Different operational mode of bioreactors influence the biosynthesis of the enzyme and related products as well as the growth of industrial microorganisms. This communication deals with the effect of mode of operation of various bioreactors with different geometric configurations, viz., batch (includes commercially available batch stirred tank, and custom-designed cylindrical and tapered reactors), batch-fed, continuous flow stirred tank reactors on the biosynthesis of penicillin amidase in Escherichia coli. Experimental findings show that the biosynthesis of penicillin amidase in E. coli show a little variation among batch reactor modes and significant variation on the continuous mode of operation. Further analysis show that the different reactor modes also influence periplasmic localization of the enzyme in the cell.  相似文献

Statistical design was used to determine the optimal levels of medium components, the optimal initial pH of the enzyme production medium, the temperature of fermentation, age of the organism in the slant growth and the age of the inoculum for the production of chitinase in shake flask fermentations. The use of high concentrations of chitin and ammonium sulphate and exclusion of peptone and urea from the medium resulted in the production of higher level of the enzyme. The optimal concentrations of the medium components were 12.5 kg/m3 and 4.2 kg/m3 for the chitin and ammonium sulphate respectively. The effect of the addition of peptone and urea to the optimized medium was studied. The optimal values of initial pH and temperature were 5.6 and 28 °C respectively. The optimal age of the slant and the inoculum were found to be 105 h and 43 h respectively. The highest level of chitinase before optimization of the above variables was 0.054 U which was maximized to the level of 0.197 U.  相似文献

A kinetic analysis and optimization of reaction conditions for the enzymatic hydrolysis of chitin using chitinase produced by Trichoderma harzianum NCIM 1185 was carried out. Swollen chitin was used as the substrate for chitinase. The central composite design was followed for this optimization. The required volume ratio of the major reactants for maximum hydrolysis was determined. The pH and temperature optima were found to be 4.75 and 47 °C respectively. K m and V max for this enzyme were 4.643 kg/m3 and 0.1542 U respectively.  相似文献

Hepatitis E virus (HEV) is a major human pathogen in the developing world. In the absence of an in vitro culture system, very little information exists on the basic biology of the virus. A small protein (approximately 13.5 kDa) of unknown function, pORF3, is encoded by the third open reading frame of HEV. We expressed pORF3 in transiently transfected COS-1 and Huh-7 cells and showed that it is a phosphoprotein which is modified at a serine residue(s). Deletion and site-directed mutants were created to establish Ser-80 as the phosphorylation site. This residue is present within a conserved primary sequence that showed consensus sites for phosphorylation by p34cdc2 kinase (cdc2K) and mitogen-activated protein kinase (MAPK). In vitro experiments with hexahistidine-tagged pORF3 expressed either in Escherichia coli or in COS-1 cells showed efficient phosphorylation with exogenously added MAPK. The pORF3 mutants also exhibited an in vitro phosphorylation profile with MAPK which was identical to that observed in vivo. In its primary sequence, pORF3 possesses two highly hydrophobic N-terminal domains. On subcellular fractionation, pORF3 was found to partition with the cytoskeletal fraction, and this association with the cytoskeleton was lost on deletion of hydrophobic domain I (amino acid residues 1 to 32). These results suggest that HEV pORF3 is a cytoskeleton-associated phosphoprotein and are discussed in terms of a possible function for pORF3 within the HEV replicative cycle.  相似文献

Summary Octoploidy was induced in Chili pepper (Capsicum annuum cultivar cerasiformis) through the application of colchicine and the cytomorphological features of two octoploid plants were described. In general, the octoploids did not exhibit gigas characters when compared to the tetraploids; on the contrary they were less vigorous, suggesting that the optimum and desirable ploidy level for Capsicum is probably tetraploid. Chromosome associations such as octovalents and hexavalents, in addition to IVs, IIIs, IIs and Is, were recorded at diakinesis and metaphase I. Meiosis was highly irregular and the pollen and seed fertility was very low. Cytological features of octoploid Chili peppers are compared with octoploids of Physalis and Petunia.  相似文献

