Dissociation of imaginal disc cells has been carried out previously to enable flow cytometry and cell sorting to analyze cell cycle progression, cell size, gene expression, and other aspects of imaginal tissues. However, the lengthy dissociation protocols employed may alter gene expression, cell behavior and overall viability. Here we describe a new rapid and gentle method of dissociating the cells of wing imaginal discs that significantly enhances cell viability and reduces the likelihood of gene expression changes. Furthermore, this method is scalable, enabling collection of large amounts of sample for high-throughput experiments without the need for data-distorting amplifications.  相似文献

Calmodulin levels are elevated three- to fourfold in the dividing cells, resulting from the lectin-induced stimulation of fresh human lymphocytes. This increase in calmodulin appears to be related mainly to progression into S phase and supports the hypothesis that calmodulin might be crucial in regulating the progression of lymphoblasts through their division cycle. Calmodulin levels are higher in a lymphoid cell line derived from human acute lymphoblastic leukemia blood cells than in a lymphoid cell line derived from normal human blood cells, suggesting that calmodulin could be an important mediator of the leukemogenetic process.  相似文献

Discovery of hydrogels has resulted in developing competent controlled-release drug delivery systems. Present study describes the synthesis and characterization of novel pH responsive hydrogels of chitosan, hydroxyl ethyl cellulose (HEC) and polyol prepared by physical blending of the three components in different ratios. Vegetable oil derived polyol seems to act as a filler and cross linking agent. The synthesized hydrogels were characterized using FT-IR spectroscopy, thermo gravimetric analysis (TGA), Optical microscopy and scanning electron microscopy (SEM). Equilibrium swelling behavior of hydrogels in water and different buffers with pH values (2, 4, 7.3, and 8) indicated the sustained expansion of the films in different pH solutions.  相似文献

Herein, we presented the synthesis and application of sodium dodecylbenzenesulfonate–based silver nanoparticles (termed as SDBS-AgNPs). The SDBS reverse micelles (RMs) in ethanol was used as nanoreactor for green AgNPs synthesis. The size, structure, and shape of SDBS-AgNPs were well distinct by UV/visible (UV/Vis), Fourier transform infrared (FTIR) spectroscopy, dynamic light scattering (DLS), and atomic force microscopy (AFM) techniques. The SDBS-AgNPs were quite stable even at high temperature (80 °C), salt concentration (up to 300 μM), and wide pH range (2 to 12). Moreover, SDBS-AgNPs were found to be highly sensitive and selective colorimetric sensor for antihypertensive drug amlodipine (AML). The interaction of AML with SDBS-AgNPs resulted as a substantial increase in the absorbance and a prominent blue shift in wavelength from 426 to 400 nm. DLS results were further confirmed that the SDBS-AgNPs break into smaller sized particles. Similarly, FTIR results also verified the SDBS-AgNPs etching–based sensing of AML molecules due to the strong attraction by amine and carbonyl functional groups on the target drug. The proposed sensor exhibited linear response in the range of 0.001–200 μM (R2 = 0.9917) with limit of detection (LOD) and quantification (LOQ) of 0.161 and 0.49 μM, respectively. The probe remained selective against AML, even in the presence of equimolar interfering species (including other drugs and metal ions). Furthermore, findings proposed that the SDBS-AgNPs might be used as effective substitute to minimize infection severity by obstructing the biofilm formation against nosocomial and urinary tract infection (UTI) causing pathogens.

in silico modeling, using Psipred and ExPASy servers was employed to determine the structural elements of Bcr-Abl oncoprotein (p210BCR-ABL) isoforms, b2a2 and b3a2, expressed in Chronic Myelogenous Leukemia (CML). Both these proteins are tyrosine kinases having masses of 210-kDa and differing only by 25 amino acids coded by the b3 exonand an amino acidsubstitution (Glu903Asp). The secondary structure elements of the two proteins show differences in five α-helices and nine β-strands which relates to differences in the SH3, SH2, SH1 and DNA-binding domains. These differences can result in different roles played by the two isoforms in mediating signal transduction during the course of CML.  相似文献

The effect of transfer factor was studied in 32 children recuperating from protein-calorie malnutrition. Half of the children were given transfer factor; the other half were given saline in a randomized, double-blind trial. Although unexpectedly low mortality in both groups precluded the adequate evaluation of transfer factor in preventing death during the nutritional recovery period, there was no difference between treated and control patients on a variety of indices of rehabilitation. In addition, transfer factor had no demonstrable effect on the evolution of delayed cutaneous hypersensitivity as both groups of patients recovered from the anergy associated with protein-calorie malnutrition.  相似文献

Cadmium (Cd) is a heavy metal with various human exposure sources. It accumulates in the liver, forming a complex with metallothionein protein and progresses to other organs. As a heavy metal, cadmium can replace calcium and other divalent ions and disturb their cascades, ultimately affecting the vital organs. Since cadmium acetate (CA) is considered more lethal than other Cd compounds, the current study examines the effect of different concentrations of CA doses in drinking water for different exposure times in murine models (Mus musculus). After the exposure period, the murine models were then examined histopathologically and biochemically. The histopathological examination of the heart, liver, and kidneys of the experimental group showed extensive degenerative effects. Atomic absorption spectroscopy was used to determine the quantity of cadmium in serum, kidney, and hepatic tissues. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis of hepatic proteins, especially metallothionein, directly related to Cd administration. The biochemical parameters, including creatine kinase, alanine aminotransferase, aspartate aminotransferase, total proteins, glucose, urea, uric acid, and creatinine, were also analyzed. After thorough histochemical and biochemical analysis, it was concluded that even low dose exposure of CA is hazardous to murine models with damaging effects.  相似文献

Refusing consent to organ donation remains unacceptably high, and improving consent rates from family or next‐of‐kin is an important step to procuring more organs for solid organ transplantation in countries where this approval is sought. We have thus far failed to translate fully our limited understanding of why families refuse permission into successful strategies targeting consent in the setting of deceased organ donation, primarily because our interventions fail to target underlying cognitive obstacles. Novel interventions to overcome these hurdles, incorporating an understanding of cognitive psychology and behavioral change therapy, may be beneficial. One potential intervention is to use the concept of nudge theory, where decision‐making is influenced by encouraging positive reinforcement and indirect suggestion. Purposefully nudging families to given consent for organ donation by understanding, and then overcoming, their inherent cognitive biases is novel but also controversial. This article explores the roles of relatives in decisions about organ donation, how nudge theory translates to organ donation and discusses the arguments for and against its application.  相似文献
