The results of the experimental investigations of the spectral composition of plasma focus X-ray radiation in the photon energy range of 1.5 keV-400 keV are presented. The test for somatic mutagenesis and recombination in Drosophila is one of the widely used approaches for determination of possible carcinogenic effects of chemical compounds. The use of heterozygotes for mutant tumor suppressor gene wts enables more direct evaluation of the blastomogenic effects of chemical compounds, by tumor formation in the adult flies. This study presents evaluation of the SMART effectiveness upon the use of Drosophila heterozygotes for the wts(P4) gene, first included into the test system. The increase of the test resolution capacity compared to the literature data for the wts(P2) allele was observed. Using wts(P4) heterozygotes, a total of 20 carcinogenic compounds, and their slightly carcinogenic and noncarcinogenic analogs were tested. Specificity of the method was about 100%, and sensitivity depended on the type of the agent tested. The latter was absolute for the direct action carcinogens, with respect to carcinogens, requiring the metabolic activation. The sensitivity was elective and was limited by the presence of the enzymes capable of activating of these compounds. To increase the test sensitivity, the RNA interference-mediated silencing of the Drosophila p53 functional activity was successfully applied. Moreover, the frequency of wts tumor induction considerably increased both in spontaneous and induced mutagenesis conditions.  相似文献

Individual protein binding sites on DNA can be measured in bits of information. This information is related to the free energy of binding by the second law of thermodynamics, but binding kinetics appear to be inaccessible from sequence information since the relative contributions of the on- and off-rates to the binding constant, and hence the free energy, are unknown. However, the on-rate could be independent of the sequence since a protein is likely to bind once it is near a site. To test this, we used surface plasmon resonance and electromobility shift assays to determine the kinetics for binding of the Fis protein to a range of naturally occurring binding sites. We observed that the logarithm of the off-rate is indeed proportional to the individual information of the binding sites, as predicted. However, the on-rate is also related to the information, but to a lesser degree. We suggest that the on-rate is mostly determined by DNA bending, which in turn is determined by the sequence information. Finally, we observed a break in the binding curve around zero bits of information. The break is expected from information theory because it represents the coding demarcation between specific and nonspecific binding.  相似文献

A new methodology for the detection of lipid flip was developed. This methodology relies on the quenching of the fluorescence of the cascade-blue-labeled lipid through complex formation with a membrane-impermeable cyclen-tetranaphthalenethiourea synthetic receptor for this dye. The high affinity of the receptor to cascade-blue label allows the use of micromolar concentrations of this receptor during the experiment. At these low concentrations, the receptor does not interfere with the membrane integrity and, therefore, renders this new methodology less invasive to the model and cell membranes than commonly utilized 7-nitro-1,2,3-benzoxadiazol-4-yl (NBD)-dithionite methodology. Unlike with the NBD-dithionite assay, where the fluorescence quenching of the NBD group is achieved through its chemical modification, this new assay relies on the noncovalent interactions between cascade-blue label and the receptor. Therefore, the quenching can be reverted by either competitive displacement of the lipid-attached label with a water-soluble substrate or by enzymatic degradation of the receptor leading to the label release and fluorescence dequenching. We demonstrate that this new methodology is suitable for the study of lipid flip in both model spherical bilayer membranes and in-vitro experiments.  相似文献

The specific morphological and biological features of dwarf chars Salvelinus malma curilus from three isolated populations of the southern Kuril Islands Kunashir and Iturup were studied. It was shown that brook dwarfs, in comparison with the migratory form of Salvelinus malma curilus, are characterized by a smaller diameter of spawned eggs (which corresponds to smaller yolk reserves and shorter embryos at hatching), retarded growth and morphogenesis of early juveniles, a mosaic pattern of paedomorphic and peramorphic morphological transformations, and a short lifespan. The population and age variability of the relative sizes and shape of otoliths was revealed. Dwarf populations are typically characterized by considerable morphological differences between males and females corresponding to those in migratory fish. The strategy of dwarfism is assumed to be built into the genome of the migratory ecotype of S. malma curilus.  相似文献

Morphological, biochemical, and molecular genetic studies of green microalgae from the collection of cyanobacteria, algae, and algal parasites of St. Petersburg State University (CALU) (presumably belonging to the Parietochloris genus) were conducted in order to estimate biotechnological potential and clarify the phylogenetic position. It was determined that the studied strains have a close relationship to two genera from different classes (Lobosphaera (Trebouxiaceae) and Deasonia (Actinochloridaceae)) and can be of biotechnological interest as producers of valuable polyunsaturated fatty acids (especially arachidonic, linoleic, and α-linolenic).  相似文献

The paper shows the possibility of using the laser Doppler flowmetry method for the assessment of system of blood microcirculation in the lower limbs of patients with diabetes mellitus. A series of experimental studies involving 76 patients with diabetes mellitus and 46 healthy volunteers was carried out. The state of peripheral blood flow was assessed during the local heating tests with different temperature regimes. The wavelet analysis of LDF-grams was used to evaluate the adaptive changes of microcirculation during the temperature tests. The data show that the proposed methodology in the form of heating tests and the use of the wavelet transform in the analysis of LDF-grams allows the evaluation of adaptation processes in the microcirculatory bed during thermal tests and the detection of the preclinical stage of trophic disorders.  相似文献

We studied biochemical features of the lipid and phospholipid composition and patter of fatty acid spectra in lenses of one and two year old salmons bearing cataracts. The lipid complex of lenses of diseased fish underwent significant changes. Cataractogenesis was accompanied by enhanced free radical oxidation and accumulation of malone dialdehyde in lenses of salmons of various ages. The intensification of oxidative processes was synchronized with the decreased level of antioxidant protection (reduced glutathione, vitamins A and E). The activity of Ca2+-dependent proteolytic enzymes in lenses was determined and its decrease in the case of cataract was shown.  相似文献

In experiments with rats, abdominal aorta was subjected to microsurgical anastomosis after local irradiation with doses of 40 and 50 Gy. Irrespective of the time interval between the operation and irradiation the iatrogenic defect was restored completely. With the operation performed 24 h after irradiation the platelet adhesion decreased, the proliferation was inhibited depending on radiation dose, and the pattern of the endotheliocyte migration changed. The above effects were absent with the operation performed one month after irradiation.  相似文献