Ammonium was shown to be a limiting nutrient for iron oxidation in cultures of Thiobacillus ferrooxidans. In addition, one strain was also able to assimilate nitrate, but not nitrite, for growth and coupled iron oxidation. Some amino acids (0.5 mM) were tested as a source of nitrogen; none clearly stimulated bacterial activity and inhibition was commonly encountered. Complex nitrogenous compounds were inhibitory at high concentrations (0.1 to 0.5%, wt/vol) and, at low concentrations, some clearly stimulated the bacterial iron oxidation in ammonium-limited cultures. Enhancement of iron oxidation by these compounds was also observed in ammonium-unlimited cultures, suggesting their possible role in providing trace nutrients and possibly carbon for the bacteria.  相似文献

Daily administration of cadmium salt for 25 days (2.5 mg per Kg body weight) in the male domestic fowl caused the end of treatment period. An increased incidences of concentration. Fertility dropped to zero at the end of the treatment period. Activity of acid and alkaline phosphomonoesterases were also drastically reduced by the end of treatment period. An increased incidences of morphological abnormalities of spermatozoa were noticed in the treated birds. After 46 days cessation of the treatment, full recovery of the above measures was found. These alterations suggest the reversible type of effect of cadmium chloride on the spermatozoa of male domestic fowl.  相似文献

Effect of oestradiol was studied on the angiotensin converting enzyme (ACE)--a component of renin angiotensin system, in oviduct of immature quails of 15 days of age. ACE was studied in whole oviduct, magnum, shell gland and the glandular epithelium of magnum and shell gland. It was found that whole oviduct had a significantly higher level of ACE in control than those treated with exogenous estrogen at three dose levels (200, 400 or 600 micrograms). ACE contents of whole muscle and glandular epithelium did not differ but magnum had higher ACE level than the shell gland. Results are explained on the basis of functional role of oviductal parts.  相似文献

Precursor feeding strategy for increasing the yield of conessine, a steroidal alkaloid of Holarrhena antidysenterica, was established in cell suspension culture. A total of 50 mg/L added cholesterol was converted into 43 mg/L of alkaloid, 90% of which constituted the conessine. By applying the precursor feeding policy to the cell suspension culture in modified Murashige and Skoog (MS) medium, a total of 143 mg/L of alkaloid was produced in 8 days. In this way the alkaloid content of the cells was increased more than six times compared to that obtained in the standard MS medium. The steps leading to biotransformation of cholesterol into alkaloids were unaffected by phosphate. The shake flask data were successfully transferred to a bench scale 6-L stirred tank bioreactor in which the specific biosynthetic rate of alkaloid production was 110 mg/100 g dry cell weight per day, about 160 times higher than that of whole plant.  相似文献

In a recent study we demonstrated that a synthetic peptide representing residues 124-147 of the major protein of hepatitis B surface Ag (HBsAg) undergoes spontaneous oligomerization to reconstruct one or more conformational group-specific determinants on HBsAg. The present study was undertaken to identify and characterize the HBsAg-related antigenic determinants on this oligomeric peptide (peptide OS[124-147]). A panel of nine analogs of this peptide was generated by either deleting, substituting, or chemical side chain modification of specific amino acid residues. With HBsAg subtype-specific antisera a single "a" epitope was identified as one that includes Met133 and Lys141. In addition a "d" epitope toward the amino-terminal end of the sequence was also observed. Perturbation of certain amino acid residues was found to enhance a antigenicity and subsequent experiments indicated that maximal expression of this a antigenicity is dependent in part on accessibility of the Lys141 side chain and in part on the primary sequence. With a total of 50 human anti-HBsAg serum samples obtained from individuals vaccinated against hepatitis B, it was demonstrated that these sera recognize the Met133-Lys141-dependent a epitope as the dominant, and in many cases the only, determinant on peptide OS[124-147]. Finally, on immunization, peptide OS[124-147] elicits an anti-HBsAg response that is predominantly anti-a though a lesser contribution from an anti-d response was also obtained.  相似文献
