but the differences between methods were greater for mice and rats. Removal of creatininase from the enzymatic method reagents gave proportionally higher values for non-creatinine chromogens in mice and rats. A single enzymatic method was not specific for creatinine when used for these species.
