Background
Toll-like receptor (TLR) 2 is a component of the innate immune system and senses specific pathogen associated molecular patterns (PAMPs) of both microbial and viral origin. Cell activation via TLR2 and other pattern recognition receptors (PRRs) contributes to sepsis pathology and chronic inflammation both relying on overamplification of an immune response. Intracellular antibodies expressed and retained inside the endoplasmatic reticulum (ER-intrabodies) are applied to block translocation of secreted and cell surface molecules from the ER to the cell surface resulting in functional inhibition of the target protein. Here we describe generation and application of a functional anti-TLR2 ER intrabody (αT2ib) which was generated from an antagonistic monoclonal antibody (mAb) towards human and murine TLR2 (T2.5) to inhibit the function of TLR2. αT2ib is a scFv fragment comprising the variable domain of the heavy chain and the variable domain of the light chain of mAb T2.5 linked together by a synthetic (Gly4Ser)3 amino acid sequence. 相似文献942.

Methane oxidation by pure cultures of the methanotrophs Methylobacter albus BG8 and Methylosinus trichosporium OB3b was inhibited by ammonium choride and sodium nitrite relative to that in cultures assayed in either nitrate-containing or nitrate-free medium. M. albus was generally more sensitive to ammonium and nitrite than M. trichosporium. Both species produced nitrite from ammonium; the concentrations of nitrite produced increased with increasing methane concentrations in the culture headspaces. Inhibition of methane oxidation by nitrite was inversely proportional to headspace methane concentrations, with only minimal effects observed at concentrations of>500 ppm in the presence of 250 μM nitrite. Inhibition increased with increasing ammonium at methane concentrations of 100 ppm. In the presence of 500 μM ammonium, inhibition increased initially with increasing methane concentrations from 1.7 to 100 ppm; the extent of inhibition decreased with methane concentrations of > 100 ppm. The results of this study provide new insights that explain some of the previously observed interactions among ammonium, nitrite, methane, and methane oxidation in soils and aquatic systems. 相似文献

We investigated the sublethal effects of a predatory crab, Cancer productus (Randall), on the behavior and growth of its snail prey, Littorina sitkana, by setting up controlled rearing and prey-size selection experiments. L. sitkana were collected from three sites on San Juan Island, WA, USA. These sites varied in snail size, abundance, and vertical distribution, and in the abundance of the crab predator C. productus. Snails from all three populations were raised for 34 days under the following treatments: no-crab control, a non-feeding C. productus encased in mesh box, and an encased C. productus feeding on L. sitkana. The non-feeding crab treatment did not affect snail foraging behavior or growth rate in comparison with the no-crab control. In contrast, the presence of a feeding crab elicited escape behavior in the snails, halted grazing, and consequently reduced growth rates. A population difference in escape behavior was observed: upward migration in snails from rocky shores and hiding in crevices in snails from a mud flat. It thus appears that chemicals leaching from crushed conspecific snails, rather than the presence of the crab predator, act as the “alarm substance” to which L. sitkana react. The magnitude of the growth depression in the presence of feeding crabs was 85%, with no difference among the three populations. Once the feeding crab stimulus was removed, snails in all populations resumed normal growth, suggesting that this response to feeding predators is reversible with changing environmental conditions. Laboratory experiments were set up to determine if all size classes of L. sitkana are equally susceptible to C. productus predation. C. productus consistently selected the largest of three size classes of L. sitkana. These results suggest that slow growth rate and small size in L. sitkana may actually be an adaptation for coexisting with high C. productus abundance, rather than simply a cost of escape behavior. 相似文献

As demands for mobile broadband services and ubiquitous network coverage in our societies are increasing, the mobile communication network infrastructure has to be expanded. Concurrently, the technical infrastructure of mobile communication technologies (base stations) raises the public's concerns about health risks due to electromagnetic fields (EMF). By applying conjoint analyses, the study empirically investigates the relationship between mobile data demands, different base station locations, the prevalence of perceived health complaints, and the impact of compensation payments. Findings show that health concerns are the most critical factor for mobile network communication scenario preferences, followed by data rate availability. In the decision scenarios, base station location and compensation payments played a minor role. Two user groups, cellphone and smart phone users who differ in their sensitivity regarding health concerns and data demands, were identified by segmentation analysis. By means of a sensitivity analysis, different mobile communication network scenarios were analyzed. Outcomes show the importance of integrating users’ preferences into the design of mobile communication networks. This especially refers to an increased sensitivity regarding health concerns in cellphone users and minimum requirements for data rates at least sufficient for the usage of mobile Internet services for smart phone users. 相似文献

