This article reviews data on studies devoted to the seasonal changes of the opiliones populations in the yew and box tree grove of the Caucasian State Nature Biosphere Reserve. Inducible nitric oxide synthase (iNOS) is a major source of nitric oxide during inflammation whose activity is thought to be controlled primarily at the expression level. The B1 kinin receptor (B1R) post‐translationally activates iNOS beyond its basal activity via extracellular signal regulated kinase (ERK)‐mediated phosphorylation of Ser745. Here we identified the signalling pathway causing iNOS activation in cytokine‐treated endothelial cells or HEK293 cells transfected with iNOS and B1R. To allow kinetic measurements of nitric oxide release, we used a sensitive porphyrinic microsensor (response time = 10 msec.; 1 nM detection limit). B1Rs signalled through Gαi coupling as ERK and iNOS activation were inhibited by pertussis toxin. Furthermore, transfection of constitutively active mutant Gαi Q204L but not Gαq Q209L resulted in high basal iNOS‐derived nitric oxide. G‐βγ subunits were also necessary as transfection with the β‐adrenergic receptor kinase C‐terminus inhibited the response. B1R‐dependent iNOS activation was also inhibited by Src family kinase inhibitor PP2 and trans‐fection with dominant negative Src. Other ERK‐MAP kinase members were involved as the response was inhibited by dominant negative H‐Ras, Raf kinase inhibitor, ERK activation inhibitor and MEK inhibitor PD98059. In contrast, PI3 kinase inhibitor LY94002, calcium chelator 1,2‐bis‐(o‐Aminophenoxy)‐ethane‐N,N,N′,N′‐tetraacetic acid, tetraacetoxymethyl ester (BAPTA‐AM), protein kinase C inhibitor calphostin C and protein kinase C activator PMA had no effect. Angiotensin converting enzyme inhibitor enalaprilat also directly activated B1Rs to generate high output nitric oxide via the same pathway. These studies reveal a new mechanism for generating receptor‐regulated high output nitric oxide in inflamed endothelium that may play an important role in the development of vascular inflammation.  相似文献

An Asian powdery mildew fungus Erysiphe (Uncinula) kenjiana (Erysiphales, Ascomycota) has been found in Ukraine. This is the first record of this fungus in Europe. In 2007, E. kenjiana was collected on four Ulmus species in Kiev. All locations adjoined railways or an airport. Development of E. kenjiana was epiphytotic. This species was not found on elms surveyed at towns situated north-east, east or south of Kiev. The fungus may have been brought directly to Ukraine by rail or air transport. In 2008, the fungus was also collected in Chernihiv situated north-east of Kiev. It is likely that E. kenjiana will spread over all Ukraine and into countries of central and western Europe in 2009 or later. Molecular phylogenetic analyses using D1/D2 domains of the 28S rDNA and ITS sequences revealed that the Ulmaceae-Cannabaceae-parasitic powdery mildews, including E. kenjiana, form a clade with strong supports, suggesting that these species diverged from a single ancestor and expanded their host ranges within the Ulmaceae and allied Cannabaceae. This hypothesis is supported by these species sharing the unique morphology of enlarged apices on their chasmothecial appendages. These fungi formed part of a larger grouping with species on Fagaceae, Nothofagaceae, Rosaceae, and Sapindaceae with strong statistical supports. These results suggest that Uncinula-like powdery mildew fungi on these plant families exhibit close evolutionary relationships with their hosts.  相似文献

A series of 5-amino-1H-pyrazoles was synthesized and studied as inhibitors of furin. The most potent compound, 5-amino-4-acetylamino-3-(4-methylphenylamino)1H-pyrazole, was found to retard the activity of furin by mixed-type inhibition with K = 288 microM. These findings permit to plan new ways for chemical modifications of the 5-amino-1H-pyrazole structure and design more potent furin inhibitors of non-peptide nature.  相似文献

Five novel heterometallic Ni/Zn coordination compounds [Ni(en)3][ZnCl4] (1), [Ni(en)(Hea)2][ZnCl4] (2), [Ni(dien)2][ZnCl4] (3), [Ni(en)3][ZnCl4] · 2DMSO (4) and [Ni(en)3][Zn(NCS)4] · CH3CN (5), where en = ethylenediamine (ethane-1,2-diamine), Hea = monoethanolamine (2-aminoethanol) and dien = diethylenetriamine (1,4,7-triazaheptane), have been synthesized by means of the open-air reaction of zinc oxide, nickel chloride (or nickel powder), NH4X (X = Cl, NCS) and ligand (en, dien, Hea) in non-aqueous solvents, such as DMSO, DMF, CH3OH and CH3CN. The choice of a counter-anion in the initial ammonium salt as well as selection of the ligand and solvent provides an effortless approach to the controlled assembly of two- or three-dimensional extended networks assisted by hydrogen bonding. Crystallographic investigations reveal that 1, 2 and 5 possess 3D, while 3 and 4 exhibit 2D H-bonded crystal structures. The structures of the compounds exhibit six-coordinated Ni(II) centers and four-coordinated Zn(II) centers in distorted octahedral and tetrahedral geometries, respectively.  相似文献

Background

Recent experimental results suggest that impairment of auditory information processing in the thalamo-cortical loop is crucially related to schizophrenia. Large differences between schizophrenia patients and healthy controls were found in the cortical EEG signals.

Methods

We derive a phenomenological mathematical model, based on coupled phase oscillators with continuously distributed frequencies to describe the neural activity of the thalamo-cortical loop. We examine the influence of the bidirectional coupling strengths between the thalamic and the cortical area with regard to the phase-locking effects observed in the experiments. We extend this approach to a model consisting of a thalamic area coupled to two cortical areas, each comprising a set of nonidentical phase oscillators. In the investigations of our model, we applied the Ott-Antonsen theory and the Pikovsky-Rosenblum reduction methods to the original system.

Results

The results derived from our mathematical model satisfactorily reproduce the experimental data obtained by EEG measurements. Furthermore, they show that modifying the coupling strength from the thalamic region to a cortical region affects the duration of phase synchronization, while a change in the feedback to the thalamus affects the strength of synchronization in the cortex. In addition, our model provides an explanation in terms of nonlinear dynamics as to why brain waves desynchronize after a given phase reset.

Conclusion

Our model can explain functional differences seen between EEG records of healthy subjects and schizophrenia patients on a system theoretic basis. Because of this and its predictive character, the model may be considered to pave the way towards an early and reliable clinical detection of schizophrenia that is dependent on the interconnections between the thalamic and cortical regions. In particular, the model parameter that describes the strength of this connection can be used for a diagnostic classification of schizophrenia patients.

A mathematical model is proposed to analyze the spinal strain-deformation condition resulting from axial and lateral g-loads that are generated by changes in the gravity field and/or pilot’s actions during high-performance aircraft maneuvering under flight overload conditions. An algorithm of solution has been developed, which takes into account changes in the intervertebral disk pressure and the fibrous ring shape at the time when loading reaches close-to-critical g-values. Calculation of the spinal-strain deformation condition was implemented using the SPLEN computer system (KOMMEK, Russia). Analysis of the spinal straindeformation condition was made for two types of external loads: normal load and unilateral load with the bending moment. Maximum permissible loads on the spinal segment were evaluated, and a pattern of distribution of strain intensity and mean strains, spinal deformation, and the destruction field was described. The developed computer models can be used as a basis for developing a technique of predicting characteristic spinal injuries due to different extreme loads and pathologies.  相似文献