Anoxia or oxygen and glucose deprivation in SH-SY5Y cells: a step closer to the unraveling of neuroglobin and cytoglobin functions 总被引:4,自引:0,他引:4
Several studies support the hypothesis that neuroglobin and cytoglobin play a protective role against cell death when cellular oxygen supply is critical. Although the underlying molecular mechanisms are unknown, previous reports suggest that this protection can be realised by the fact that they act as ROS scavengers. In this study, expression of neuroglobin and cytoglobin was evaluated in a human neuroblastoma cell line (SH-SY5Y) under conditions of anoxia or oxygen and glucose deprivation (OGD). The cells could survive prolonged anoxia without significant loss of viability. They became anoxia sensitive when deprived of glucose. OGD induced significant cell death after 16 h resulting in 54% dead cells after 32 h. Necrosis was the main process involved in OGD-induced cell death. After reoxygenation, apoptotic neurons became more abundant. Real-time quantitative PCR and Western blotting revealed that neuroglobin and cytoglobin were upregulated, the former under OGD and the latter under anoxic conditions. Under OGD, cell survival was significantly reduced after inhibiting cytoglobin expression by transfection with antisense ODN. Moreover, cell survival was significantly enhanced by neuroglobin or cytoglobin overexpression. When neuroglobin or cytoglobin protein expression increased or decreased, the H(2)O(2) level was found to be lower or higher, respectively. Males of socially monogamous species can increase their siring success via within‐pair and extra‐pair fertilizations. In this study, we focused on the different sources of (co)variation between these siring routes, and asked how each contributes to total siring success. We quantified the fertilization routes to siring success, as well as behaviors that have been hypothesized to affect siring success, over a five‐year period for a wild population of great tits Parus major. We considered siring success and its fertilization routes as “interactive phenotypes” arising from phenotypic contributions of both members of the social pair. We show that siring success is strongly affected by the fecundity of the social (female) partner. We also demonstrate that a strong positive correlation between extra‐pair fertilization success and paternity loss likely constrains the evolution of these two routes. Moreover, we show that more explorative and aggressive males had less extra‐pair fertilizations, whereas more explorative females laid larger clutches. This study thus demonstrates that (co)variation in siring routes is caused by multiple factors not necessarily related to characteristics of males. We thereby highlight the importance of acknowledging the multilevel structure of male fertilization routes when studying the evolution of male mating strategies. 相似文献

Laboratory experiments, were carried out on nymphs and adults of the desert locust, Schistocerca gregaria, and the African migratory locust, Locusta migratoria migratorioides, in view of the field observation that locusts become temporarily less gregarious after the final moult. The experiments showed no reduction in grouping after the final moult but did show a reduction in grouping in mature Schistocerca. Targeted disruption of the different ER genes has generated experimental animal models that are very useful in evaluating the distinct and cooperative roles of the two estrogen receptors, ER and ERβ, in reproductive but also non-reproductive tissues of both sexes. Phenotypic analysis has provided definitive experimental findings for estrogen receptor mediated physiological actions, involving ER in uterine, mammary gland and neuroendocrine sites. ERβ is involved most dramatically in the ovary as is ER. More detailed studies in combination with tissue specific or inducible ER knock outs will be important for future research. 相似文献

Successful vitrification of organ slices is hampered by both osmotic stress and chemical toxicity of cryoprotective agents (CPAs). In the present study, we focused on the effect of osmotic stress on the viability of precision-cut liver slices (PCLS) by comparing different CPA solutions and different methods of loading and unloading the slices with the CPAs. For this purpose, we developed a gradient method to load and unload CPAs with the intention of minimizing sudden changes in osmolarity and thereby avoiding osmotic stress in the slices in comparison with the commonly used step-wise loading/unloading approach. With this gradient method, the CPA solution was introduced at a constant rate into a specially designed mixing chamber containing the slices. We showed that immediate mixing of the infused CPA and the chamber constituents occurred, which enabled us to control the CPA concentration to which PCLS were exposed as a function of time. 相似文献

The lens epithelium-derived growth factor p75 (LEDGF/p75), coded by the PSIP1 gene, is an important host co-factor that interacts with HIV-1 integrase to target integration of viral cDNA into active genes. The aim of this study was to investigate the association of SNPs in the PSIP1 gene with disease outcome in HIV-1 infected patients. We performed a genetic association study in a cohort of 171 HIV-1 seropositive Brazilian individuals classified as rapid progressors (RP, n = 69), typical progressors (TP, n = 79) and long-term nonprogressors (LTNP, n = 23). The exonic SNP rs61744944 and 9 tag SNPs were genotyped. A group of 192 healthy subjects was analyzed to determine the frequency of SNPs and haplotypes in the general population. Linkage disequilibrium (LD) analyses indicated that the SNPs analyzed were not in high LD (r2<0.8). Logistic regression models suggested that patients carrying the T allele rs61744944 (472L) were more likely to develop a LTNP phenotype (OR = 4.98; p = 0.05) as compared to TP group. The same trend was observed when LTNPs were compared to the RP group (OR = 3.26). Results of haplotype analyses reinforced this association, since the OR values obtained for the haplotype carrying allele T at rs61744944 also reflected an association with LTNP status (OR = 6.05; p = 0.08 and OR = 3.44; p = 0.12 for comparisons to TP and RP, respectively). The rare missense variations Ile436Ser and Thr473Ile were not identified in the patients enrolled in this study. Gene expression analyses showed lower LEDGF/p75 mRNA levels in peripheral blood mononuclear cells obtained from HIV-1 infected individuals. However, these levels were not influenced by any of the SNPs investigated. In spite of the limited number of LTNPs, these data suggest that the PSIP1 gene could be associated with the outcome of HIV-1 infection. Further analyses of this gene may guide the identification of causative variants to help predict disease course. 相似文献
